Dry seeds of Leucadendron laureolum (Lam.) Fourc. (Proteaceae) were exposed for different intervals (range: 7 to 84 days) to visible, UV-A and UV-B radiation of different biologically effective dose (range: 0 to 11.43 kJ m-2 d-1). Changes in seed germination, physiology and ultrastructure, and residual UV effects on seedling performance, were examined. Germination was depressed in seeds following short (7-day) exposures to UV radiation. This depression was intensified with increased UV exposure dose, and most pronounced at shorter UV-B wavelengths. Also glutathione reductase (GR) activities increased in seeds exposed to shorter UV-B wavelengths, but these were unaffected by irradiation dose level in the UV-B range. Electrolyte leakage rates from UV-irradiated seeds were unaltered, which indicated that germination depression did not result from intrinsic membrane damage. The reversal of germination depression (UV-induced dormancy) in UV-irradiated seeds by red light pointed to the possible involvement of phytochrome in this photo-response. Germination depression disappeared in seeds after 56-days irradiation, possibly due to photoreceptor damage by excess UV light. At this stage, all UV irradiated seeds, irrespective of treatment wavelength or dose level, exhibited increased electrolyte leakage rates, which indicated membrane perturbation. Also, increased GR activities were observed in irradiated seeds, but these were proportionately smaller in seeds exposed to shorter wavelength UV-B radiation (9.1 to 35.8% increase) than longer wavelength UV-A (73.4% increase) and visible (97.7% increase) radiation. This implied a metabolic limitation for scavenging of free radicals and peroxides in aging seeds exposed to UV-B radiation, which pointed to accelerated seed deterioration. It was indirectly supported by ultrastructural evidence of sub-cellular damage (lipid coagulation and plasmalemma withdrawal from cell walls) in embryonic tissues of seeds after 84 days UV-B exposure, and reflected in decreased leaf numbers, photochemical efficiencies, and foliar chlorophyll a and carotenoid levels in seedlings cultured from these seeds.  相似文献

In mammalian cells, signal peptide-dependent protein transport into the endoplasmic reticulum (ER) is mediated by a dynamic protein-conducting channel, the Sec61 complex. Previous work has characterized the Sec61 channel as a potential ER Ca(2+) leak channel and identified calmodulin as limiting Ca(2+) leakage in a Ca(2+)-dependent manner by binding to an IQ motif in the cytosolic aminoterminus of Sec61α. Here, we manipulated the concentration of the ER lumenal chaperone BiP in cells in different ways and used live cell Ca(2+) imaging to monitor the effects of reduced levels of BiP on ER Ca(2+) leakage. Regardless of how the BiP concentration was lowered, the absence of available BiP led to increased Ca(2+) leakage via the Sec61 complex. When we replaced wild-type Sec61α with mutant Sec61αY344H in the same model cell, however, Ca(2+) leakage from the ER increased and was no longer affected by manipulation of the BiP concentration. Thus, BiP limits ER Ca(2+) leakage through the Sec61 complex by binding to the ER lumenal loop 7 of Sec61α in the vicinity of tyrosine 344.  相似文献

Evaluating whether protected areas reduce tropical deforestation in Sumatra   总被引:2,自引:0,他引:2  
Aim  This study determines whether the establishment of tropical protected areas (PAs) has led to a reduction in deforestation within their boundaries or whether deforestation has been displaced to adjacent unprotected areas: a process termed neighbourhood leakage.
Location  Sumatra, Indonesia.
Methods  We processed and analysed 98 corresponding LANDSAT satellite images with a c.  800 m2 resolution to map deforestation from 1990 to 2000 across 440,000 km2 on the main island of Sumatra and the smaller island of Siberut. We compared deforestation rates across three categories of land: (1) within PAs; (2) in adjacent unprotected land lying with 10 km of PA boundaries; and (3) within the wider unprotected landscape. We used the statistical method of propensity score matching to predict the deforestation that would have been observed had there been no PAs and to control for the generally remote locations in which Sumatran PAs were established.
Results  During the period 1990–2000 deforestation rates were found to be lower inside PAs than in adjacent unprotected areas or in the wider landscape. Deforestation rates were also found to be lower in adjacent unprotected areas than in the wider landscape.
Main conclusions  Sumatran PAs have lower deforestation rates than unprotected areas. Furthermore, a reduction in deforestation rates inside Sumatran PAs has promoted protection, rather than deforestation, in adjacent unprotected land lying within 10 km of PA boundaries. Despite this positive evaluation, deforestation and logging have not halted within the boundaries of Sumatran PAs. Therefore the long-term viability of Sumatran forests remains open to question.  相似文献

The anti-methicillin-resistant Staphylococcus aureus (anti-MRSA) activity and the possible mechanism of action of a crude extract from red bulbs of Eleutherine americana Merr. were investigated. The crude ethanolic extract from E. americana produced minimum inhibitory concentrations (MICs) of 62.5–1000 and 250 μg mL−1 against MRSA isolates and the reference strains, respectively. Treatment of S. aureus ATCC 27664 with a crude extract at 2MIC reduced the inoculum size by 5 log at 24 h compared with the control. The combined effect of the extract and 7.5% NaCl on the enterotoxin-producing ATCC strain resulted in no detection of organisms within 24 h compared with the control. The release of cell materials after extract treatment was determined by measuring OD260 nm, the treatment resulted in cytoplasmic leakage. Determination of OD620 nm showed that the extract did not cause gross cell wall damage. However, observation of S. aureus cells under an electron microscope after treatment with 2MIC and 4MIC of the crude extract revealed that the extract caused damage to membrane morphology. A knowledge of the mechanism of action of the E. americana extract may offer useful hints in the search for novel antibacterial substance.  相似文献

Mass increases in raffinose family oligosaccharides (RFOs, α 1,6-galactosyl extensions of sucrose) are well documented in the generative tissues of many plants upon cold acclimation, and they (i.e. mainly the two shortest RFO members, raffinose and stachyose) have been suggested as frost stress protectants. Our focus here was on the longer RFO members as they commonly occur in the frost-hardy evergreen labiate Ajuga reptans in its natural habitat, and accumulate to their highest concentrations in winter when the plant is faced with sub-zero temperatures. We examined the effects of RFO concentration and chain length on frost tolerance using excised leaves which accumulate long-chain RFOs under both cold and warm conditions, thereby uncoupling the acclimation temperature from RFO production. We demonstrated that frost tolerance in excised A. reptans leaves correlates positively with long-chain RFO accumulation under both acclimation temperatures. After 24 d post-excision in the warm, the leaves had increased their RFO concentrations (mainly long-chain RFOs) 22-fold to 78 mg g−1 fresh weight, and decreased their EL50 values (temperature at which 50% leakage occurred) from −10.5 to −24.5 °C, suggesting a protective role for these oligosaccharides in the natural frost tolerance of A. reptans .  相似文献

International trade transfers social and environmental impacts across national borders. The consumption of forest products often takes place far away from industrial production sites, and mills procure raw material from remote forests. Finland produces about 10% of forest products that are traded internationally, with the majority of its exports destined for other European countries. Here we report and analyze data that demonstrate that international leakage, in relative terms, increased faster than the production of commodities. The international consumption of products made in Finland increased, and an increase in wood imports from Russia provided the raw material for most of the incremental production. The international consumption‐production system translated the increasing global demand for Finnish products into increased harvests in Russia, until the Russian customs duties started to increase in 2007. We argue that national and regional policies for the promotion of sustainable consumption and production must be analyzed and assessed from an international, holistic perspective.  相似文献

研究了300mmol/L NaCl胁迫对19个不同类型南瓜品种幼苗盐害指数、电解质渗透率、脯氨酸及可溶性糖含量的影响。结果表明,随NaCl胁迫时间延长,南瓜幼苗盐害指数、电解质渗透率、脯氨酸含量和可溶性糖含量均呈升高趋势。盐害指数和电解质渗透率能较好反映南瓜幼苗的耐盐性,可作为南瓜品种耐盐性的筛选指标。在NaCl胁迫下,青栗南瓜(Q1)表现出很强的耐盐性,黑蛮南瓜(H2)和黑籽南瓜(H3)为盐敏感品种,而其他为耐盐性中等品种。  相似文献

Proteinase K-containing liposomes with highly selective membrane permeability properties were prepared. The selectivity obtained was with respect to the two substrate molecules added to the external aqueous phase of the liposomes: acetyl-L-Ala-Ala-Ala-p-nitroanilide (Ac-AAA-pNA) and succinyl-L-Ala-Ala-Ala-p-nitroanilide (Suc-AAA-pNA). The liposome-forming lipid used was POPC (1-palmitoyl-2-oleoyl-sn-glycero-3-phosphocholine) and modulation of the membrane permeability was achieved using the detergent cholate. Proteinase K-containing mixed liposomes (PKCL) were prepared by adding cholate to preformed proteinase K-containing POPC liposomes (PKL) at a defined effective cholate/POPC molar ratio in the liposomal bilayer membrane R(e). Proteinase K was kept inside PKCL with a negligible amount of leakage into the bulk aqueous phase at R(e) < or = 0.30. At higher R(e), leakage of proteinase K was pronounced, even under conditions where POPC/cholate mixed liposomes seemed to be still intact (0.30 < R(e) < or = 0.39). At R(e) < or = 0.30, the reactivity of proteinase K in the PKCL measured with the externally added substrate Ac-AAA-pNA increased with increasing R(e), while the reactivity measured with Suc-AAA-pNA remained low, regardless of the R(e) value. This showed that externally added Ac-AAA-pNA molecules permeated the liposomal membrane more easily than Suc-AAA-pNA by modulating the membrane with cholate. Consequently, Ac-AAA-pNA was hydrolyzed in PKCL with considerably higher apparent substrate selectivity in comparison with the cases of proteinase K in PKL and free proteinase K (without liposomal encapsulation). The results obtained clearly demonstrate that the prepared PKCL can be utilized as a kind of nano-scaled bioreactor system which can take up a particular target substrate with high apparent substrate selectively from the external phase of the liposomes. Inside the liposomes, the target substrate is then converted into the corresponding products.  相似文献
