Binding of GnRH and its metal complexes to a sheep pituitary receptor have been investigated showing that Cu(II)-GnRH complex is more effectively bound to the receptor than the metal-free ligand, while Ni(II) and Co(II) complexes are less effective than the metal-free GnRH. Earlier studies have explained reasonably well the complex formation with cupric ion, while in this work extensive 1H NMR measurements have been performed for free gonadotropin-releasing hormone (GnRH) and its complexes with Ni(II) in DMSO (dimethyl sulfoxide) solution. This study shows the high order of organization of the metal-free peptide in DMSO solution with two structured 'domains' whose relative orientation is modulated by the mobility of the central glycine. Furthermore, theoretical calculations were performed for the Ni(II)-GnRH complex. The data obtained in this work supports previous studies on the co-ordination of Ni(II) ions with GnRH in aqueous solutions at high pH [J. Inorg. Biochem. 33 (1988) 11] and suggest an experimental procedure to reproduce high pH in DMSO solution. In the Ni(II) complex, the metal ion was found to co-ordinate with four nitrogen atoms inducing a well definite arrangement of aromatic side-chains and a rigid backbone structure.  相似文献

Salicylic acid (SA) mediates plant response to pathogen invasion, resulting in hypersensitive response and in the formation of systemic acquired resistance. It is well known that Nicotiana tabacum and other plants respond to Tobacco Mosaic Virus (TMV) infection by increasing the content of SA but the details of SA biosynthesis are still not fully understood. Generally, SA may originate directly from isochorismate ( Arabidopsis thaliana ), or its C6–C1 skeleton could be synthesized via the phenylpropanoid pathway by β-oxidation of trans -cinnamic acid ( N. tabacum ), 2-coumaric acid (OCA) ( Gaulteria procumbens , Lycopersicum esculentum ) or by retro-aldol reaction of trans -cinnamoyl-CoA ( Hypericum androsaemum ). We report here a novel putative enzyme activity from tobacco, salicylic aldehyde synthase (SAS), catalysing non-oxidative formation of salicylic aldehyde (SALD) directly from OCA. This chain-shortening activity is similar to that of 4-hydroxybenzaldehyde synthase from Vanilla planifolia , Lithospermum erythrorhizon , Daucus carota , Solanum tuberosum and Polyporus hispidus but the enzyme differs in the kinetics of the reaction, substrate specificity and requirements for reducing cofactors. SAS activity is constitutively expressed in healthy tobacco leaves and doubles as a result of infection with TMV. Moreover, the product of SAS activity—SALD, applied exogenously on tobacco leaves, stimulates peroxidase activity and enhances resistance to consecutive infection with TMV. These observations could suggest a contribution of SAS and SALD to the response of tobacco to TMV infection.  相似文献

During cold acclimation, winter rye ( Secale cereale L.) plants develop the ability to tolerate freezing temperatures by forming ice in intercellular spaces and xylem vessels. In this study, proteins were extracted from the apoplast of rye leaves to determine their role in controlling extracellular ice formation. Several polypeptides in the 15 to 32 kDa range accumulated in the leaf apoplast during cold acclimation at 5°C and decreased during deacclimation at 20°C. A second group of polypeptides (63, 65 and 68 kDa) appeared only when the leaves were maximally frost tolerant. Ice nucleation activity, as well as the previously reported antifreeze activity, was higher in apoplastic extracts from cold-acclimated than from nonacclimated rye leaves. These results indicate that apoplastic proteins exert a direct influence on the growth of ice. In addition, freezing injury was greater in extracted cold-acclimated leaves than in unextracted cold-acclimated leaves, which suggests that the proteins present in the apoplast are an important component of the mechanism by which winter rye leaves tolerate ice formation  相似文献

Abstract: P0 glycoprotein, the major protein of PNS myelin, contains approximately 1 mol of covalently bound long-chain fatty acids. To determine the chemical nature of the fatty acid-protein linkage, P0 was labeled in rat sciatic nerve slices with [3H]palmitic acid and subsequently treated with various reagents. The protein-bound palmi-tate was released by incubation with the reducing agents dithiothreitol and 2-mercaptoethanol, and with 1 M hydrox-ylamine at pH 7.5. In addition, P0 was deacylated by treatment with 10 m M NaBH4 with the concomitant production of [3H]hexadecanol, indicating that the fatty acid is bound in a thioester linkage. This conclusion was supported further by the fact that deacylation with hydroxylamine generated free thiol groups, which were titrated with [14C]-iodoacetamide. To identify the cysteine residue involved in the thioester linkage, [14C]carboxyamidomethylated P0was digested with trypsin and the resulting peptides analyzed by reversed-phase HPLC. Identification of the radioactive protein fragments by amino acid analysis and amino-terminal peptide sequencing revealed that Cys153 in rat P0 glycoprotein is the acylation site. The acylated cysteine is located at the junction of the putative transmem-brane and cytoplasmic domains. This residue is also present in the P0 glycoprotein of other species, including human, bovine, mice, and chicken.  相似文献

Metabolic profiles of amniotic fluid and maternal blood are sources of valuable information about fetus development and can be potentially useful in diagnosis of pregnancy disorders. In this study, we applied 1H NMR-based metabolic profiling to track metabolic changes occurring in amniotic fluid (AF) and plasma (PL) of healthy mothers over the course of pregnancy. AF and PL samples were collected in the 2nd (T2) and 3rd (T3) trimester, prolonged pregnancy (PP) until time of delivery (TD). A multivariate data analysis of both biofluids reviled a metabolic switch-like transition between 2nd and 3rd trimester, which was followed by metabolic stabilization throughout the rest of pregnancy probably reflecting the stabilization of fetal maturation and development. The differences were further tested using univariate statistics at α = 0.001. In plasma the progression from T2 to T3 was related to increasing levels of glycerol, choline and ketone bodies (3-hydroxybutyrate and acetoacetate) while pyruvate concentration was significantly decreased. In amniotic fluid, T2 to T3 transition was associated with decreasing levels of glucose, carnitine, amino acids (valine, leucine, isoleucine, alanine, methionine, tyrosine, and phenylalanine) and increasing levels of creatinine, succinate, pyruvate, choline, N,N-dimethylglycine and urocanate. Lactate to pyruvate ratio was decreased in AF and conversely increased in PL. The results of our study, show that metabolomics profiling can be used to better understand physiological changes of the complex interdependencies of the mother, the placenta and the fetus during pregnancy. In the future, these results might be a useful reference point for analysis of complicated pregnancies.  相似文献

The genetic diversity of all available culture strains of the Tribonemataceae ( Stramenopiles , Xanthophyceae ) from Antarctica was assessed using the chloroplast-encoded psbA /rbcL spacer region sequences, a highly variable molecular marker, to test for endemism when compared with their closest temperate relatives. There was no species endemic for Antarctica, and no phylogenetic clade corresponded to a limited geographical region. However, species of the Tribonemataceae may have Antarctic populations that are distinct from those of other regions because the Antarctic strain spacer sequences were not identical to sequences from temperate regions. Spacer sequences from five new Antarctic isolates were identical to one or more previously available Antarctic strains, indicating that the Tribonemataceae diversity in Antarctic may be rather limited. Direct comparisons of the spacer sequences and phylogenetic analyses of the more conserved rbcL gene revealed that current morphospecies were inadequate to describe the actual biodiversity of the group. For example, the genus Xanthonema , as currently circumscribed, was paraphyletic. Fortunately, the presence of distinctive sequence regions within the psbA/rbcL spacer, together with differences in the rbcL phylogeny, provided significant autoapomorphic criteria to re-define the Tribonemataceae species.  相似文献

Ammonium assimilation enzymes from several strains of ectendo- and ectomycorrhizal fungi were assayed after three weeks culture on a buffered synthetic medium containing ammonium as sole nitrogen source. Activity of NADP-dependent glutamate dehydrogenase (GDH, EC 1.4.1.4) of ectomycorrhizal strains was very low despite excellent mycelial growth. Only ectendomycorrhizal fungus MrgX isolated from roots of Pinus sylvestris showed high GDH activity. Similar results were obtained when the enzyme extracts were subjected to starch gel electrophoresis. Growth of the fungi, except ectendomycorrhizal MrgX, was arrested when inhibitors of glutamine synthetase (GS, EC 6.3.1.2) or glutamate synthase (GOGAT. EC 1.4.7.1) (methionine sulphoximine or albizine, respectively) were included in the culture medium. Glutamine synthetase activity was found in all fungi tested. The results suggest that the GS pathway for ammonium assimilation is potentially operative in ectomycorrhizal fungi and imply only a minor role for GDH in ammonium assimilation by the studied ectomycorrhizal symbionts of pine. Some physiological and ecological implications of these results are discussed.  相似文献

The work of adhesion between water and a heptane solution of phospholipids extracted from spring (cv. Jara) and winter (cv. Grana) wheat ( Triticum aestivum L.) grown at 2°C and 20°C was measured in order to determine the effect of changes in the composition of fatty acids on the hydrophilic properties of the lipid bilayer of membranes. The work of adhesion water/phospholipids was positively correlated with the contents of the oleic and linoleic acids (18:1 and 18:2, respectively) and negatively correlated with the content of linolenic acid (18:3) in the phospholipids. The sensitivity of the work of adhesion to changes in the particular fatty acids decreased with increasing number of double bonds. In addition, it depended on the genotype and on the temperature of growth of the seedlings.  相似文献

SUMMARY 1. A biomanipulation experiment was carried out in a small (10 ha), but relatively deep (17 m) and highly eutrophic lake in northern Poland. The lake had been stocked in 1996, 1997 and 1998 with a variety of piscivorous fish (pike, catfish, trout and pikeperch), in order to reduce numbers of cyprinid planktivores.
2. Piscivore stocking was associated with a threefold decrease in the offshore fish density (night echosounding). Despite this reduction, the large planktonic cladoceran, Daphnia hyalina , remained scarce, whereas the density of small-sized zooplankton increased greatly.
3. The lack of demographic response in D. hyalina was probably due to the anoxia in the hypolimnetic refuge of this vertically migrating species. The anoxic hypolimnion, below 3–4 m depth, was inhabited day and night by numerous Chaoborus flavicans larvae.
4. Changes in zooplankton were associated with shifts in the taxonomic composition (from single-cell green algae to filamentous cyanobacteria), size structure (from nano- to net phytoplankton) and seasonal dynamics of phytoplankton, but not in the average biomass of planktonic algae. A clear-water phase, which was absent in the prestocking years, developed in spring, with Secchi depth reaching 2.5 m, a value which had never been recorded in the 20 years preceding the biomanipulation. In general, the lake's status was switched from hypertrophic to eutrophic.
5. Deteriorating food conditions, resulting from qualitative changes in the phytoplankton community, combined with predation pressure by the remaining fish and Chaoborus larvae were associated with the ultimate elimination of D. hyalina from the lake.  相似文献
