We investigated growth interactions between the dinophyte Prorocentrum minimum and the bacillariophyte Skeletonema costatum using bi-algal cultures under axenic conditions. When low cell densities of P. minimum and high cell densities of S. costatum were inoculated into the same medium, growth of P. minimum was suppressed. Other inoculum combinations resulted in reduced S. costatum maximum cell densities. A mathematical model was used to simulate growth and interactions of P. minimum and S. costatum in bi-algal cultures. The model indicated that P. minimum always outcompeted S. costatum over time. Enriched filtrate from low-density P. minimum cultures significantly stimulated S. costatum growth, but enriched filtrate from high-density P. minimum cultures notably inhibited the growth of S. costatum. Growth of P. minimum was not affected by enriched filtrate from cultures of P. minimum at any density. Filtrates of P. minimum cultures were fractionated by ultrafiltration (molecular weight cutoff >3000 Da), and retentate that included polysaccharide(s) significantly inhibited the growth of S. costatum. 相似文献

During global deprotection using a strong acid in Boc chemistry, the electrophilic alkylating species, i.e. carbocations,
and formaldehyde generated from the side-chain protecting groups and the benzyloxymethyl group on the His residue, respectively,
can cause alkylation of susceptible residues. To reduce these side reactions, a deprotection procedure using a strong acid
such as HF or trifuloromethanesulfonic acid must always be carried out in the presence of scavengers. We found that addition
of hydroxylamine derivatives could efficiently suppress the side reactions associated with formaldehyde and that addition
of 2-mercaptopyridine could not only specifically circumvent alkylation of the Met residue but also convert its sulfonium

The growth, lipid content, and fatty acid composition of Aurantiochytrium sp. strain mh0186 at different temperatures were investigated. Strain mh0186 grew well at 15–30°C, but weakly at 10°C. The
biomass at 15–30°C was significantly higher than at 10 and 35°C, and the total lipid at 15–35°C was significantly higher than
that at 10°C. The amount of DHA in the total fatty acid was highest at 10°C and decreased in response to temperature increase.
The content of DHA (mg/g-dry cell weight) at 15–30°C were significantly higher than those at 35°C and those at 15–25°C were
significantly higher than those at 10 and 35°C. The DHA yield at 15–35°C was significantly higher than those at 10 and 35°C.
Unsaturation of fatty acid was regulated by temperature and was enhanced in response to temperature decrease. The ratio of

Since Robert Koch and colleagues found agar to be an effective gelling agent over a century ago, the pure culture method using agar plates has long been a standard of microbiology. Agar is undoubtedly easy to handle and useful for culture of microorganisms, but recent discovery of the ubiquity of microorganisms that cannot be cultured on agar raises a question: is agar really the best agent? In this study, we investigated the effect of two gelling agents, agar and gellan gum, on colony formation of a diverse array of microorganisms (total 108 strains) newly isolated from freshwater sediments and a representative microorganism as a slow grower on agar medium, Gemmatimonas aurantiaca, to clarify (i) whether they can grow on both agar and gellan gum plates, and (ii) the difference in time required for colony formation between the two gelling agents. Interestingly, 22 of 108 isolates showed no ability to form any visible colonies on the agar medium but did so on the gellan gum medium, and showed low 16S rRNA gene sequence similarities to their closest species. The remaining 86 isolates grew on both agar and gellan gum, but 52 of them grew much faster on gellan gum than on agar. Moreover, gellan gum also significantly stimulated the colony formation of the representative slow‐growing microorganism G. aurantiaca. Our results demonstrate that the gelling agent is a crucial factor for the growth of bacteria on plate media, and that alternatives to agar will be very important for increasing the culturability of yet‐to‐be cultured microorganisms. 相似文献

The effects of population density on male and female reproductive success of Betula maximowicziana were evaluated in two mixed and two post-fire stands, with various population densities, ranging from 1.9 to 300.0 trees
per ha, in central Hokkaido, Japan. First, we investigated ecological determinants of reproductive success (seed set and germination)
of both seeds collected from the trees (tree seeds) and dispersed seeds collected from seed traps (dispersed seeds). We then
evaluated the effects of population density on seed set and germination of tree seeds and dispersed seeds using a generalized
linear mixed model (GLMM). Subsequently, we genotyped 950 seeds collected from mother trees and 940 seeds trapped after dispersal
derived from tree seeds and dispersed seeds, respectively, using eleven microsatellite loci. Using the acquired data, we then
evaluated the outcrossing rate and effective number of pollen donors (N
ep) of the tree seeds, and the genetic structure of both pollen pools and dispersed seed populations. The seed set and germination
rate of dispersed seeds was significantly lower both in the lowest-density stand and in the highest-density stand. The GLMM
revealed that seed set and germination rates of dispersed seeds may be maximal at approximately 120 trees per hectare (optimal
density). Outcrossing rates were consistently high (t
m = 0.995), regardless of the population density. In contrast, N
ep was lower in the lowest-density stands. Significant genetic structure of the dispersed seed population was found in two low-density

Fourteen lactose-fermenting strains of Kluyveromyces marxianus , including its anamorph, Candida kefyr , were grown in two media containing 20% (w/v) sugar as either beet molasses or cheese whey. Strain NBRC 1963 of K. marxianus converted sucrose and lactose to ethanol in both media most efficiently. However, ethanol was produced from sucrose and not from lactose by strain NBRC 1963 in the medium containing equal amounts of sugar from beet molasses and cheese whey. The spontaneous mutants resistant to 2-deoxyglucose in the minimal medium composed of galactose as the sole carbon source were isolated from strain NBRC 1963. Among them, strain KD-15 vigorously produced ethanol in the media containing beet molasses, cheese whey, or both. The mutant strain KD-15 was insensitive to catabolite repression, as shown by the observation that β-galactosidase was not repressed in the presence of sucrose from beet molasses. 相似文献

Objective: In an attempt to clarify the conflicting data on resistin mRNA expression and protein analysis by western blotting in adipose tissue and serum, we developed a sensitive enzyme‐linked immunosorbent assay (ELISA) for direct measurement of mouse resistin. Research Methods and Procedures: We developed polyclonal antibodies directed to the N (21 to 40) and C (79 to 91) termini of mouse resistin. Then, affinity‐purified anti‐C‐terminal resistin immunoglobin G (IgG) was biotinylated. ELISA was based on the sandwiching of antigen between antibody IgG coated on polystyrene plates and biotinylated antibody IgG. The bound biotinylated antibody was quantified with streptavidin‐linked horseradish peroxidase. Results: New ELISA can measure a concentration as low as 0.5 ng/mL of recombinant mouse resistin and is sensitive and specific enough to measure resistin protein in various adipose tissues and in sera. In normal mice, decreases in resistin concentrations in both white adipose tissue and serum were age dependent during 6 to 24 weeks of development. Resistin concentrations were significantly higher in omental adipose tissue in comparison with perirenal and abdominal adipose tissues and were 2‐ to 5‐fold higher in females than males during the growth period. ob/ob mice had significantly lower resistin concentrations than the control mice in both sera and the white adipose tissues, particularly in the omental fat. The treatment by testosterone, but not progesterone or β‐estradiol, in cultured adipocytes reduces resistin protein levels in a dose‐dependent manner. Discussion: New sensitive ELISA for mouse resistin clarified that the resistin concentrations in normal mice were markedly elevated in the omental adipose depots as compared with the perirenal and abdominal adipocyte depots and significantly elevated compared with adipose tissues in genetically obese mice. 相似文献

Src‐like adaptor protein (SLAP) is a hematopoietic adaptor containing Src homology (SH)3 and SH2 motifs and a unique carboxy terminus. Unlike c‐Src, SLAP lacks a tyrosine kinase domain. We investigated the role of SLAP in osteoclast development and resorptive function. Employing SLAP‐deficient mice, we find lack of the adaptor enhances in vitro proliferation of osteoclast precursors in the form of bone marrow macrophages (BMMs), without altering their survival. Furthermore, osteoclastogenic markers appear more rapidly in SLAP?/? BMMs exposed to RANK ligand (RANKL). The accelerated proliferation of M‐CSF‐treated, SLAP‐deficient precursors is associated with enhanced ERK activation. SLAP's role as a mediator of M‐CSF signaling, in osteoclastic cells, is buttressed by complexing of the adaptor protein and c‐Fms in lipid rafts. Unlike c‐Src, SLAP does not impact resorptive function of mature osteoclasts but induces their early apoptosis. Thus, SLAP negatively regulates differentiation of osteoclasts and proliferation of their precursors. Conversely, SLAP decreases osteoclast death by inhibiting activation of caspase 3. These counterbalancing events yield indistinguishable bones of WT and SLAP?/? mice which contain equal numbers of osteoclasts in basal and stimulated conditions. J. Cell. Introduction of genetic material into cells is an essential prerequisite for current research in molecular cell biology. Although
transfection with commercially available reagents results in excellent gene expression, their high costs are obstacles to
experimentation with a large number or large scales of transfection. The cationic polymer linear-polyethylenimine (MW 25,000)
(PEI), one of the most cost-effective vehicles, facilitates DNA compaction by polyplex formation, which leads to efficient
delivery of DNA into cells by endocytosis. However, the use of PEI is still limited because of substantial cytotoxicity and
intolerable deterioration in transfection efficiency by its low stability. Here, we show that acidification of PEI is important
for its transfection activity. Dissolving PEI powder in 0.2N HCl confers a long shelf-life for PEI storage at 4 and −80 °C,
and the polyplex formation of plasmid DNA with PEI is optimized in lactate-buffered saline at pH 4.0. Furthermore, changing
the culture medium at 8–12 h posttransfection can minimize the cytotoxicity of PEI without sacrificing the high transfection
efficiency comparable to that of commercial reagents. The cost per test using acidified PEI is drastically reduced to approximately
1:10,000, compared with commercial reagents. Thus, we conclude that acidification of PEI satisfactorily accomplishes cost-effective,

Betula maximowicziana is an ecologically and economically important tree species in Japan. In order to examine the phylogeographical pattern of
the species in detail, maternally inherited chloroplast (cp) DNA variations of 25 natural populations of Betula maximowicziana and a total of 12 populations of three related species were evaluated by PCR-RFLP analysis. Two main haplotypic groups of
B. maximowicziana populations (northern and southern) were detected, with the main boundary passing through the Tohoku region in northeastern
Japan; in addition there was high genetic differentiation among the 25 populations studied (G
ST = 0.950,
G\textST¢ = 0. 9 7 7 G_{\text{ST}}^{\prime } = 0. 9 7 7 ). The phylogeographical pattern exhibited by B. maximowicziana was much more similar to that of alpine plants than to that of beech and oak. Comparison of the patterns of genetic structure
obtained from the cpDNA with previously and newly acquired data on bi-parentally inherited nuclear DNA indicates that the
nuclear genome was transferred via pollen from the northern haplotypic group to the southern group more frequently than it
moved in the opposite direction. Although common haplotypes were detected among B. maximowicziana and the two related species examined, these haplotypes were not shared sympatrically, suggesting very rare hybridization
among the species currently occurring in their natural populations. 相似文献
