Endocytosis of the nicotinic acetylcholine receptor (AChR) is a proposed major mechanism of neuromodulation at neuromuscular junctions and in the pathology of synapses in the central nervous system. We show that binding of the competitive antagonist alpha-bungarotoxin (alphaBTX) or antibody-mediated cross-linking induces the internalization of cell surface AChR to late endosomes when expressed heterologously in Chinese hamster ovary cells or endogenously in C2C12 myocytes. Internalization occurs via sequestration of AChR-alphaBTX complexes in narrow, tubular, surface-connected compartments, which are indicated by differential surface accessibility of fluorescently tagged alphaBTX-AChR complexes to small and large molecules and real-time total internal reflection fluorescence imaging. Internalization occurs in the absence of clathrin, caveolin, or dynamin but requires actin polymerization. alphaBTX binding triggers c-Src phosphorylation and subsequently activates the Rho guanosine triphosphatase Rac1. Consequently, inhibition of c-Src kinase activity, Rac1 activity, or actin polymerization inhibits internalization via this unusual endocytic mechanism. This pathway may regulate AChR levels at ligand-gated synapses and in pathological conditions such as the autoimmune disease myasthenia gravis.  相似文献

We investigated the possibility of decreased feeding deterrent response following prolonged exposure in three specialist herbivores, Plutella xylostella (larvae), Pseudaletia unipuncta (larvae), and Epilachna varivestis (adults) through leaf disc choice and no‐choice tests. Neonate larvae (<24 h old) of the three species were reared on their host plants sprayed with the antifeedants until tested. Our results demonstrated that Ps. unipuncta did not show a decrease in feeding deterrent response to extracts of Melia volkensii (choice and no‐choice tests) or oregano (Origanum vulgare) (choice test) following prolonged exposure. Plutella xylostella exhibited a decrease in feeding deterrent response to M. volkensii in a choice test only, but not to oregano. Although not significant, we did find a trend toward decreasing feeding deterrent response to M. volkensii by both species in no‐choice tests. However, both species exhibited a significant decrease in feeding deterrent response to pure allelochemicals (Ps. unipuncta to thymol, and P. xylostella to thymol and toosendanin) compared with the naïve groups, following prolonged exposure in leaf disc choice tests. Epilachna varivestis showed a significant decrease in feeding deterrent responses to both oregano and thymol in leaf disc choice tests. We conclude from our present and previous studies that not only are there interspecific differences between generalist and specialist species but also among specialist species.  相似文献

Increasing anthropogenic pCO2 alters seawater chemistry, with potentially severe consequences for coral reef growth and health. Octocorals are the second most important faunistic component in many reefs, often occupying 50% or more of the available substrate. Three species of octocorals from two families were studied in Eilat (Gulf of Aqaba), comprising the zooxanthellate Ovabunda macrospiculata and Heteroxenia fuscescens (family Xeniidae), and Sarcophyton sp. (family Alcyoniidae). They were maintained under normal (8.2) and reduced (7.6 and 7.3) pH conditions for up to 5 months. Their biolological features, including protein concentration, polyp weight, density of zooxanthellae, and their chlorophyll concentration per cell, as well as polyp pulsation rate, were examined under conditions more acidic than normal, in order to test the hypothesis that rising pCO2 would affect octocorals. The results indicate no statistically significant difference between the octocorals exposed to reduced pH values compared to the control. It is therefore suggested that the octocorals' tissue may act as a protective barrier against adverse pH conditions, thus maintaining them unharmed at high levels of pCO2.  相似文献

Summary The mechanism of higher grain production of wheat (Triticum aestivum L.) by four tillage methods was explored on a sandy calcareous soil of Sahl-Al Jafara, Libya, during the year 1976–77. Tillage methods increased grain yield and dry matter weight at the boot stage in the order of no-tillage<rotovator=disc plough < subsoiler. Rotovation to 15 cm and disc ploughing to 25 cm depth enhanced grain production mainly by eliminating weed competition. In both the cases, roots penetrated < 25 cm where plough-pan existed.Further yield increases by 50 cm deep subsoiling seems to be mainly explained by N and Cu rise in plants from their marginal to optimum levels and by reduction of Mn deficiency in plant shoots. Penetration of roots beyond the 25 cm plough-pan apparently resulted in higher absorption of these nutrients from leached or native soil supplies. Subsoiling also resulted in greater Zn concentration in plant shoots which, under marginal to deficient conditions, will also increase grain production.  相似文献
