The current laser atherectomy technologies to treat patients with challenging (to-cross) total chronic occlusions with a step-by-step (SBS) approach (without leading guide wire), are lacking real-time signal monitoring of the ablated tissues, and carry the risk for vessel perforation. We present first time post-classification of ablated tissues using acoustic signals recorded by a microphone placed nearby during five atherectomy procedures using 355 nm solid-state Auryon laser device performed with an SBS approach, some with highly severe calcification. Using our machine-learning algorithm, the classification results of these ablation signals recordings from five patients showed 93.7% classification accuracy with arterial vs nonarterial wall material. While still very preliminary and requiring a larger study and thereafter as commercial device, the results of these first acoustic post-classification in SBS cases are very promising. This study implies, as a general statement, that online recording of the acoustic signals using a noncontact microphone, may potentially serve for an online classification of the ablated tissue in SBS cases. This technology could be used to confirm correct positioning in the vasculature, and by this, to potentially further reduce the risk of perforation using 355 nm laser atherectomy in such procedures. 相似文献

Galactosyltransferase specific activities in embryonic chick retina, optic tectum, and telecephalon were found to decline during embryonic development. Incorporation of galactose from nucleotide sugar into exogenously added glycoprotein acceptor was measured in the presence of excess of glycoprotein acceptor. This ensured that the specific activity measurements were reflections of true enzyme specific activity rather than availability of acceptor. Moreover, we have shown that the decline in specific activity is not due to degradation of the nucleotide sugar, UDP-galactose, under our in vitro assay conditions.Enzymatic specific activity declined sharply with embryonic age for all tissues tested. This decline was not affected by the presence of 5-bromodeoxyuridine during in vitro culture of embryonic chick neutral retina above that caused by the culturing alone. Galactosyltransferase activity was not found to be associated with the plasma membrane fraction from homogenized tissue but rather with the microsomal fraction. Thus, the changes in galactosyltransferase specific activity detected here do not reflect changes at the cell surface. 相似文献

Enteropathogenic (EPEC) and enterohaemorrhagic Escherichia coli (EHEC) constitute a significant risk to human health worldwide. Both pathogens colonize the intestinal mucosa and, by subverting intestinal epithelial cell function, produce a characteristic histopathological feature known as the 'attaching and effacing' (A/E) lesion. Although EPEC was the first E. coli to be associated with human disease in the 1940s and 1950s, it was not until the late 1980s and early 1990s that the mechanisms and bacterial gene products used to induce this complex brush border membrane lesion and diarrhoeal disease started to be unravelled. During the past few months, there has been a burst of new data that have revolutionized some basic concepts of the molecular basis of bacterial pathogenesis in general and EPEC pathogenesis in particular. Major breakthroughs and developments in the genetic basis of A/E lesion formation, signal transduction, protein translocation, host cell receptors and intestinal colonization are highlighted in this review. 相似文献

Phylogenetic relationships within Pectinidae (Bivalvia, Pteriomorphia) have been investigated primarily for Pacific and Western Atlantic or commercially valuable taxa. Most molecular phylogenetic studies have revealed monophyly of pectinid bivalves but interrelationships of the different clades are still inconsistent. However, non-commercial European Pectinidae has mostly been neglected in earlier investigations and therefore the evolution and radiation of the European Pectinidae is poorly understood. Since the fossil record of this group is well investigated, the evolutionary age of phylogenetic diversification and radiation events within this group can be dated. Thus, the connection of geological and climatic changes to radiation events within this group can be assessed. We investigated the phylogenetic relationships within European Pectinidae using mitochondrial (12S and 16S) and nuclear (18S, 28S and H3) gene markers and performed relaxed molecular clock approaches to gain information on the evolutionary age and the connection between Cenozoic climatic changes and diversification within this group. The results show concordance of radiation events with the Middle Miocene cooling event and the following climatic period with slowly decreasing temperatures. However, geological changes such as the uplift of the Gomphotherium Landbridge or the closure and re-opening of the Strait of Gibraltar also had great impact on diversification and distribution patterns within European Pectinidae. 相似文献

Heparin uptake and desulphation by cultured macrophages were investigated. Histones, polyamino-acids, protamine and eosinophil-basic protein stimulated both heparin uptake and desulphation, processes found to be non-related. Poly-l-ornithine and poly-dl-lysine increased the heparin uptake by about 33-fold, and histone produced up to 7.5-fold increase in the desulphation.
