The mechanism of therapeutic activity of recombinant murine interferon-gamma (rMu IFN-gamma) and the IFN inducer polyinosinic-polycytidylic acid solubilized with poly-L-lysine in carboxy methyl cellulose (pICLC) in treating metastatic disease was investigated by comparing effector cell augmentation with therapeutic activity in mice bearing experimental lung metastases (B16-BL6 melanoma). Effector cell functions in spleen, peripheral blood, and lung (the organ with tumor) were tested after 1 and 3 weeks of rMu IFN-gamma or pICLC administration (intravenous, three times a week). In these studies, natural killer (NK), lymphokine-activated killer (LAK), cytolytic T lymphocytes (CTL) (against specific and nonspecific targets), and macrophage tumoricidal and tumoristatic activities were measured. rM IFN-gamma and pICLC had therapeutic activity and immunomodulatory activity in most assays of immune function examined. Specific CTL activity of pulmonary parenchymal mononuclear cells (PPMC), but not in splenocytes or peripheral blood lymphocytes (PBL), during week 3 and not during week 1, correlated with the therapeutic activity of rMu IFN-gamma and of pICLC. Macrophage tumoricidal activity in PPMC, but not in alveolar macrophages, also correlated with the therapeutic activity of rMu IFN-gamma, but the opposite was true for the therapeutic activity of pICLC. NK activity of PPMC, but not of splenocytes or PBL, during week 1 correlated with the therapeutic activity of pICLC; in contrast, NK activity at any site did not correlate with the therapeutic activity of rMu IFN-gamma. LAK activity at any site did not correlate with the therapeutic activity of either agent.  相似文献

Human alpha or beta interferons inhibit the proliferation of Daudi Burkitt lymphoma cells and induce the differentiation of these cells towards a mature plasma cell phenotype. Similar responses are seen when Daudi cells are treated with the phorbol ester, TPA. Both interferons and TPA down-regulate expression of the c-myc oncogene in these cells. Although TPA can mimic the effect of interferon on cell differentiation, it does not induce 2'5' oligoadenylate synthetase or the interferon-sensitive mRNAs, 6-16 or 9-27. Thus chronic stimulation of protein kinase C by TPA cannot mimic all of the effects of interferon treatment on gene expression. Inhibition of ADP-ribosyl transferase activity by 3-methoxybenzamide impairs interferon- or TPA-induced differentiation of Daudi cells. This agent induces a higher level of c-myc mRNA in the cells and stimulates the incorporation of [3H]thymidine into DNA; although these effects are partially counteracted by interferon or TPA treatment, the elevated expression of the c-myc gene may be sufficient to prevent terminal differentiation and allow cell proliferation to continue.  相似文献

Neurotransmitter receptors and ion channels play a critical role in the transduction of signals at chemical synapses. The modulation of neurotransmitter receptor and ion channel function by protein phosphorylation is one of the major regulatory mechanisms in the control of synaptic transmission. The nicotinic acetylcholine receptor (nAcChR) has provided an excellent model system in which to study the modulation of neurotransmitter receptors and ion channels by protein phosphorylation since the structure and function of this receptor have been so extensively characterized. In this article, the structure of the nAcChR from the electric organ of electric fish, skeletal muscle, and the central and peripheral nervous system will be briefly reviewed. Emphasis will be placed on the regulation of the phosphorylation of nAcChR by second messengers and by neurotransmitters and hormones. In addition, recent studies on the functional modulation of nicotinic receptors by protein phosphorylation will be reviewed.  相似文献

Summary The effects of the antibiotics methotrexate and chloramphenicol on somatic embryogenesis inCitrus were evaluated. Relatively low levels (0.1 to 1.0 μg/ml) of these antibiotics did not inhibit embryo production from undeveloped ovules of ‘Key’ lime [C. aurantifolia) (Christm.) Swing.]. Surprisingly, both antibiotics induced the formation of embryogenic callus in these cultures. This is usually a rare event in cultures of undevelopedCitrus ovules, and ‘Key’ lime is especially recalcitrant. The effects of these antibiotics on embryogenic callus appeared to be limited to the induction stage, because there was no consistent effect, either stimulatory or inhibitory, on established, lines of embryogenic callus. Summary Lactic acid production by an isolated ofLactobacillus plantarum was standardised on enzyme-hydrolysed tapioca (Manihot esculenta) flour, tapioca starch and soluble starch. Calculated yields of lactic acid (g from 100 g reducing sugars used) in nutrient media containing the abovementioned hydrolysates (10% reducing sugars) were 21.8%, 16.2% and 16.2%, respectively. Higher yields (29–34%) were obtained in media containing 5% reducing sugars. A conversion efficiency of 80–99% was achieved when the acid produced in the broth was neutralised periodically. One hundred milliliters of the medium (5% sugars) yielded 4.0–4.5 g of calcium lactate. These results indicate that unrefined starchy material can be successfully employed for the economic production of lactic acid. The same substrate can also be utilised for biomass production, as viable lactobacilli are being used for therapy in medicine.  相似文献

The error inin vivo 14C incubator measurements of primary production in the Eastern Scheldt when neutral density filters were used and the error obtained when no account was taken of the spectral changes in submarine irradiance that occur with increasing depth, were evaluated theoretically. By multiplying the photosynthetic action spectra of two marine algae by calculated irradiance in the euphotic layer using Kd and Kd() respectively, the gross primary production P[Ed(400–700)] and P[Ed()] was computed. In the green-brown waters of the Eastern Scheldt estuary the use of neutral density filters was sufficient to simulate the underwater light conditions. In clear waters it can cause an overestimation of the gross production.  相似文献

Influence of the food plants ofHeliothis armigera (Hb.) on the degree of parasitism by exotic parasiteCotesia kazak Telenga (Hymenoptera: Braconidae) was studied in cages in the laboratory on 7 food plants such as cotton (Gossypium hirsutum L.), tomato (Lycopersicon esculentum Mill), okra [Abelmoschus esculentus (L.) Moench], Dolichos (dolichos lablab L.), pigeonpea [Cajanus cajan (L.) Millsp.], Cowpea (Vigna unquiculata (L.) and chickpea (Cicer arietium L.). To determine the preference of the parasite 2 test methods were employed. In single plant choice test cotton was most preferred. Next in order of preference were tomato and okra. Dolichos, pigeonpea, cowpea and chickpea were least preferred. In multiple choice test, however, cotton and okra were preferred followed by tomato. Parasites were seen visiting these plants very frequently and high parasitism was recorded on these plants. Chick pea, pigeon pea, cowpea and Dolichos were the least preferred food plants. There appears to be some difference in fecundity as affected by some food plants. Exposure on okra, cotton and tomato resulted in higher cocoon production as compared to pigeonpea, Dolichos, cowpea and chickpea. There was, however, no difference in sex-ratio and longevity of the progeny as affected by food plants. This exotic parasite should be released first in crops such as cotton, okra and tomato on whichH. armigera is a very serious pest in India and elsewhere.
