miRNA-21对肺癌细胞放疗敏感性调控及肺损伤的影响

[目的]探究miRNA-21对肺癌细胞放疗敏感性的调控及放疗所致肺损伤的影响。[方法]选取人小细胞肺癌细胞系NCI-H446作为肺癌细胞模型、人支气管上皮细胞HBE作为放疗所致肺损伤细胞模型，取对数生长期NCI-H446和HBE细胞，分为空白对照组(不转染细胞)、阴性对照组(转染阴性对照序列细胞)和anti-miRNA-21组(转染anti-miRNA-21序列细胞),检测各组细胞中miRNA-21表达水平细胞活率、细胞周期进程。[结果]NCI-H446中miRNA-21表达量显著高于BEAS-2B,5.0Gy组的NCI-H446和HBE细胞活率均显著低于0.0Gy组，与0.0Gy比较，2.5、5.0Gy组的NCI-H446和HBE细胞的G2/M期细胞百分比显著增高(P<0.05),G1/S期细胞百分比显著降低，5.0Gy γ照射后，在8 h前，NCI-H446细胞中的miRNA-21表达水平显著上调，8 h后逐渐回落；在12 h前，HBE细胞中的miRNA-21表达水平显著上调，12 h后逐渐回落。转染后，anti-miRNA-21组NCI-H446和HBE细胞中的miRNA-...     

Directional migration of leukocytes：their pathological roles in inflammation and strategies for development of anti－inflammatory therapies

Directional migration of leukocytes is indispensable to innate immunity for host defense. However, recruitment of leukocytes to a site of tissue injury also constitutes a leading cause for inflammatory responses. Mechanistically, it involves a cascade of cellular events precisely regulated by temporal and spatial presentation of a repertoire of molecules in the migrating leukocytes and their surroundings (microenvironments). Here I will summarize the emerging evidence that has shed lights on the underlying molecular mechanism for directional migration of leukocytes, which has guided the therapeutical development for innovative anti-inflammatory medicines.

Calcium-Mediated Mitochondrial Permeability Transition Involved in Hydrogen Peroxide-Induced Apoptosis in Tobacco Protoplasts

In the present study, we focused on whether Intracellular free Ca^2＋ （[Ca^2＋],） regulates the formation of mltochondrlal permeability transition pore （MPTP） In H2O2-induced apoptosis In tobacco protoplasts. It was shown that the decrease In mltochondrlal membrane potential （△ψm） preceded the appearance of H2O2-Induced apoptosls; pretreatment with the specific MPTP Inhibitor cyclosporine A, which also Inhibits Ca^2＋ cycling by the mitochondria, effectively retarded apoptosls and the decrease In △ψm. Apoptosls and decreased △ψm were exacerbated by CaCl2, whereas the plasma membrane voltage-dependent Ca^2＋ channel blocker lanthanum chloride （LaCl3） attentuated these responses. Chelation of extracellular Ca^2＋ with EGTA almost totally Inhibited apoptosls and the decrease In △ψmInduced by H2O2. The time-course of changes In [Ca^2＋]l In apoptosls was detected using the Ca^2＋ probe Fiuo-3 AM. These studies showed that [Ca^2＋]1 was Increased at the very early stage of H2O2-Induced apoptosls. The EGTA evidently Inhibited the Increase In [Ca^2＋]1 Induced by H=O=, whereas It was only partially Inhibited by LaCl3. The results suggest that H2O2 may elevate cytoplasmic free Ca^2＋ concentrations In tobacco protoplasts, which mainly results from the entry of extracellular Ca^2＋, to regulate mltochondrlal permeability transition. The signaling pathway of [Ca^2＋]1-medlated mltochondrlal permeability transition was associated with H2O2-Induced apoptosis In tobacco protoplaete.
