Herein, for the first time, we comparatively report the opening and closing of apo plasmepsin I – V. Plasmepsins belong the aspartic protease family of enzymes, and are expressed during the various stages of the P. falciparum lifecycle, the species responsible for the most lethal and virulent malaria to infect humans. Plasmepsin I, II, IV and HAP degrade hemoglobin from infected red blood cells, whereas plasmepsin V transport proteins crucial to the survival of the malaria parasite across the endoplasmic reticulum. Flap‐structures covering the active site of aspartic proteases (such as HIV protease) are crucial to the conformational flexibility and dynamics of the protein, and ultimately control the binding landscape. The flap‐structure in plasmepsins is made up of a flip tip in the N‐terminal lying perpendicular to the active site, adjacent to the flexible loop region in the C‐terminal. Using molecular dynamics, we propose three parameters to better describe the opening and closing of the flap‐structure in apo plasmepsins. Namely, the distance, d1, between the flap tip and the flexible region; the dihedral angle, ?, to account for the twisting motion; and the TriCα angle, θ1. Simulations have shown that as the flap‐structure twists, the flap and flexible region move apart opening the active site, or move toward each other closing the active site. The data from our study indicate that of all the plasmepsins investigated in the present study, Plm IV and V display the highest conformational flexibility and are more dynamic structures versus Plm I, II, and HAP. The influence of psychosocial components on back and abdominal endurance tests in patients with persistent non-specific low back pain should be investigated to ensure the correct interpretation of these measures. Three-hundred and thirty-two patients (291 men and 41 women) from 19 to 63 years performed an abdominal and back muscle endurance test after completing some psychosocial questionnaires. During the endurance tests, surface electromyography signals of the internal obliques, the external obliques, the lumbar multifidus and the iliocostalis were recorded. Patients were dichotomized as underperformers and good performers, by comparing their real endurance time, to the expected time of endurance derived from the normalized median frequency slope. Independent t-tests were performed to examine the differences on the outcome of the questionnaires. In the back muscle endurance test, the underperformers had significantly lower (p < 0.05) scores on some of the physical subscales of the SF-36. The underperformers group of the AE test scored significantly higher on the DRAM MZDI (p = 0.018) and on the PCS scale (p = 0.020) and showed also significantly lower scores on the SF-36 (p < 0.05). Back muscle endurance tests are influenced by physical components, while abdominal endurance tests seem influenced by psychosocial components.  相似文献

Tissue engineering has brought new hopes for urethral reconstruction. However, the absence of pre‐vascularization and the subsequent degradation of materials often lead to the failure of in vivo application. In this study, with the assistance of hypoxia‐activated human umbilical cord mesenchymal stem cells (hUCMSCs), pedicled muscle flaps were used as materials and pre‐incubated in ventral penile subcutaneous cavity of rabbit for 3 weeks to prepare a pre‐vascularized urethral construct. We found that small vessels and muscle fibres were scattered in the construct after 3 weeks' pre‐incubation. The construct presented a fibrous reticular structure, which was similar to that of the corpus spongiosum under microscope examination. The produced constructs were then used as a patch graft for reconstruction of the defective rabbit urethra (experimental group), natural muscular patch was used as control (control group). Twelve weeks after the reconstructive surgery, urethrography and urethroscope inspections showed wide calibres of the reconstructed urethra in the experimental group. Histopathological studies revealed that fibrous connective tissues and abundant muscle fibres constituted the main body of the patch‐grafted urethra. In contrast, in the control group, only adipose tissue was found in the stenosis‐reconstructed urethra, replacing the originally grafted muscular tissue. To our knowledge, this is the first report that successfully constructed a pre‐vascularized urethral construct by using hypoxia‐activated hUCMSC and pedicled muscle flaps. More importantly, the pre‐vascularized construct showed a good performance in urethral reconstruction when applied in vivo. The study provided a novel strategy for tissue engineering of pre‐vascularized urethral construct for the defective urethra, representing a further advancement in urethral reconstruction.  相似文献

The propensity for chronic wounds in humans increases with ageing, disease conditions such as diabetes and impaired cardiovascular function, and unrelieved pressure due to immobility. Animal models have been developed that attempt to mimic these conditions for the purpose of furthering our understanding of the complexity of chronic wounds. The model described herein is a rat ischemic skin flap model that permits a prolonged reduction of blood flow resulting in wounds that become ischemic and resemble a chronic wound phenotype (reduced vascularization, increased inflammation and delayed wound closure). It consists of a bipedicled dorsal flap with 2 ischemic wounds placed centrally and 2 non-ischemic wounds lateral to the flap as controls. A novel addition to this ischemic skin flap model is the placement of a silicone sheet beneath the flap that functions as a barrier and a splint to prevent revascularization and reduce contraction as the wounds heal. Despite the debate of using rats for wound healing studies due to their quite distinct anatomic and physiologic differences compared to humans (i.e., the presence of a panniculus carnosus muscle, short life-span, increased number of hair follicles, and their ability to heal infected wounds) the modifications employed in this model make it a valuable alternative to previously developed ischemic skin flap models.  相似文献

目的:探讨颌面部皮肤软组织大面积缺损凹陷的理想修复方法。方法:本组6例均为爆炸伤后颌面部皮肤软组织缺损及严重凹陷畸形,采用胸三角皮肤扩张形成带蒂皮瓣修复上述皮肤缺损及自体脂肪移植纠正残存凹陷畸形。手术分五步进行:1.胸三角深筋膜浅层埋植500mL-800mL皮肤扩张器并注水扩张3个月。2井艮据面颈部预计皮肤缺损大小及形状作皮瓣预制并面部局部皮瓣纠正器官移位。3.带蒂皮瓣转移修复颌面部缺损。4.蒂部延迟及断蒂微整形。5.自体脂肪移植。结果:所有皮瓣成活良好,皮瓣色质接近面颈部周围正常皮肤,缺损畸形修复,外观形态好,供区直接缝合无需植皮,取得了较好的面部改观效果。结论:对于面部大面积皮肤软组织缺损,合并面部凹陷、面部器官缺损及移位,采取胸三角扩张延迟预制皮瓣并自体脂肪移植修复可取得良好的整复效果,为颌面部战创伤畸形提供了理想的修复方法。  相似文献

目的:分析子宫不同占位性病变的声像学特点和和误诊原因,探讨超声诊断价值.方法:回顾性分析我院185例不同占位性病变经腹部超声图像资料,观察病变形态、大小、病变位置、包膜、边缘及后方回音等.结果:185例患者中,经体表超声检测出139例不同占位性病变患者,符合率为75.14%;误诊46例,误诊率为24.86%,误诊原因与病变形态、内部回音、宫腔线所表现出的相似性有关.结论:腹部超声能够较准确地诊断出子宫不同占位性病变,结合患者病史和临床症状,可以实现其更好的利用价值.  相似文献

该研究利用光学和扫描电子显微镜对采自湖南东洞庭湖国家级自然保护区的硅藻标本进行了观察,鉴定出其中一种硅藻为比提基金长篦藻(Neidium beatyi Hamilton et al.)。结果表明,该长篦藻具有以下鉴定特征:(1)线性椭圆至线性披针形的壳瓣外形。(2)壳面两侧具有3~5条纵向线。(3)向相反方向弯曲的两近缝端,呈叉状分枝的远缝端末梢及顶端的三角形硅质顶盖。(4) 16~18条/10μm的线纹密度和14~18个/10μm的孔纹密度。(5)一些孔纹内部开口周围的4~7个豆状突出物。本研究结果与比提基金长篦藻模式种群的形态特征和数量性状均吻合,故确定其为比提基金长篦藻;增加了比提基金长篦藻的新地理分布区域——中国洞庭湖。  相似文献

摘要 目的:探讨口腔颌面部肿瘤患者术后缺损的外侧皮瓣修复术与前臂皮瓣修复术对比。方法:选取遂宁市中心医院(我院)2015年8月到2020年8月共收治的120例口腔颌面部肿瘤患者,所有患者通过肿瘤切除术后均出现组织缺损现象,对所有组织缺损的患者依照不同的皮瓣修复方式分为两组,其中应用外侧皮瓣修复术的68例患者分为外侧皮瓣修复组,应用前臂皮瓣修复术的52例患者分为前臂皮瓣修复组,对比两组的皮瓣修复效果,治疗后的口腔功能恢复情况,瓣成活率、术后皮瓣危象率以及血管吻合时间,并发症情况。结果:外侧皮瓣修复组的总有效率为95.59 %,前臂皮瓣修复组的总有效率为84.62 %,外侧皮瓣修复组明显高于前臂皮瓣修复组(P<0.05);外侧皮瓣修复组患者的外形修复、语言功能、咀嚼功能、吞咽功能以及口腔闭合评分明显高于前臂皮瓣修复组(P<0.05);两组的皮瓣成功率均比较高对比无明显差异(P>0.05),两组的术后皮瓣危象率比较低,对比无明显差异(P>0.05),血管吻合时间对比无明显差异(P>0.05);对比两组并发症发生情况发现,外侧皮瓣修复组的总并发症发生率为2.94 %,前臂皮瓣修复组并发症发生率为17.31 %,外侧皮瓣修复组明显低于前臂皮瓣修复组(P<0.05)。结论:对口腔颌面部肿瘤患者术后缺损患者应用外侧皮瓣修复术能够提升患者的皮瓣修复效果,提升患者的口腔功能恢复情况,减少并发症的发生,安全性好,值得临床应用推广。  相似文献

目的:探讨沙漠干热环境下猪腹部肠管火器伤后肝脏环氧化酶-2(COX-2)的表达与肝细胞凋亡之间的关系。方法:沙漠干热环境组和常温环境组各42头长白仔猪随机等分为对照组和伤后1h、2h、4h、8h、12h和24h组7个时间组,实验各组在建立腹部肠管火器伤模型后,用免疫组化图像分析法测定各组肝组织COX-2的表达,采用TUNEL法观察肝细胞凋亡情况,并检测血清ALT的变化。结果:沙漠干热环境组和常温环境组中各实验组COX-2表达水平及肝细胞凋亡率均明显高于对照组(P〈0.01);在沙漠干热环境组,各实验组肝脏COX-2表达水平明显高于相对应的常温组(P〈0.01),肝细胞凋亡率在1h、2h、4h、8h明显高于相对应的常温组(P〈0.05或P〈0.01);肝脏COX-2的表达水平和肝细胞凋亡率在沙漠干热环境组和常温组中的第1个高峰值均出现在伤后2h组,沙漠干热环境组的第2个高峰值出现在伤后8h组,而常温环境组的第2个高峰出现在伤后12h组;血清ALT的变化趋势与COX-2的表达及细胞凋亡的趋势相似,肝脏COX-2表达水平和肝细胞凋亡率在沙漠干热环境组和常温环境组均具有相关性,相关系数分别为0.973和0.945,(P〈0.01)。结论:沙漠干热环境腹部肠管火器伤后肝脏COX-2的水平明显高于常温组,COX-2的表达与肝细胞凋亡的变化趋势一致,且密切相关,提示在沙漠干热环境腹部肠管火器伤后COX-2可能通过促进肝细胞凋亡在继发性肝脏损害过程中起重要作用。  相似文献
