MRE11-Deficiency Associated with Improved Long-Term Disease Free Survival and Overall Survival in a Subset of Stage III Colon Cancer Patients in Randomized CALGB 89803 Trial

Purpose

Colon cancers deficient in mismatch repair (MMR) may exhibit diminished expression of the DNA repair gene, MRE11, as a consequence of contraction of a T₁₁ mononucleotide tract. This study investigated MRE11 status and its association with prognosis, survival and drug response in patients with stage III colon cancer.

Patients and Methods

Cancer and Leukemia Group B 89803 (Alliance) randomly assigned 1,264 patients with stage III colon cancer to postoperative weekly adjuvant bolus 5-fluorouracil/leucovorin (FU/LV) or irinotecan+FU/LV (IFL), with 8 year follow-up. Tumors from these patients were analyzed to determine stability of a T₁₁ tract in the MRE11 gene. The primary endpoint was overall survival (OS), and a secondary endpoint was disease-free survival (DFS). Non-proportional hazards were addressed using time-dependent covariates in Cox analyses.

Results

Of 625 tumor cases examined, 70 (11.2%) exhibited contraction at the T₁₁ tract in one or both MRE11 alleles and were thus predicted to be deficient in MRE11 (dMRE11). In pooled treatment analyses, dMRE11 patients showed initially reduced DFS and OS but improved long-term DFS and OS compared with patients with an intact MRE11 T₁₁ tract. In the subgroup of dMRE11 patients treated with IFL, an unexplained early increase in mortality but better long-term DFS than IFL-treated pMRE11 patients was observed.

Conclusions

Analysis of this relatively small number of patients and events showed that the dMRE11 marker predicts better prognosis independent of treatment in the long-term. In subgroup analyses, dMRE11 patients treated with irinotecan exhibited unexplained short-term mortality. MRE11 status is readily assayed and may therefore prove to be a useful prognostic marker, provided that the results reported here for a relatively small number of patients can be generalized in independent analyses of larger numbers of samples.

Coccidia in Camels (Camelus dromedarius) in Saudi Arabia1

Saudi Arabian camels {Camelus dromedarius) are infected with three species of Eimeria: E. dromedarii (28.4%), E. rajasthani < 22.2%), and E. cameli (19.2%); 41.6% of the animals examined were positive. The highest prevalence of infection was reported in the western region of the country. Mixed infection with two Eimeria species is most common; E. dromedarii was most frequently and generally the most predominant species. Eimeria dromedarii and E. rajasthani are described for the first time from Saudi Arabian camels.     

Deep Subwavelength Confinement of Mid-infrared Plasmon Modes by Coupling Graphene-Coated Nanowire with a Dielectric Substrate

In this paper, we report the realization of highly confined mid-infrared plasmon modes by coupling freestanding graphene-coated nanowire with a dielectric substrate. It is shown that the presence of a nearby dielectric substrate not only breaks the azimuthal symmetry of the plasmon modes but also has a strong impact on the coupling and hybridization of these modes. The degree of interactions with the substrate depends on the permittivity of the substrate, the key structural parameters of the nanowire, the operation frequency and chemical potential of graphene, as well as the gap distance between the nanowire and supporting substrate. It is found that compared to freestanding case, using a high-index substrate and adjusting the gap distance can result in the following benefits: (i) an ultra-small mode area and a long propagation length can be realized simultaneously, (ii) a stronger field enhancement in the low-index gap region and improved figure of merit can be achieved, and (iii) a huge reduction in the crosstalk can be made which is crucial for the realization of high-density integrated nanophotonic devices.     

Ectomycorrhizal fungi increase the vitality of Norway spruce seedlings under the pressure of Heterobasidion root rot in vitro but may increase susceptibility to foliar necrotrophs

We tested if root colonisation by ectomycorrhizal fungi (EMF) could alter the susceptibility of Norway spruce (Picea abies) seedlings to root rot infection or necrotic foliar pathogens. Firstly, spruce seedlings were inoculated by various EMF and challenged with Heterobasidion isolates in triaxenix tubes. The ascomycete EMF Meliniomyces bicolor, that had showed strong antagonistic properties towards root rot causing Heterobasidion in vitro, protected spruce seedlings effectively against root rot. Secondly, spruce seedlings, inoculated with M. bicolor or the forest humus, were subjected to necrotrophic foliar pathogens in conventional forest nursery conditions on peat substrates. Botrytis cinerea infection after winter was mild and the level of needle damage was independent of substrate and EMF colonisation. Needle damage severity caused by Gremminiella abietina was high in seedlings grown in substrates with high nutrient availability as well as in seedlings with well-established EMF communities. These results show that albeit M. bicolor is able to protect spruce seedlings against Heterobasidion root rot in axenic cultures it fails to induce systemic protection against foliar pathogens. We also point out that unsterile inoculum sources, such as the forest humus, should not be considered for use in greenhouse conditions as they might predispose seedlings to unintended needle damages.

Effects of cerivastatin on adrenergic pathways, hypertrophic growth and TGFbeta expression in adult ventricular cardiomyocytes

The effects of statin treatment in the setting of heart failure have already been shown. Nevertheless, there is little knowledge about its influence on adrenergic pathways in cardiomyocytes. Therefore, this study investigated the impact of cerivastatin on adrenoceptor-mediated signalling pathways in isolated adult ventricular cardiomyocytes. It focused on two endpoints: hypertrophic growth and TGFbeta expression. Cultured cardiomyocytes were used to study rac activation (analysed by its translocation into the membrane fraction), ROS formation (H(2)DCF fluorescence) and hypertrophic growth ((14)C-phenylalanine incorporation). Alpha- and beta-adrenoceptor stimulation showed significant differences regarding rac activation, ROS formation, and p38 MAP kinase activation. Both alpha- and beta-adrenoceptor stimulation induced TGFbeta expression. Upon activation of alpha-adrenergic signalling - although ROS formation was not influenced by cerivastatin - TGFbeta expression decreased. Following beta stimulation, TGFbeta expression as well as rac and p38 MAP kinase activation were reduced after pre-treatment with cerivastatin. Statin treatment did not show any influence on hypertrophic growth. In summary, this study clearly demonstrates the ability of adrenoceptor stimulation to increase TGFbeta expression. One component of the beneficial effects of statin therapy on heart failure might therefore be due to a dominant reduction and inhibition of TGFbeta, which is involved in many pathophysiological processes in cardiomyocytes.     

Evaluation of oil recovery by rhamnolipid produced with isolated strain from Iranian oil wells

A rhamnolipid producing bacterium,Pseudomonas aeruginosa HR was isolated from south of Iranian oil wells. In the previous study, effects of main factors of cultivation on production of biosurfactant by this strain were evaluated and their optimized values were determined. In the present paper, the ability of produced biosurfactant in oil recovery was investigated. For this purpose, its affect on enhanced oil recovery in sand pack and core holder obtained from oil wells was evaluated. It was found that the maximum oil recovery in saturated sand pack column and in saturated core holder of crude oil were 23.6 and 64%, respectively. Then, in core test mathematical model was proposed by considering interactions between given parameters. Furthermore, dependence of residual oil recovery on the process of core washing during injection of biosurfactant was investigated. Finally, capillary desaturation curve was compared with core flood data.
