目的了解不同等级环境(屏障环境和普通环境)对实验大小鼠的一般生理表现和抗应激能力以及对药物反应的影响,初步判定严格的微生物控制是否影响实验动物的人类模型作用。方法将40只SPF级Wistar大鼠和200只SPF级BALB/c小鼠分别饲养于屏障环境和普通环境内,通过以下实验对比观察不同等级环境对实验大、小鼠及其应用的影响:每周测量动物的体重增长情况;饲养35d后,测定大鼠15项血液学指标、14项血液生化指标和9个脏器的相对重量;以抗缺氧实验和游泳实验判断对小鼠体质和抗应激能力的影响;以5.氟尿嘧啶急性毒性实验和戊巴比妥钠麻醉实验测试其对药物作用的异同。结果在5周观察期内,屏障环境饲养小鼠和大鼠的体重增长均明显快于普通环境的对照动物(P〈0.05);普通环境饲养大鼠的血液RBC、WBC、PCT、PLT、GOT、GGT、GPT、CK、TB值高于屏障环境大鼠(P〈0.05),而CHO、LDL-C和HDL-C值低于屏障环境大鼠(P〈0.05);不同微生物环境条件饲养小鼠的游泳耐力和抗缺氧实验结果接近,但是屏障环境组动物所获数据的变异较小;不同等级环境条件饲养小鼠的戊巴比妥钠麻醉反应和5-氟尿嘧啶中毒反应相似,而屏障环境组动物的麻醉维持时间较短,LD50较低,95%可信限狭窄。结论饲养于屏障环境和普通环境的实验大、小鼠在一般生理表现上出现部分差异,动物的抗应激能力和对药物的反应未发生显著变化;在屏障环境条件中,动物的药物反应更敏感、所获数据更稳定,因此,初步认为严格微生物控制条件下的SPF级动物可以作为生活在自然环境中的人类的模型动物。  相似文献

This study investigated body size to fecundity relationships of a reef fish species targeted by line fishing, and examines the potential benefits of increased batch fecundity in no-take reserves compared to fished areas around the Palm, Whitsunday and Keppel Island Groups, Great Barrier Reef, Australia. Lutjanus carponotatus batch fecundity increased with fork length in a non-linear relationship that was best described by a power function. Batch fecundity differed by more than 100-fold among individuals, with a range from 7,074 to 748,957 eggs in fish ranging from 184 to 305 mm fork length. Furthermore, egg diameter increased with fish size. Based on underwater visual census, the potential batch fecundity per unit area in all three island groups ranged from 1.0 to 4.2 times greater in the no-take reserves than in the fished areas between 2001 and 2004. In 2002, a mean 2.3-fold difference in biomass between no-take reserves and fished areas converted to a mean 2.5-fold difference in batch fecundity per unit area. Greater batch fecundity, longer spawning seasons and potentially greater larval survival due to larger egg size from bigger individuals might significantly enhance the potential benefits of no-take marine reserves on the Great Barrier Reef.  相似文献

Gambierdiscus is a genus of benthic dinoflagellates found worldwide. Some species produce neurotoxins (maitotoxins and ciguatoxins) that bioaccumulate and cause ciguatera fish poisoning (CFP), a potentially fatal food‐borne illness that is common worldwide in tropical regions. The investigation of toxigenic species of Gambierdiscus in CFP endemic regions in Australia is necessary as a first step to determine which species of Gambierdiscus are related to CFP cases occurring in this region. In this study, we characterized five strains of Gambierdiscus collected from Heron Island, Australia, a region in which ciguatera is endemic. Clonal cultures were assessed using (i) light microscopy; (ii) scanning electron microscopy; (iii) DNA sequencing based on the nuclear encoded ribosomal 18S and D8‐D10 28S regions; (iv) toxicity via mouse bioassay; and (v) toxin profile as determined by Liquid Chromatography‐Mass Spectrometry. Both the morphological and phylogenetic data indicated that these strains represent a new species of Gambierdiscus, G. lapillus sp. nov. (plate formula Po, 3′, 0a, 7″, 6c, 7‐8s, 5?, 0p, 2″″ and distinctive by size and hatchet‐shaped 2′ plate). Culture extracts were found to be toxic using the mouse bioassay. Using chemical analysis, it was determined that they did not contain maitotoxin (MTX1) or known algal‐derived ciguatoxin analogs (CTX3B, 3C, CTX4A, 4B), but that they contained putative MTX3, and likely other unknown compounds.  相似文献

The biological significance of embryo mortality in demersally spawning coral reef fishes is poorly understood. Here we describe patterns of variation in embryo mortality in Pomacentrus amboinensis (Pomacentridae) at Lizard Island (Great Barrier Reef). The aim was to determine whether numbers of embryos hatched substantially differed from egg production, and if so, identify whether predatory fishes were a source of embryo mortality. Spawning success (number of eggs laid), embryo mortality (proportion of embryos that died prior to hatching) and number of embryos hatching were estimated from daily maps of clutches laid on artificial surfaces (PVC tiles) defended by nesting males. Patterns of variation in eggs laid, embryo mortality and numbers of embryos hatched were examined at three spatial scales: (1) among widely-spaced locations around the island; (2) between adjacent reef slope and patch reef habitats occupied by P. amboinensis at a single location; and (3) among different males within these two habitats. The embryo mortality was extremely high, with a mean of 25.9pm ± 6.2% (S.E.) for 4 locations examined in 1994 and a mean of 69.2pm ± 2.9% for two habitats surveyed in 1995. There were no significant differences in embryo mortality among locations or habitats in either year. This meant that spatial patterns in the number of embryos hatching reflected differences in the number of eggs laid on tiles. Embryo mortality was extremely variable on the scale of individual territories, with embryo mortality commonly ranging from 0% to 100%. Much of the mortality could be attributed to diurnal predatory fishes, especially the wrasse Thalassoma lunare. However, variation in predator densities did not explain spatial patterns in embryo mortality rates. Both solitary and group predatory behaviour was observed, with groups often causing 100% embryo mortality. The level of embryo mortality observed suggests that predation prior to hatching may have a substantial effect on the reproductive output of populations of this demersal-nesting fish.  相似文献

Methamphetamine (METH) is a drug of abuse with neurotoxic and neuroinflammatory effects, which include disruption of the blood-brain barrier (BBB) and alterations of tight junction protein expression. This study focused on the actin cytoskeletal rearrangement as a modulator of METH-induced redistribution of tight junction protein occludin in brain endothelial cells. Exposure to METH resulted in a shift of occludin localization from plasma membranes to endosomes. These changes were accompanied by activation of the actin-related protein 2/3 (Arp2/3) complex, which stimulates actin polymerization by promoting actin nucleation. In addition, METH-induced coronin-1b phosphorylation diminishes the inhibitory effect of nonphosphorylated coronin-1b on actin nucleation. Blocking actin nucleation with CK-666, a specific inhibitor of the Arp2/3 complex, protected against METH-induced occludin internalization and increased transendothelial monocyte migration. Importantly, treatment with CK-666 attenuated a decrease in occludin levels in brain microvessels and BBB permeability of METH-injected mice. These findings indicate that actin cytoskeletal dynamics is detrimental to METH-induced BBB dysfunction by increasing internalization of occludin.  相似文献

A combined laboratory and field study examined the potential for a symbiotic association between the actinorhizal shrub Myrica cerifera and vesicular arbuscular mycorrhizal (VAM) fungi on a Virginia barrier island. M. cerifera seedlings and two test species, Zea mays and Strophostyles umbellata, were grown in an environmental chamber on soils collected from four sites differing in soil age (< 5 to over 130 years), salinity (1–35 g/g total soil chloride), and edaphic characteristics. Seedling root infection was significantly lower for all three species in the youngest soils from the beach where salinity was highest. Stained M. cerifera roots revealed all the components for a functional VAM association; however, there were significantly fewer arbuscules and vesicles relative to the test species. Among field-collected M. cerifera, infection was not detected in mature shrubs from the bay side of the island, where M. cerifera thickets were in a state of degeneration. Infection was highest in soils from the young, developing thickets, and in the most stable thickets of the island interior. Despite the dynamic nature of the barrier island environment, VAM associations with M. cerifera appear to be present, especially in seedlings and developing shrub thickets.  相似文献

Gametogenesis and spawning have been studied since 1986 in a population of Protopalythoa sp. (Anthozoa, Zoanthidea) from Orpheus Island, Great Barrier Reef. Fertile zooids were gonochoric or hermaphroditic with oocytes and sperm vesicles intermingled in the same mesentery; colonies were single sex or mixed. Oocytes arose in macrocnemes below the actinopharynx and were first observed in July. Their mean diameter increased from < 50 µm to 160–240 µm by November, by which time they contained zooxanthellae. Female zooids each produced 800–2400 ova. No testicular vesicles were visible before October but spermatozoa were present in November and December. Spawning in each of three years occurred in November (4th to 6th nights after full moon) simultaneously with mass spawning of scleractinian corals. The buoyant eggs (along with sperm in the hermaphroditic zooids) were released in bundles. Some large oocytes and mature sperm were still present in December.  相似文献

Resistivity and water level measurements were made on a barrier island on the south shore of Long Island, New York to examine the distribution of fresh groundwater and the potential for recirculation of saline groundwater. The depth to the base of the freshwater lens was overpredicted by calculations of the static-equilibrium depth to a sharp interface apparently because of the sensitivity of the calculation to the low water-table elevations which are in turn sensitive to variations in sea level because of the existence of a transition zone between fresh and saline groundwater. Mixing and recirculation of saline groundwater at the base of the lens produced a transition zone up to 9.65 m thick. Measurements also support model forecasts of a mean bay level several centimeters above sea level, augmented by atmospheric forcing and wave setup. A time lag of about 8 hours between the response of the ocean level to longshore winds and the corresponding response of the bay level can result in a difference in elevation between the bay and the ocean that is up to four times that produced by other agents such as Stokes transport and density differences. In the presence of differential hydraulic head, bay and ocean water may be exchanged via groundwater flow between the base of the freshwater lens under the barrier beach and a deeper clay layer.  相似文献
