Morphometric differentiation among freshwater fish populations is a commonplace occurrence, although the underlying causes for this divergence often remain elusive. We analysed the degree and patterns of morphological differentiation among nine freshwater three‐spined stickleback (Gasterosteus aculeatus) populations inhabiting isolated karst rivers of the Adriatic Sea basin, to characterise the phenotypic diversity and differentiation in these populations. The analyses revealed marked and significant morphometric differentiation – especially in traits related to predator defence amongst most populations – even among those located within close geographic proximity in the same catchment system. Accordingly, the degree of morphometric and neutral genetic differentiation, as assessed from variability in 15 microsatellite loci from a parallel study, were uncorrelated across the populations. However, PST/FST comparisons revealed that the degree of phenotypic differentiation (PST) among populations exceeded that to be expected from genetic drift alone, suggesting a possible adaptive basis for the observed differentiation. In fact, avian predation pressure and several physiochemical environmental variables were identified as the main putative drivers of the observed differentiation, particularly in the dorsal spines, ascending process and lateral plates. The olive (Olea europaea L. subsp. europaea) is an old traditional crop which was domesticated from the wild at various locations around the Mediterranean basin, resulting in a vast number of accessions worldwide. This is the case of the northeast part of Spain (Aragon), where 163 genotypes were prospected and characterised. The majority of these genotypes (90 %) seem to be autochthonous accessions of this area and the rest are synonyms of accessions from other areas of Spain. In this study, 11 nuclear SSR markers were used to discriminate between the accessions and to understand the genetic relationship among them. All primers produced a successful amplification giving a total of 176 fragments in the genotypes studied, with an average of 16 alleles per SSR, ranging from 5 (DCA13) to 24 (DCA11). Allele size ranged from 120 bp at locus UDO99 to 284 bp at locus DCA15. The dendrogram generated using the variability observed classified most of the genotypes according to their geographical origin (Huesca, Teruel and Zaragoza provinces), confirming the particular evolution of different olive ecotypes. Structure software was used to investigate the genetic population structure among olive accessions, showing that the maximum rate of change in the log probability of the data occurred at K = 2. In addition, the SSR markers have consequently shown their usefulness for cultivar identification in olive, for establishing the genetic closeness among its accessions, and for establishing genealogical relationships.  相似文献

Mining is a relatively highly monitored industry. While chemical pollutants (toxic ions, radionuclides, etc.) have mostly been eliminated from mining waters, other types of environmental pollution (temperature regime alterations, high concentrations of various anions, etc.) can affect benthic invertebrates. In this study, we focused on the effect of mining water effluent on the diversity and density of aquatic Clitellata. Four sampling sites were selected. Three sites in a natural stream (the Nedvědi?ka River, Czech Republic), one upstream and two downstream from the mining effluent, and one site on the mining waters were sampled monthly during 2008–2009. Environmental variables were recorded in and samples were collected from two types of habitats — riffles and pools. The response of clitellate assemblages was evaluated using principal component analysis and generalised estimating equations. The results indicated that the mining effluent caused partial species exchange and had negative effects on clitellate taxa richness and abundance. These responses were specific to both the habitat (riffle/pool) and species sampled. In each of the different taxa studied, we observed one of four typical clitellate responses: (a) elimination of stenotherm species; (b) reduction of clitellate species followed by quick recovery; (c) neutral response; or (d) positive influence. We found that aquatic clitellates, which are considered to be eurytopic with broad ecological valences, are also sensitive to even slight environmental pollution.  相似文献

Sphingosine [(2S, 3R, 4E)-2-amino-4-octadecen-1, 3-diol] is the most common sphingoid long chain base in sphingolipids. It is the precursor of important cell signaling molecules, such as ceramides. In the last decade it has been shown to act itself as a potent metabolic signaling molecule, by activating a number of protein kinases. Moreover, sphingosine has been found to permeabilize phospholipid bilayers, giving rise to vesicle leakage. The present contribution intends to analyze the mechanism by which this bioactive lipid induces vesicle contents release, and the effect of negatively charged bilayers in the release process. Fluorescence lifetime measurements and confocal fluorescence microscopy have been applied to observe the mechanism of sphingosine efflux from large and giant unilamellar vesicles; a graded-release efflux has been detected. Additionally, stopped-flow measurements have shown that the rate of vesicle permeabilization increases with sphingosine concentration. Because at the physiological pH sphingosine has a net positive charge, its interaction with negatively charged phospholipids (e.g., bilayers containing phosphatidic acid together with sphingomyelins, phosphatidylethanolamine, and cholesterol) gives rise to a release of vesicular contents, faster than with electrically neutral bilayers. Furthermore, phosphorous 31-NMR and x-ray data show the capacity of sphingosine to facilitate the formation of nonbilayer (cubic phase) intermediates in negatively charged membranes. The data might explain the pathogenesis of Niemann-Pick type C1 disease.  相似文献

We studied the properties of bilayers formed by ether-and ester-containing phospholipids, whose hydrocarbon chains can be either linear or branched, using sn-1,2 dipalmitoyl, dihexadecyl, diphytanoyl, and diphytanyl phosphatidylcholines (DPPC, DHPC, DPhoPC, and DPhPC, respectively) either pure or in binary mixtures. Differential scanning calorimetry and confocal fluorescence microscopy of giant unilamellar vesicles concurred in showing that equimolar mixtures of linear and branched lipids gave rise to gel/fluid phase coexistence at room temperature. Mixtures containing DHPC evolved in time (0.5 h) from initial reticulated domains to extended solid ones when an equilibrium was achieved. The nanomechanical properties of supported planar bilayers formed by each of the four lipids studied by atomic force microscopy revealed average breakdown forces Fb decreasing in the order DHPC ≥ DPPC > DPhoPC >> DPhPC. Moreover, except for DPPC, two different Fb values were found for each lipid. Atomic force microscopy imaging of DHPC was peculiar in showing two coexisting phases of different heights, probably corresponding to an interdigitated gel phase that gradually transformed, over a period of 0.5 h, into a regular tilted gel phase. Permeability to nonelectrolytes showed that linear-chain phospholipids allowed a higher rate of solute + water diffusion than branched-chain phospholipids, yet the former supported a smaller extent of swelling of the corresponding vesicles. Ether or ester bonds appeared to have only a minor effect on permeability.  相似文献

The trimeric light-harvesting complexes II (LHCII) of plants and green algae are pigment-protein complexes involved in light harvesting and photoprotection. Different conformational states have been proposed to be responsible for their different functions. At present, detergent-solubilized LHCII is used as a model for the “light-harvesting conformation”, whereas the “quenched conformation” is mimicked by LHCII aggregates. However, none of these conditions seem to perfectly reproduce the properties of LHCII in vivo. In addition, several monomeric LHC complexes are not fully stable in detergent. There is thus a need to find conditions that allow analyzing LHCs in vitro in stable and, hopefully, more native-like conformations. Here, we report a study of LHCII, the major antenna complex of plants, in complex with amphipols. We have trapped trimeric LHCII and monomeric Lhcb1 with either polyanionic or non-ionic amphipols and studied the effect of these polymers on the properties of the complexes. We show that, as compared to detergent solutions, amphipols have a stabilizing effect on LHCII. We also show that the average fluorescence lifetime of LHCII trapped in an anionic amphipol is ~30 % shorter than in α-dodecylmaltoside, due to the presence of a conformation with 230-ps lifetime that is not present in detergent solutions.  相似文献

Electroporation is a method of treatment of plant tissue that due to its nonthermal nature enables preservation of the natural quality, colour and vitamin composition of food products. The range of processes where electroporation was shown to preserve quality, increase extract yield or optimize energy input into the process is overwhelming, though not exhausted; e.g. extraction of valuable compounds and juices, dehydration, cryopreservation, etc. Electroporation is—due to its antimicrobial action—a subject of research as one stage of the pasteurization or sterilization process, as well as a method of plant metabolism stimulation. This paper provides an overview of electroporation as applied to plant materials and electroporation applications in food processing, a quick summary of the basic technical aspects on the topic, and a brief discussion on perspectives for future research and development in the field. The paper is a review in the very broadest sense of the word, written with the purpose of orienting the interested newcomer to the field of electroporation applications in food technology towards the pertinent, highly relevant and more in-depth literature from the respective subdomains of electroporation research.  相似文献

Helix is a genus of large Western Palaearctic land snails, particularly diverse in the Mediterranean region. Despite the large size and attractiveness of its members, it has an unsettled taxonomy, and no data are available on its intrageneric phylogenetic relationships. One of the problematic Helix taxa is the widely distributed, economically important, and conchologically very variable H. lucorum. Two distinct forms may be encountered under this name in the Apennine Peninsula: a typical one in the north, and a form originally described as Helix straminea in central and southern Italy. To evaluate the status of H. straminea and its relationships to Italian and Balkan Helix fauna, we combined shell morphology, geometric morphometrics, and phylogenetic analysis based on two mitochondrial genes. Distribution data were improved by drawing information from unambiguously identifiable photographs posted online. Based on our results, Helix straminea is redescribed as a separate species, and we find it to have a disjunctive trans‐Adriatic range with closest relatives in the western Balkans. We provide insight into relationships and intraspecific variability within Helix, a first step towards a comprehensive revision of the genus. On the example of Italian Helix fauna, we demonstrate how understanding of snail zoogeography may change with improving taxonomy. © 2014 The Linnean Society of London  相似文献
