The regenerating scale and tissues comprising the scale pocket of Fundulus olivaceus were examined microscopically at specific intervals. Scale removal resulted in a thickening of the epidermis which persisted through the early stages of regeneration. This thickening was due in part to the appearance of columnar basal cells which divided producing cells that became mucous cells and squamous cells. The scale regenerated as a relatively large plate of bone which first appeared between layers of scleroblasts on the floor of the scale pocket and then grew producing circuli and radii. By the fourth day of regeneration, calcium was observed in the cytoplasm of the scleroblasts and at randomly distributed foci in the osseous portion of the scale. The osseous layer was completely calcified by 15 days of regeneration.  相似文献

Birth seasonally at high latitudes is a complex phenomenon which is undoubtedly affected by a subtle interaction between environmental rhythmicity (most notably in photoperiod and temperature) and cultural adaption. There is intriguing evidence that human gonadotrophic activity (and hence fertility) may be affected by seasonal fluctuations in light intensity and duration. Nevertheless, cultural factors are important insofar as they mediate between environmental rhythmicity and human fertility/birth patterns. This article examines the distribution of births over several decades in an Inuit community located 300 miles north of the Arctic Circle. Several shifts in birth seasonality are noted, the most significant of which is a dramatic shift from pronounced seasonality in the 1970s to non-seasonality in the 1980s. Longitudinal ethnographic fieldwork has allowed an examination of social and economic changes accounting for the rather sudden disappearance of birth seasonality. These include increasing reliance upon wage employment and social assistance, decreased dependence upon subsistence hunting and trapping, changing attitudes on the part of young people entering their prime reproductive years, and the introduction of television, radio, and southern-style recreational activities.  相似文献

Blue Lake, an insoluble dye, was evaluated as an indicator of potential bacterial penetration into eggs. Various groups of eggs (fresh-laid; commercial; water-washed) were dipped in 0.25% (w/v) Blue Lake in 0.1% Triton X-100 solution for 2 min and incubated at room temperature up to 1 ± 24 h. Penetration was detected by counting the blue dots on shell membranes after breaking the eggs. Commercial eggs allowed the easiest penetration. All commercial eggs showed blue dots even at 2 min incubation and the average count reached 111/egg at 1 h. Water-washed eggs allowed much less penetration than commercial eggs and the counts of blue dots on those eggs were 10/egg at 2 min and 22 at 1 h. Fresh-laid eggs did not allow any penetration up to 24 h. Above results corresponded very well with the penetration study with Salmonella enteritidis and also with the morphological study of eggshell surfaces using electron microscopy where fresh-laid eggs had intact cuticle layers, but commercial eggs did not. Thus, Blue Lake dye might be used as a rapid indicator of bacterial penetration through eggshells.  相似文献

Loblolly pine (Pinus taeda L.) produces an induced defensive hypersensitive response in inner bark colonized by the southern pine beetle, Dendroctonus frontalis Zimm. (Coleoptera: Scolytidae), and its associated fungi. Adult beetles forced to colonize the induced response tissue in a laboratory study constructed galleries that were the same length as adults boring in normal phloem. However, each female laid fewer eggs in the induced tissue. Larval and pupal mortality were also higher in this tissue when compared to surrounding phloem. Beetles colonizing trees in response to pheromone baits constructed less gallery and laid fewer eggs in induced tissue than in surrounding normal phloem. These results suggest that the lesions produced by the induced defense system in conifers may not only contain the growth of fungi inoculated into trees during the attack phase of beetle colonization, but may also affect survival of bark beetle progeny.
Résumé Une réaction hypersensible de défense a été induite dans les couches profondes de l'écorce de P. taeda colonisée par D. frontalis et le champignon qui lui est associé. Au laboratoire, des adultes, contraints de coloniser les tissus où une réaction hypersensible e été induite, ont foré des galeries de même longueur que celles creusées dans du phloème sain. Cependant les femelles ont pondu moins d'oeufs dans les tissus induits; les mortalités larvaires et nymphales étaient aussi plus fortes dans ce tissu que dans le phloème voisin. Les scolytes, ayant colonisé les arbres après attraction par des pièges à phéromones, forent moins de galeries et pondent moins d'oeufs dans le tissu induit que dans le phloème sain voisin. Ces résultats suggèrent que les lésions, provoquées par le système de défense induit des conifères, peuvent non seulement limiter la croissance du champignon inoculé dans l'arbre au cours de la colonisation des scolytes, mais aussi affecter la survie des descendants dans l'écorce.

Acetate, a by-product of ethanol fermentation by Saccharomyces cerevisiae, has been shown to inhibit cell growth if present in high concentrations. Consequently, acetate has been considered undesirable in systems where the production rate depends upon steady-state growth. Acetate, however, may be desirable in some systems since it increases the specific rate of ethanol production by increasing the maintenance requirements of yeast. In immobilized cell reactors using the crosslinking method, steady state is not achieved and cell overgrowth is a problem. This article presents the results of a study aimed at taking advantage of the use of acetate, both to reduce cell overgrowth and to increase productivity. Various concentrations of acetate were added to batch and plug flow systems, while monitoring the effects on cell growth and ethanol production. The productivity was increased by as much as 50% in an immobilized cell reactor (ICR), while cell growth was greatly reduced.  相似文献

The effects of inoculum size on the kinetics of ethanol fermentation are not well defined in the literature. The purpose of this article is to examine the influence of the initial cell concentration on the modeling of ethanol inhibition. Experimental results show that increasing the inoculum level decreases the severity of ethanol inhibition. The effect of cell concentration can be related to the different inhibitory effects of autogeneously produced versus extracellularly added ethanol. On this basis, it is concluded that the extracellular ethanol concentration in the fermentation media is not the only variable to account for product inhibition modeling. Other fermentation parameters, such as yields and maintenance coefficients, are presented at different levels of initial cell concentration.  相似文献

The gas exchange properties of whole plant canopies are an integral part of crop productivity and have attracted much attention in recent years. However, insufficient information exists on the coordination of transpiration and CO2 uptake for individual leaves during the growing season. Single-leaf determinations of net photosynthesis (Pn), transpiration (E) and water use efficiency (WUE) for field-grown cotton (Gossypium hirsutum L.) leaves were recorded during a 2-year field study. Measurements were made at 3 to 4 day intervals on the main-stem and first three sympodial leaves at main-stem node 10 from their unfolding through senescence. Results indicated that all gas exchange parameters changed with individual main-stem and sympodial leaf age. Values of Pn, E and WUE followed a rise and fall pattern with maximum rates achieved at a leaf age of 18 to 20 days. While no significant position effects were observed for Pn, main-stem and sympodial leaves did differ in E and WUE particularly as leaves aged beyond 40 days. For a given leaf age, the main-stem leaf had a significantly lower WUE than the three sympodial leaves. WUE's for the main-stem and three sympodial leaves between the ages of 41 to 50 days were 0.85, 1.30, 1.36 and 1.95 μmol CO2 mmol−1 H2O, respectively. The mechanisms which mediated leaf positional differences for WUE were not strictly related to changes in stomatal conductance (gs·H2O) since decreases in gs·H2O with leaf age were similar for the four leaves. However, significantly different radiant environments with distance along the fruiting branch did indicate the possible involvement of mutual leaf shading in determining WUE. The significance of these findings are presented in relation to light competition within the plant canopy during development.  相似文献

Intrinsic growth and substrate uptake parameters were obtained for Peptostreptococcus productus, strain U-1, using carbon monoxide as the limiting substrate. A modified Monod model with substrate inhibition was used for modeling. In addition, a product yield of 0.25 mol acetate/mol CO and a cell yield of 0.034 g cells/g CO were obtained. While CO was found to be the primary substrate, P. productus is able to produce acetate from CO2 and H2, although this substrate could not sustain growth. Yeast extract was found to also be a growth substrate. A yield of 0.017 g cell/g yeast extract and a product yield of 0.14 g acetate/g yeast extract were obtained. In the presence of acetate, the maximum specific CO uptake rate was increased by 40% compared to the maximum without acetate present. Cell replication was inhibited at acetate concentrations of 30 g/l. Methionine was found to be an essential nutrient for growth and CO uptake by P. productus. A minimum amount of a complex medium such as yeast extract (0.01%) is, however, required.  相似文献

Embroys excised from dormant seeds of sugar pine ( Pinus lambertiana Dougl.) incubated at 25°C (non-dormancy-breaking) or stratified at 5°C (dormancy-breaking) were analyzed to determine temperature effects on the relative activities of respiration and fermentative metabolism, the levels of soluble sugers and the activities of the hydrolytic enzymes, invertase and sucrose synthase, as related to the release of dormancy and germinatio. At 25°C, despite a sharp drop in embryo oxygen uptake after 48 h, a simultaneous decline in acetaldehyde and ethanol concentrations indicated that there was not a shift to fermentative metabolism. The concentrations of soluble sugars showed no treatment effects. Embryo invertase (EC 3.2.1.26) activity changed only slightly at either temperature, while stratification was accompanied by a 4-fold increase in sucrose synthase (EC 2.4.1.13) activity (cleavage direction). Upon transfer of stratified seeds to 25°C, embryo sucrose synthase activity rapidly increased almost 10-fold, with the increase beginning prior to germination, while mvertase activity increased 20-fold, concomitant with germination.  相似文献

Factors involved in the dissolution of polyhedra of Autographa californica nuclear polyhedrosis virus (AcNPV) by digestive fluid collected from fifth stage Trichoplusia ni larvae were studied in vitro. Observations were made at timed intervals using phase contrast microscopy and transmission electron microscopy. When digestive fluid was heated at 50°C proteases retained activity. Exposure of polyhedra to digestive fluid previously heated to 50°C resulted in polyhedral matrix dissolution and envelope disruption in a manner similar to that of unheated digestive fluid, only delayed slightly. After exposure of polyhedra for 3 min, only enveloped virons were observed. Heating the digestive fluid to 60° or higher inactivated the proteases and altered the effect on polyhedra. Dissolution of the occlusion body matrix occurred but the polyhedral envelope remained and only a few weakened areas were observed in its structure. Within the polyhedral envelope, enveloped virons were not observed, only nucleocapsids and capsids. Exposure of polyhedra to 0.1 m sodium carbonate buffer at pHs of 9.5 or higher had effects similar to those of the digestive fluid with heat (60°C)-inactivated proteases. The addition of trypsin and chymotrypsin to the 0.1 m sodium carbonate buffer had no effect, while the addition of a bacterial protease (Streptomyces griseus) at pHs of 9.5 or higher resulted in dissolution of the matrix and disruption of the polyhedral envelope like the digestive fluid. Material infectious to TN-368 cells was obtained by exposure of AcNPV to T. ni digestive fluid. Maximum infectivity resulted from a 5-min exposure to unheated digestive fluid, with a dramatic decrease in infectivity with longer exposure. Exposure to digestive fluid with heat (60°C)-inactivated proteases resulted in a slower release of infectious material from the occlusion body, with a steady increase in the level of infectivity throughout the 30-min digestion period.  相似文献
