Reliable Method for Fabricating Tissue‐Mimicking Materials With Designated Relative Permittivity and Conductivity at 128 MHz

Artificial materials that can simultaneously mimic the relative permittivity and conductivity of various human tissues are usually used in medical applications. However, the method of precisely designing these materials with designated values of both relative permittivity and conductivity at 3 T MRI resonance frequency is lacking. In this study, a reliable method is established to determine the compositions of artificial dielectric materials with designated relative permittivity and conductivity at 128 MHz. Sixty dielectric materials were produced using oil, sodium chloride, gelatin, and deionized water as the main raw materials. The dielectric properties of these dielectric materials were measured using the open‐ended coaxial line method at 128 MHz. Nonlinear least‐squares Marquardt–Levenberg algorithm was used to obtain the formula, establishing the relationship between the compositions of the dielectric materials and their dielectric properties at 128 MHz. The dielectric properties of the blood, gall bladder, muscle, skin, lung, and bone at 128 MHz were selected to verify the reliability of the obtained formula. For the obtained formula, the coefficient of determination and the expanded uncertainties with a coverage factor of k = 2 were 0.991% and 4.9% for relative permittivity and 0.992% and 6.4% for conductivity. For the obtained artificial materials measured using the open‐ended coaxial line method, the maximal difference of relative permittivity and conductivity were 1.0 and 0.02 S/m, respectively, with respect to the designated values. In conclusion, the compositions of tissue‐mimicking material can be quickly determined after the establishment of the formulas with the expanded uncertainties of less than 10%. 途径特异性调控因子介导的链霉菌来源天然产物的产量提升

链霉菌是活性天然产物的重要来源。基因组学研究揭示了链霉菌有巨大的生物合成潜力,平均每株菌拥有20–40个生物合成基因簇。在常规的实验室条件下,大多数链霉菌来源的天然产物产量较低,影响了对其进一步研究和产业化开发。由于链霉菌中天然产物的生物合成受到严格的调控,对于这些调控因子和调控网络的深入研究将有力地促进链霉菌来源的天然产物的发现和开发利用。文中主要综述了近5年来链霉菌来源天然产物生物合成中途径特异性调控因子的研究进展,重点介绍其在提高相应天然产物产量中的应用。

Genetic Analysis of Human Adenovirus Type 7 Strains Circulating in Different Parts of China

To investigate the molecular epidemiology and genetic variation of human adenovirus type 7(HAdV-7) in children with acute respiratory infections(ARI) in China. HAdV-7-positive respiratory samples collected from children with ARI in Beijing, Shijiazhuang, Wenzhou and Guangzhou from 2014–2018 were selected for gene amplification and sequence analysis. Fifty-seven HAdV-7 clinical strains with hexon, penton base and fiber gene sequences were obtained. Meanwhile17 strains were selected randomly from different cities for whole genome sequencing. Phylogenetic and variation analyses were performed based on the obtained sequences, HAdV-7 prototype strain Gomen(AY594255), vaccine strains(AY495969 and AY594256) and representative sequences of strains. The phylogenetic trees constructed based on whole genome sequences, major capsid protein genes(hexon, penton base and fiber) and the early genes(E1, E2, E3 and E4) were not completely consistent. The HAdV-7 strains obtained in this study always clustered with most of the circulating strains worldwide from the 1980 s to the present. Compared with the HAdV-7 prototype strain Gomen(AY594255), some amino acid mutations in loop1 and loop2 of hexon and the RGD loop region of the penton base gene were observed. Recombination analysis showed that partial regions of 55 k Da protein and 100 kDa hexon-assembly associated protein genes among all HAdV-7 strains in this study were from HAdV-16 and HAdV-3, respectively. Our study demonstrated the molecular evolution characteristics of HAdV-7 strains circulating in China and provided basic reference data for the prevention, control and vaccine development of HAdV-7.     

Genome‐wide distribution and functions of the AAE complex in epigenetic regulation in Arabidopsis

Heterochromatin is widespread in eukaryotic genomes and has diverse impacts depending on its genomic context. Previous studies have shown that a protein complex, the ASI1‐AIPP1‐EDM2 (AAE) complex, participates in polyadenylation regulation of several intronic heterochromatin‐containing genes. However, the genome‐wide functions of AAE are still unknown. Here, we show that the ASI1 and EDM2 mostly target the common genomic regions on a genome‐wide level and preferentially interacts with genetic heterochromatin. Polyadenylation (poly(A) sequencing reveals that AAE complex has a substantial influence on poly(A) site usage of heterochromatin‐containing genes, including not only intronic heterochromatin‐containing genes but also the genes showing overlap with heterochromatin. Intriguingly, AAE is also involved in the alternative splicing regulation of a number of heterochromatin‐overlapping genes, such as the disease resistance gene RPP4. We provided evidence that genic heterochromatin is indispensable for the recruitment of AAE in polyadenylation and splicing regulation. In addition to conferring RNA processing regulation at genic heterochromatin‐containing genes, AAE also targets some transposable elements (TEs) outside of genes (including TEs sandwiched by genes and island TEs) for epigenetic silencing. Our results reveal new functions of AAE in RNA processing and epigenetic silencing, and thus represent important advances in epigenetic regulation.     

罗氏沼虾MrLc3a基因cDNA的克隆及在副溶血弧菌胁迫下的表达分析

虾类和果蝇同属节肢动物.果蝇的相关研究表明自噬与免疫关系密切,而虾类自噬机制研究鲜少.微管相关蛋白1轻链3 (microtubule-associated protein 1 light chain 3,Lc3)与自噬基因Atg8同源,其与自噬体的形成密切相关,是自噬活性的标志分子.本研究利用RACE技术克隆了罗氏沼虾的MrLc3a基因的全长cDNA,用RT-qPCR检测了该基因在罗氏沼虾主要组织中的表达量;并研究了正常和副溶血弧菌感染两种情况下MrLc3a基因和免疫基因Relish的表达变化情况,为其在病害防御方面的应用提供了前期数据.试验结果表明:MrLc3a基因全长653 bp,其中包括195 bp的5'-UTR、378 bp的ORF开放阅读框和80 bp的3'-UTR,共编码126个氨基酸;序列比对结果显示,其编码的氨基酸序列和南美白对虾Lc3a编码的氨基酸序列具有较高的同源性,并在系统发育树上聚为一支;RT-qPCR结果显示,MrLc3a基因在罗氏沼虾各个组织均有表达,其中在脑、鳃、胃中的表达量较高,在肝胰腺和性腺中的表达量较少;副溶血弧菌感染罗氏沼虾后显著影响了MrLc3a和Relish基因在罗氏沼虾肝胰腺组织中的转录情况,MrLc3a和Relish基因随时间变化都呈现出先上升后下降的趋势,表明MrLc3a基因通过参与细胞自噬过程而参与了免疫反应.
