Summary An alternative approach to the regeneration of coenzymes using immobilized hydrogen dehydrogenase (hydrogenase) is described. Hydrogenase isolated from Alcaligenes Eutrophus was immobilized to porous glass particles and used in combination with alanine dehydrogenase for formation of alanine, while the NADH consumed was regenerated by molecular hydrogen. Different physical arrangements of the two enzymes were compared. Alanine was conveniently assayed with a specially designed enzyme thermistor method.  相似文献

The establishment of permanent T-lymphocyte cell lines by transformation with DNA viruses has not yet been achieved. This paper reports the successful transfer of polyoma virus genome into T-lymphocyte cells by somatic hybridization. A T-lymphocyte clone, HB1, derived from (DBA/ 2J×AKR) spleen cells, isolated in vitro by cloning in semi-solid agar, was fused with a polyoma (Py) virus-transformed fibroblast C3HPy, clone 1. The authenticity of the hybrid C3H/HB was established by chromosome and histocompatibility antigen studies. This initial population and the various clones retained T-lymphocyte characteristics such as morphological appearance, growth properties (suspension culture) and differentiation antigen (Thy 1–2). The hybrid cell line and the various clones presented all the characteristics of Py transformation. Namely, they carried the Py genome originating from the fibroblastic parent and maintained Py virus tumour-associated antigens (TSTA, TSSA and T antigens). In most respects, this hybrid population resembled the C3HPy/C11 parent and exhibited the same tumorigenicity.  相似文献

A lot of numerous rhynchonellid shells referedto the main species of genus SphenorhynchiaBuckman have been sampled in stratigraphically well-defined beds of Dogger period in the Mâconnais and in the Jura (France). Here, morphologic and anatomic characters and variability of S. plicatella, S. matisconensis, S. bugeysiaca, S. ferryi and S. dominula are studied.The main interest of this revision is to establish a complete diagnosis of the genus, not studied since its creation in 1918, with very limited knowledge of the internal structures. The settlement of this diagnosis is based on the type-species, S. plicatella, sometimes a little «unconventional form (beak, outline of the young shells, orientation of dental lamellae, delthyrial cavity), and also on the other species' characters. So, I expect that a later diagnosis will be avoided.Other advantages are observations on the taxonomic place of Sphenorhynchia in the RhynchonellidaeGray (in the TetrarhynchiinaeAger rather than in the CyclothyridinaeMakridin), on its evolution in the course of the Dogger period and the establishment of the biostratigraphy of its main species.  相似文献

The study of imprints of fossil plants collected in the Permian from the country of Hammanskraal (Namibia) leads to the following identifications: Glossopteris sp., Gangamopteris sp., Noegerrathiopsis sp., Gondwanidium sp., Asterotheca sp. and Lycopodiopsis derbyiB. Renault, Lycopodiopsis costilatus n. sp., Brasilodendron pedroanum(W. Carruthers) W.G. Chaloner & alii, Brasilodendron africanum n. sp. This flora has affinities with some fossil floras from South Africa and from Brazil, and is, probably, of upper Permian age.  相似文献

The presence of a ligula is showed off. Mechanisms controlling disintegration or breakdown of the germinal vesicle (GVBD) in Rana oocytes were investigated. A secondary cytoplasmic maturation promoting factor (MPF), produced in response to steroid stimulation, was shown to induce maturation when injected into immature recipient oocytes. Exposure of immature Rana oocytes to cycloheximide following injection of MPF or steroid treatment completely inhibited such maturation. Results indicate that injected MPF required protein synthesis for germinal vesicle breakdown and thus acted at some translational level. These results contrast with data obtained in Xenopus oocytes where injected MPF induced maturation in the presence of cycloheximide. Cytoplasmic MPF was also produced in Rana oocytes following treatment with lanthanum salts. This activity was similarly inhibited by cycloheximide. Time course studies conducted to compare the onset of cycloheximide insensitivity in steroid-treated and MPF-injected oocytes demonstrated that MPF-injected oocytes become insensitive to cycloheximide prior to steroid-treated germ cells. These results suggest that MPF acts as an intermediary in progesterone-induced maturation. Insensitivity to cycloheximide occurred several hours prior to the onset of germinal vesicle breakdown in both MPF-injected and steroid-treated oocytes. The data indicate that injected MPF in Rana does not induce nuclear disintegration directly, but rather requires amplification and/or autocatalytic synthesis of additional MPF or other factors for maturation to be induced. Molecular mechanisms involved in nuclear disintegration are discussed in relation to these species differences.  相似文献

Reciprocal crosses were made between pairs of plants showing the "aberrant ratio" (AR) effect at the a locus, and simultaneously between these plants and an aa tester. The results suggested that the AR effect in the particular line that was used could be explained by inactivation of a gene other than A, but also required for aleurone color. The development of orientation selectivity in the primary visual cortex is described by first-order kinetics between three functional compartments chained in a catenary mode. A first model is presented, in which two unidirectional kinetics with constant exchange coefficients, symmetrical in their effects, function in an alternating mode depending on the presence or absence of visual experience. The failure of this model to simulate the modifications induced by a delayed visual experience, when the exchange coefficients are identified to fit normal and dark rearing, supports the hypothesis that the maturation process consequent to interaction with visual environment is dependent on the date at which it is allowed to take place. A second model is then proposed, in which exchange coefficients are piecewise linear functions of time. In order to correctly predict the functional effects of restricted visual experience following prior dark rearing, it is assumed that visuomotor experience during the critical period permits the expression of a non-linear modifiability gradient which may have been masked up to this point by the absence of vision or eye movements.  相似文献
