The role and the importance of the sulfonate moiety in phenyl 4-(2-oxo-3-alkylimidazolidin-1-yl)benzenesulfonates (PAIB-SOs) were assessed using its bioisosteric sulfonamide equivalent leading to new cytochrome P450 1A1 (CYP1A1)-activated prodrugs designated as 4-(3-alkyl-2-oxoimidazolidin-1-yl)-N-phenylbenzenesulfonamides (PAIB-SAs). PAIB-SAs are active in the submicromolar to low micromolar range showing selectivity toward CYP1A1-expressing MCF7 cells as compared to cells devoid of CYP1A1 activity such as MDA-MB-231 and HaCaT cells. The most potent, PAIB-SA 13, bearing a trimethoxyphenyl group on ring B blocks the cell cycle progression in G2/M phase, disrupts the microtubule dynamics and is biotransformed by CYP1A1 into CEU-638, its potent antimicrotuble counterpart. Structure-activity relationships related to PAIB-SOs and PAIB-SAs evidenced that PAIB-SOs and PAIB-SAs are true bioisosteric equivalents fully and selectively activatable by CYP1A-expressing cells into potent antimitotics.  相似文献

Parasitoid wasps (Hymenoptera) play a significant role in reducing the pest population of the bagworm species Metisa plana. This study presents the abundance and DNA barcoding information of eight parasitoid wasps species, Dolichogenidea metesae (47%), Brachymeria carinata (19%), Buysmania oxymora (12%), Goryphus bunoh (7%), Pediobius anomalus (5%), Eupelmus cotoxanthae (2%), Apanteles aluella (5%), Apanteles sp.1 (3%) and that emerged from M. plana collected from three highly infested oil palm plantations in Selangor (west), Perak (north) and Johor (south) in Peninsular Malaysia. Samples of infested M. plana were collected from the field and reared in a rearing room. The parasitoid wasp species D. metesae recorded the highest emergence numbers and the broadest presence in all the sampling sites. The relationships among the parasitoids species were estimated and visualized using Neighbor Joining (NJ) phylogenetic analyses with the Ceraphronidae family as an outgroup. The resulting NJ tree showed that the identified parasitoid wasps were successfully classified into specific species and supported with bootstraps values between 55% to 100%. This study provides important information on potential biological control agents for M. plana that may be useful for the Malaysian oil palm industry.  相似文献

The SH group of 2-mercaptoethylpyridine adds to the double bond of dehydroalanine residues in alkali-treated casein and acetylated casein to form S-β-(2-pyridylethyl)-L-cysteine (2-PEC) side chains. The generated cysteine derivative can be assayed, after protein hydrolysis, by standard amino acid analysis techniques. The novel transformation may be useful for assessing chemical and biological functions of dehydroalanine residues in proteins.  相似文献

Eight derivatives of cysteine and penicillamine with 2- and 4-vinyl-pyridine, p-nitrobenzyl bromide, and p-nitrostyrene were evaluated as potential internal standards for the short and long (physiological) basic columns in amino acid analysis by ion-exchange chromatography. S-β-(4-pyridylethyl)-dl-penicillamine (4-PEP) was found to have an advantage over the previously proposed S-β-(4-pyridylethyl)-l-cysteine (4-PEC) since the elution position of 4-PEP on the short basic column is insensitive to minor changes in pH of the eluting citrate buffer. 4-PEP was found to be stable to acid hydrolysis as used for proteins and its recovery from protein hydrolysates was unaffected by the presence of starch during hydrolysis. However, an extra 14 min is required to elute 4-PEP on the short column.Of the eight compounds studied, six appcar suitable as internal standards on the physiological (long) column. These elute in widely differing positions between histidine and arginine, thus offering a choice of internal standards for special analysis on the basic long column.  相似文献

Xylella fastidiosa has been reported as responsible for a devastating disease on olive trees in Apulia region (south‐eastern Italy), characterized by a quick decline syndrome. In Lebanon, the pathogen was recently associated with leaf scorch symptoms on oleander, and reports on leaf scorch and dieback of olive trees branches by technicians and farmers have shown an increasing trend in the main agricultural areas. To assess the occurrence and distribution of the pathogen in Lebanon, samples of twigs from olive trees (82), olive seedlings (26), grapevine (30), oleander (32) and ornamentals imported from Italy (48) were analysed by isolation on four agarized media, serological techniques (ELISA and DTBIA) using Xylella fastidiosa‐specific antibodies and by PCR, using three specific sets of primers. Results unequivocally demonstrated that all the collected samples were free from the pathogen. As well, both detection protocols and attempts at isolating the pathogen on agarized media demonstrated that oleander samples gathered from American University campus in Beirut, where X. fastidiosa was previously reported, were not infected. Nevertheless, continuous monitoring and rigorous control measures of propagative materials are necessary to prevent the introduction of Xylella fastidiosa in Lebanon.  相似文献

Muscle paths play an important role in musculoskeletal simulations by determining a muscle’s length and how its force is distributed to joints. Most previous approaches estimate the way in which muscles ‘wrap’ around bones and other structures with smooth analytical wrapping surfaces. In this paper, we employ Newton’s method with discrete differential geometry to permit muscle wrapping over arbitrary polygonal mesh surfaces that represent underlying bones and structures. Precomputing distance fields allows us to speed up computations for the common situation where many paths cross the same wrapping surfaces. We found positive results for the accuracy, robustness, and efficiency of the method. However the method did not exhibit continuous changes in path length for dynamic simulations. Nonetheless this approach provides a valuable step toward fast muscle wrapping on arbitrary meshes.  相似文献

Phosphoinositide 3-kinase (PI3K) mediates essential functions of vascular endothelial growth factor (VEGF), including the stimulation of endothelial cell proliferation and migration. Nevertheless, the mechanisms coupling the receptor VEGFR-2 to PI3K remain obscure. We observed that the Grb2-bound adapter Gab1 is tyrosine-phosphorylated and relocated to membrane fractions upon VEGF stimulation of endothelial cells. We could detect the PI3K regulatory subunit p85 in immunoprecipitates of endogenous Gab1, and vice versa, and measure a Gab1-associated lipid kinase activity upon VEGF stimulation. Furthermore, transfection of the Gab1-YF3 mutant lacking all p85-binding sites strongly repressed PI3K activation measured in vitro. Moreover, Gab1-YF3 severely decreased the cellular amount of phosphatidylinositol 3,4,5-trisphosphate (PtdIns(3,4,5)P3) generated in response to VEGF. Furthermore, adenoviral expression of Gab1-YF3 suppressed both Akt phosphorylation and recovery of wounded human umbilical vein endothelial cell monolayers, a VEGF-dependent process involving cell migration and proliferation under PI3K control. Transfection of other Gab1 mutants, lacking Grb2-binding sites or the pleckstrin homology (PH) domain, also prevented Akt activation, further demonstrating Gab1 involvement in PI3K activation. These mutants were also used to show that interactions with both Grb2 and PtdIns(3,4,5)P3 mediate Gab1 recruitment by VEGFR-2. Importantly, Gab1 mobilization was impaired by (i) PI3K inhibitors, (ii) deletion of Gab1 PH domain, (iii) PTEN (phosphatase and tensin homolog deleted on chromosome 10) overexpression to repress PtdIns(3,4,5)P3 production, and (iv) overexpression of a competitor PH domain for PtdIns(3,4,5)P3 binding, which altogether demonstrated that PI3K is also an upstream regulator of Gab1. Gab1 thus appears as a primary actor in coupling VEGFR-2 to PI3K/Akt, recruited through an amplification loop involving PtdIns(3,4,5)P3 and its PH domain.  相似文献
