Thermal response in murine L929 cells lacking αB-crystallin expression and αB-crystallin expressing L929 transfectants

We investigated the role of B-crystallin expression in the development of thermotolerance in murine L929 cells. An initial heat-shock of 10 min at 45°C induced thermotolerance in these cells to a heat challenge at 45°C administered 24 h later. The thermotolerance ratio at 10^–1 isosurvival was 1.7. Expression of B-crystallin gene was not detected during the 24 h incubation at 37°C following heat shock by either northern or western blots. In contrast, inducible HSP70 synthesis was observed during this time period. Thus, this cell line provided an unique system in which to examine the effects of transfected B-crystallin on thermoresistance and thermotolerance. Cells stably transfected with B-crystallin under the control of an inducible promoter did not show a significant increase in the ability to develop thermotolerance. However, a stably transfected L929 clone expressing high levels of constitutive B-crystallin exhibited an approximately 50% increase in thermal resistance over parental and control cells. Though expression of B-crystallin is not requisite for the development of thermotolerance in L929 cells, overexpression of transfected B-crystallin can contribute to increased thermoresistance.     

Simultaneous Determination of Dermatan Sulfate and Oversulfated Dermatan Sulfate in Plasma by High-Performance Liquid Chromatography with Postcolumn Fluorescence Derivatization

A chemical method for the determination of dermatan sulfate (DS) and oversulfated dermatan sulfate has been developed and applied to the pharmacokinetic study of these polysaccharides in experimental animals. The analytical procedure includes a simple preparation step of administered DS and oversulfated DS from blood plasma, HPLC for the separation and detection of DS and oversulfated DS using an Asahipak NH₂P-50 column, fluorometric reaction of the polysaccharides with guanidine in a strong alkaline medium. DS and oversulfated DS were extracted from plasma by treating it with proteinase to remove plasma proteins and recovered with endogenous plasma glycosaminoglycans by ethanol precipitation. Finally, DS and oversulfated DS were analyzed by fluorometric HPLC. The detection limits of DS and oversulfated DS were 10 and 20 ng, respectively. Furthermore, we demonstrated that artificial oversulfation of DS increased its biological half-life after intravenous administration to rats.

我国仓贮昆虫病原芽孢杆菌的研究

对９５１个样品分离鉴定,有７４７个样品含芽孢杆菌,有菌率为７８．５５％．共分离得到芽孢杆菌１１３８株,其中苏云金杆菌（Ｂａｃｉｌｌｕｓｔｈｕｒｉｎｇｉｅｎｓｉｓ,简称Ｂ．ｔ）１４３株,占１２．５％;球形芽孢杆菌（Ｂａｃｉｌｌｕｓｓｐｈａｅｒｉｃｕｓ,简称Ｂ．ｓ）１１株,占０．９７％;其他芽孢杆菌９８４株,占８６．４０％．从芽孢杆菌中选出产生晶体、苏云金素或磷酸酯酶Ｃ（ＰｈｏｓｐｈａｌｉｐａｓｅＣ,简称ＰＬＣ）的毒素菌株１６８株,其中Ｂ．ｔ占１４３株,Ｂ．ｓ有５株,其他芽孢杆菌１０株．在产毒素菌株中,经测定有１２０株菌对供试昆虫毒性达标．占７７．９２％．不同菌株的杀虫毒素、杀虫范围和毒力各异,认为这种差异取决于毒素和虫种两方面的特异性．     

粤西及相邻地区太阳总辐射、光合有效辐射能量和光量子通量时空分布

应用普适全国的计算太阳辐射、光合有效辐射和光量子通量模型,系统地研究了粤西的高要、封开和临近地区梧州的太阳辐射、光合有效辐射和光量子通量的年总量、月总量以及相应的年平均日总量和日平均日总量。结果表明,太阳辐射、光合有效辐射和光量子通量的年变化有相似的规律;而地区变化有以下特点：梧州和封开明显类似,而高要与上两地差异稍大。     

转译优化对EPO基因在COS7细胞中表达的影响

利用ＣＯＳ７细胞暂时表达系统,研究转译起始序列对ＥＰＯ－ｃＤＮＡ表达的影响。通过ＤＮＡ重组技术,构建了原ＥＰＯ－ｃＤＮＡ表达载体ｐＣＳＶ－ＥＰＯ（１）,其转译起始序列为５＇ＡＡＴＴＣＡＴＧＧ３＇。同时通过定点突变技术,将起始序列改变成５＇ＣＣＡＣＣＡＴＧＧ３＇,而构建了另一表达载体ＰＣＳＶ－ＥＰＯ（２）。后经序列分析证明无误后和前均通过ＤＥＡＥ－ｄｅｘｔｒａｎ法转染ＣＯＳ７细胞上清,测定结果为     

棉铃虫对氰戊菊酯抗性和敏感品系的选育

用氰戊菊酯对来自阳谷的棉铃虫(YG)Heliothis armigera(Hubncr) 进行抗性晶系的筛选。在15代期间经过9代的室内选育,获得抗性品系(Fcn-R),抗性倍数高达2“3倍,筛选后F₁₅代LD₅₀值(24.1412μg/头)比筛选前F1代lD₅₀值(0.2020μg/头)提高了119.5倍。对来自偃师的棉铃虫(YS)进行了连续两代单对筛选,得到敏感品系(Fen-S),敏感晶系的LD50值为0.0116μg/头,接近1983年东台敏感晶系的LD₅₀值(0.0098μg/头)Fcn-R抗性晶系筛选前后分别测定了七种杀虫剂的剂量-死亡回归线,发现Fen-R抗性品系对溴氰菊酯[LD₅₀(Fen-R)/LD₅₀(YG)=5.2X] 和氯氰菊酯(2.5X)具有一定程度的交互抗性;而对功夫菊酯(0.66X),氯菊酯(0.89x)、灭多威(0.74X)及久效磷(1.5x)没有交互抗性。氰戊菊酯加Pb的增效试验结果表明棉铃虫对氰戊菊酯的抗性主要是由于多功能氧化酶的代谢作用。毒理学资料还暗示抗性为多因子(基因)的。     

Application of fuzzy reasoning to control of glucose and ethanol concentrations in baker's yeast culture

Fuzzy reasoning was applied to control both ethanol and glucose concentrations in fed-batch cultures of baker's yeast. This fuzzy controller consisted of three membership functions (concentrations of dissolved oxygen (DO), ethanol and glucose) and 18 production rules. Fuzzy inference was carried out by IF {A is a and B is b,...#x007D;, THEN {C is c} from the on-line measured concentrations of DO, ethanol and glucose. When medium concentrations of ethanol and glucose in fed-batch culture of baker's yeast were set at 2 g/l and 0.2 g/l, both ethanol and glucose concentrations were controlled at 2.67±0.35 g/l and 0.27±0.25 g/l, respectively, ethanol production was reduced from 26 g/l to 34 g/l, cell yield increased from 0.38 to 0.53 g dry cell/g consumed glucose and ethanol yield decreased from 0.50 to 0.14 g ethanol/g consumed glucose, respectively, as compared with those of the glucose only control at 0.2 g/l.
