The phylogeny of the Rhabdocoela, a species-rich taxon of free-living flatworms, is reconstructed based on complete 18S rDNA sequences. The analysis includes 62 rhabdocoels and 102 representatives of all major flatworm taxa. In total, 46 new sequences are used, 41 of them from rhabdocoel species, five from proseriates. Phylogenetic analysis was performed using maximum parsimony and Bayesian inference. Clade support was evaluated with parsimony jackknifing, Bremer support indices and Bayesian posterior probabilities. The resulting cladogram corroborates that the Rhabdocoela is monophyletic, but its sister group remains uncertain. The 'Dalyellioida' and the 'Typhloplanoida', both former rhabdocoel subtaxa, are polyphyletic. Within the Rhabdocoela the monophyletic Kalyptorhynchia, characterized by a muscular proboscis, forms the sister group of all other rhabdocoels. The Schizorhynchia is a monophyletic subtaxon of the Kalyptorhynchia, with the split proboscis as a synapomorphy. Except for the Dalyelliidae and the Typhloplanidae, both freshwater taxa, none of the 'families' previously included in the 'Typhloplanoida' and the 'Dalyellioida' appears to be monophyletic. As a result of this analysis, three existing and four new taxon names are formally defined following the rules of the Phylocode.  相似文献

Biomarkers in CSF can offer improved diagnostic accuracy for Alzheimer's disease (AD). The present study investigated whether the glycoprotein and putative tumor suppressor Dickkopf homolog 3 (Dkk-3) is secreted into CSF and evaluated its applicability as a diagnostic marker for AD. Using our highly specific immunoenzymometric assay, Dkk-3 levels were measured in plasma and/or CSF of patients suffering from depression, mild cognitive impairment (MCI), or AD and compared with healthy subjects. Dkk-3 identity was verified by western blot and matrix-assisted laser desorption/ionization time-of-flight (MALDI-TOF) mass spectrometry (MS)/MS. High concentrations of Dkk-3 were detected in CSF compared with plasma (28.2 ± 1.3 vs. 1.22 ± 0.04 nmol/L, respectively). Consistently Dkk-3 expression was demonstrated in neurons of the cortex and epithelial cells of the choroid plexus, the major source of CSF. Significantly increased Dkk-3 levels in plasma and CSF were observed for AD patients compared with healthy subjects but not patients suffering from MCI or depression. In summary, our data indicate that elevated Dkk-3 levels are specifically associated with AD and might serve as a potential non-invasive AD biomarker in plasma.  相似文献

Chaenotheca phaeocephala and C. subroscida have been characterized by differences in their morphology and ecology. They are, however, morphologically plastic and sometimes seem to intergrade. It has been suspected that the differences in morphology between these two taxa could possibly be phenotypic and caused by their occurrence in different habitats and/or by the association with different photobionts. To test if there is a genetic difference between the species ITS1–5.8S-ITS2 rDNA sequences of six specimens of the mycobiont of C. phaeocephala and five of C. subroscida were compared. Further the ITS1–5.8S-ITS2 rDNA sequences of the photobiont of one specimens of C. phaeocephala and six of C. subroscida were compared to each other and to other species of Trebouxia. The mycobiont sequences of both C. phaeocephala and C. subroscida were monophyletic, substantially different and the groups had strong supports. They also proved to be associated with different Trebouxia species, C. phaeocephala with T. arboricola and Chaenotheca subroscida with T. jamesii. The morphologically based concepts of C. phaeocephala and C. subroscida thus not merely represent phenotypic variation but also are associated with considerable genetic differences in the extremes of the ITS1–5.8S-ITS2 rDNA region.  相似文献

AimAlthough patterns of biodiversity across the globe are well studied, there is still a controversial debate about the underlying mechanisms and their generality across biogeographic scales. In particular, it is unclear to what extent diversity patterns along environmental gradients are directly driven by abiotic factors, such as climate, or indirectly mediated through biotic factors, such as resource effects on consumers.LocationAndes, Southern Ecuador; Mt. Kilimanjaro, Tanzania.MethodsWe studied the diversity of fleshy‐fruited plants and avian frugivores at the taxonomic level, that is, species richness and abundance, as well as at the level of functional traits, that is, functional richness and functional dispersion. We compared two important biodiversity hotspots in mountain systems of the Neotropics and Afrotropics. We used field data of plant and bird communities, including trait measurements of 367 plant and bird species. Using structural equation modeling, we disentangled direct and indirect effects of climate and the diversity of plant communities on the diversity of bird communities.ResultsWe found significant bottom‐up effects of fruit diversity on frugivore diversity at the taxonomic level. In contrast, climate was more important for patterns of functional diversity, with plant communities being mostly related to precipitation, and bird communities being most strongly related to temperature.Main conclusionsOur results illustrate the general importance of bottom‐up mechanisms for the taxonomic diversity of consumers, suggesting the importance of active resource tracking. Our results also suggest that it might be difficult to identify signals of ecological fitting between functional plant and animal traits across biogeographic regions, since different species groups may respond to different climatic drivers. This decoupling between resource and consumer communities could increase under future climate change if plant and animal communities are consistently related to distinct climatic drivers.  相似文献

There is a wealth of smaller-scale studies on the effects of forest management on plant diversity. However, studies comparing plant species diversity in forests with different management types and intensity, extending over different regions and forest stages, and including detailed information on site conditions are missing. We studied vascular plants on 1500 20 m × 20 m forest plots in three regions of Germany (Schwäbische Alb, Hainich-Dün, Schorfheide-Chorin). In all regions, our study plots comprised different management types (unmanaged, selection cutting, deciduous and coniferous age-class forests, which resulted from clear cutting or shelterwood logging), various stand ages, site conditions, and levels of management-related disturbances. We analyzed how overall richness and richness of different plant functional groups (trees, shrubs, herbs, herbaceous species typically growing in forests and herbaceous light-demanding species) responded to the different management types. On average, plant species richness was 13% higher in age-class than in unmanaged forests, and did not differ between deciduous age-class and selection forests. In age-class forests of the Schwäbische Alb and Hainich-Dün, coniferous stands had higher species richness than deciduous stands. Among age-class forests, older stands with large quantities of standing biomass were slightly poorer in shrub and light-demanding herb species than younger stands. Among deciduous forests, the richness of herbaceous forest species was generally lower in unmanaged than in managed forests, and it was even 20% lower in unmanaged than in selection forests in Hainich-Dün. Overall, these findings show that disturbances by management generally increase plant species richness. This suggests that total plant species richness is not suited as an indicator for the conservation status of forests, but rather indicates disturbances.  相似文献

Although Eurycorypha is the most species-rich Phaneropterinae genus in Africa, little is known about the distribution and the ecology of the species. In this study data on distribution, ecology, song and on chromosomes of some East African species are provided. The nymphs of some species are shown, famous for their ant-like appearance and behaviour. The male of E. punctipennis Chopard and three species of Eurycorypha are newly described. These are E. resonans n. sp. and E. combretoides n. sp. occurring in different habitats on Mount Kilimanjaro, and E. conclusa n. sp from forest habitats along the Tanzanian coast. Bioacoustically, the four recorded species are unusually divers, presenting non-resonant and resonant songs. As four Eurycorypha species occur syntopically on geological young Mount Kilimanjaro, the biogeographical pattern and possible speciation mechanisms in the genus Eurycorypha are discussed in context of the climatical history of eastern Africa.  相似文献

Seed mass is one of the most important plant traits. It is strongly related to plant fitness and life-history strategy, and is one of the key determinants of the ability of plants to spread and thus to respond to changing environments. While substantial empirical work has been devoted to understanding seed-mass variation across species, we know less about seed-mass variation within species, its geographical and ecological differentiation, and the degree to which it is influenced by environmental change. Here, we studied intraspecific variability in seed mass of six common grassland plants (Arrhenatherum elatius, Bromus hordeaceus, Cerastium holosteoides, Heracleum sphondylium, Trifolium repens and Veronica chamaedrys) across three regions in Germany and a broad range of land-use types and intensities. We found substantial seed-mass variation among regions, populations, and individuals within all of the studied species. In five species, seed mass had a strong and consistent geographic component, and in three species we found significant effects of land use – fertilisation, grazing intensity or mowing frequency – on seed mass. In several species, land use and geographic region not only affected mean seed mass, but also the variability of seed mass within populations. Our study demonstrates that seed mass is geographically and ecologically differentiated in common grassland species. It is likely that both phenotypic plasticity and genetic factors contribute to this differentiation. Our results also show that seed mass is a highly variable trait with typically around 10-fold variation within species.  相似文献

RAFT polymerization successfully controlled the synthesis of phosphonium-based AB diblock copolymers for nonviral gene delivery. A stabilizing block of either oligo(ethylene glycol(9)) methyl ether methacrylate or 2-(methacryloxy)ethyl phosphorylcholine provided colloidal stability, and the phosphonium-containing cationic block of 4-vinylbenzyltributylphosphonium chloride induced electrostatic nucleic acid complexation. RAFT polymerization generated well-defined stabilizing blocks (M(n) = 25000 g/mol) and subsequent chain extension synthesized diblock copolymers with DPs of 25, 50, and 75 for the phosphonium-containing block. All diblock copolymers bound DNA efficiently at ± ratios of 1.0 in H(2)O, and polyplexes generated at ± ratios of 2.0 displayed hydrodynamic diameters between 100 and 200 nm. The resulting polyplexes exhibited excellent colloidal stability under physiological salt or serum conditions, and they maintained constant hydrodynamic diameters over 24 h. Cellular uptake studies using Cy5-labeled DNA confirmed reduced cellular uptake in COS-7 and HeLa cells and, consequently, resulted in low transfection in these cell lines. Serum transfection in HepaRG cells, which are a predictive cell line for in vivo transfection studies, showed successful transfection using all diblock copolymers with luciferase expression on the same order of magnitude as Jet-PEI. All diblock copolymers exhibited low cytotoxicity (>80% cell viability). Promising in vitro transfection and cytotoxicity results suggest future studies involving the in vivo applicability of these phosphonium-based diblock copolymer delivery vehicles.  相似文献
