During the degradation of Microcystis spp. blooms, ammonia and microcystin are released into water column at the same time, which may impact aquatic organisms. This study assessed the responses of oxidative stress to a combination of ammonia and microcystin in larval fish. Newly-hatched larvae and 21-day-old larvae of silver carp Hypophthalmichthys molitrix were exposed to solutions with different concentrations of ammonia (NH3–N) and microcystin-LR (MC-LR). Results indicated significant interaction between NH3–N and MC-LR on glutathione (GSH) and malondialdehyde (MDA) levels in newly-hatched larvae. In the 21-day-old larvae, MDA fluctuated significantly with time and the two toxins, and significant interactions were detected among any two or three factors (time, NH3–N, MC-LR); the superoxide dismutase (SOD) and GSH changed significantly with time and concentrations of toxins and were time or dose-dependent on NH3–N and MC-LR exposure; results also indicated significant interaction on SOD or GSH among any two or three factors.  相似文献

The finless porpoise, Neophocaena phocaenoides, is endemic to the coastal waters of the Indo-Pacific, ranging from the Persian Gulf to Japan. Nine tetranucleotide microsatellite loci were isolated from the finless porpoise (Neophocaena phocaenoides). Polymorphism of each locus was assessed in 39 unrelated individuals from the Yellow Sea and the South China Sea of the Chinese waters. The number of alleles per locus varied from 2 to 11. The ranges of observed and expected heterozygosity were 0.154–0.795 and 0.146–0.839, respectively. Cross-species amplification of these loci was tested in other cetacean species. These microsatellite markers described here will be suitable for population genetic studies of finless porpoises and other cetacean species.  相似文献

The metabolic response of the freshwater prawn, Macrobrachium nipponense to nitrite toxicity was evaluated. The prawns were exposed to 0, 1, 2, 3 and 4 mg L?1 NO2-N concentrations for 48 h. The metabolic parameters in muscle were measured after 12, 24 and 48 h. Glucose level significantly increased after 24 h. Exposure to lower nitrite concentrations (1 and 2 mg L?1) resulted in significant increases in alanine aminotransferase (ALT) activities after 24 and 48 h. Aspartate aminotransferase (AST) activities treated with 2 and 3 mg L?1 nitrite-N at 48 h were significantly higher than those at 12 and 24 h. Intermediate sublethal nitrite concentrations produced significant elevations in lactate dehydrogenase (LDH) activities from 12 h up to 48 h. No significant changes were detected in any of the groups for triglycerides and creatine kinase (CK). To satisfy the increased energy demands caused by acute nitrite exposure, mobilization of lipids is not the main reason while utilization of amino acids seems to play a more important role. The results would be helpful for aquaculture farmers to prevent a potential depression of productivity caused by elevated nitrite levels.  相似文献

The remarkable antiquity, diversity, and significance in the ecology and evolution of Cetartiodactyla have inspired numerous attempts to resolve their phylogenetic relationships. However, previous analyses based on limited samples of nuclear genes or mitochondrial DNA sequences have generated results that were either inconsistent with one another, weakly supported, or highly sensitive to analytical conditions. Here, we present strongly supported results based upon over 1.4 Mb of an aligned DNA sequence matrix from 110 single-copy nuclear protein-coding genes of 21 Cetartiodactyla species, which represent major Cetartiodactyla lineages, and three species of Perissodactyla and Carnivora as outgroups. Phylogenetic analysis of this newly developed genomic sequence data using a codon-based model and recently developed models of the rate autocorrelation resolved the phylogenetic relationships of the major cetartiodactylan lineages and of those lineages with a high degree of confidence. Cetacea was found to nest within Artiodactyla as the sister group of Hippopotamidae, and Tylopoda was corroborated as the sole base clade of Cetartiodactyla. Within Cetacea, the monophyletic status of Odontoceti relative to Mysticeti, the basal position of Physeteroidea in Odontoceti, the non-monophyly of the river dolphins, and the sister relationship between Delphinidae and Monodontidae + Phocoenidae were strongly supported. In particular, the groups of Tursiops (bottlenose dolphins) and Stenella (spotted dolphins) were validated as unnatural groups. Additionally, a very narrow time frame of ∼3 My (million years) was found for the rapid diversification of delphinids in the late Miocene, which made it difficult to resolve the phylogenetic relationships within the Delphinidae, especially for previous studies with limited data sets. The present study provides a statistically well-supported phylogenetic framework of Cetartiodactyla, which represents an important step toward ending some of the often-heated, century-long debate on their evolution.  相似文献

Upon microbial invasion the innate immune system of Drosophila melanogaster mounts a response that comes in two distinct but complimentary forms, humoral and cellular. A screen to find genes capable of conferring resistance to the Gram-positive Staphylococcus aureus upon ectopic expression in immune response tissues uncovered imd gene. This resistance was not dependent on cellular defenses but rather likely a result of upregulation of the humoral response through increased expression of antimicrobial peptides, including a Toll pathway reporter gene drosomycin. Taken together it appears that Imd pathway is capable of playing a role in resistance to the Gram-positive S. aureus, counter to notions of traditional roles of the Imd pathway thought largely to responsible for resistance to Gram-negative bacteria.  相似文献

Fluctuations in incubation temperature affect incubation duration but not morphology, locomotion and growth of hatchlings in the sand lizard Lacerta agilis (Lacertidae). —Acta Zoologica (Stockholm) 00 : 1–8. Studies looking for potential effects of temperature and temperature fluctuations on phenotypic traits of reptile hatchlings have shown species variation, but have not always allowed a distinction between effects of fluctuation per se and temperature extremes themselves. To examine whether incubation temperature fluctuation has a key role in influencing the phenotype of offspring, we incubated eggs of the sand lizard Lacerta agilis at one of the four temperature regimes (27, 27 ± 2, 27 ± 4 and 27 ± 6 °C). We found that: (1) hatchlings incubated under the four temperature regimes did not differ from each other in any of the morphological and physiological traits examined; (2) interactions that included temperature treatment did not affect any trait examined; (3) the mean incubation length was longer in the 27 ± 6 °C treatment than in the other three treatments; and (4) female hatchlings were shorter in head length and width but longer in snout‐vent length as well as abdomen length than males derived from the same‐sized egg. Our data show that both the type and the magnitude of temperature variation can affect incubation length. We found no evidence for phenotypic divergence in responses to temperature fluctuations during incubation and therefore suggest that temperature variation does not affect the phenotype of hatchlings in L. agilis.  相似文献

The morphological traits and mitochondrial genes of sympatric sea slaters from the genus Ligia were analysed from samples collected along northeastern coastal China and adjacent coastal East Asia to assign the species and to obtain interpretations of their distributions. Using 338 sequences and 199 diagnoses of 16S polymerase chain reaction‐restriction fragment length polymorphism (PCR‐RFLP) from 331 specimens sampled from 26 localities, and homological sequences from counterparts from South Korea and the west coast of the USA, we assigned these taxa into two distinct species, Ligia exotica and Ligia cinerascens. Both species are recovered as two monophyletic clades in the robust phylogenetic hypothesis presented here. The northern boundaries of these species in China and the Korean peninsula are more southern than those of counterparts in Japan. Ligia exotica in northeastern coastal China is further split into two geographical subclades (B1 and B2), coexisting near 35°N. The distribution range of L. cinerascens covers the whole range of L. exotica subclade B1. Counterparts in Japan show a similar pattern. Oceanic rafting, shipping, damming and planting of seaweed are proposed as the media for the contemporary connectivity among populations of Ligia species inhabiting coastal China. © 2013 The Linnean Society of London  相似文献

Differential expression of ubiquitin was previously reported between Plutella xylostella strains that are resistant or susceptible to the pesticide deltamethrin (DM). This finding hinted at the potential involvement of ubiquitin in deltamethrin resistance, a theory that demanded further testing. Real-time PCR analyses revealed that one of the ubiquitin genes, UBL40, was overexpressed in the deltamethrin-resistant strain during the fourth instar. To investigate the functional relationship between this gene and deltamethrin resistance, RNA interference (RNAi) and cell transfection were utilised. UBL40 knockdown was observed to significantly reduce the level of resistance in RNAi-treated larvae after 48 h. Conversely, overexpression of UBL40 in Drosophila Kc cells conferred a degree of protection against deltamethrin. These results represent the first evidence that UBL40 plays a role in the regulation of deltamethrin resistance in P. xylostella.  相似文献
