Huge tsunami waves associated with megathrust earthquakes have a severe impact on shallow marine ecosystems. We investigated the impact of a tsunami generated by the 2011 M9.0 Tohoku-Oki earthquake on the seafloor and large benthic animals in muddy and sandy ria coasts (Otsuchi and Funakoshi bays) in northeastern Japan. We conducted underwater field surveys using scuba equipment in water depths of <20 m before the tsunami (September 2010) and after the tsunami (September 2011 and September 2012). During the study period, episodic changes in topography and grain-size composition occurred on the seafloor of the study area. Megabenthos sampling revealed a distinct pattern of distribution succession for each benthic species. For example, the protobranch bivalve Yoldia notabilis (Bivalvia: Nuculanidae) and the heterodont bivalve Felaniella usta (Bivalvia: Ungulinidae) disappeared after the tsunami event, whereas the distribution of the venus clam Gomphina melanaegis (Bivalvia: Veneridae) remained unchanged. In addition, the patterns of succession for a single species, such as the giant button top shell Umbonium costatum (Gastropoda: Trochidae) and the heart urchin Echinocardium cordatum (Echinoidea: Loveniidae), varied between the two bays studied. Our data also show that reestablishment of some benthic animal populations began within 18 months of the tsunami disturbance.  相似文献

The significance of a halocline for nutrient cycles in a shallow coastal lagoon, Lake Nakanoumi, was investigated. The study was made possible when the northern part of the lagoon was enclosed by polder dikes prior to land reclamation for agricultural purposes. The disappearance of the halocline allowed vertical mixing to extend to the whole depth and vertical gradients in water properties then decreased remarkably. Despite the disappearance of the halocline, dissolved oxygen content of bottom waters decreased considerably during summer months. Summer peaks in inorganic nitrogen and phosphate were still observed after disappearance of the halocline.  相似文献

Granulocyte colony-stimulating factor (G-CSF) has been used for the treatment of neutropenia in hematologic disorders. The neuroprotective effects of G-CSF were reported in neurological disease models. In the present study, we examined whether G-CSF can protect dopaminergic neurons against MPTP-induced cell death in a mouse model of Parkinson's disease. Mice of one group were injected intraperitoneally with MPTP for five consecutive days, those of another group with MPTP and intraperitoneal G-CSF at 2 days and 1 day before the first MPTP injection, and 30 min before each MPTP injection, while control mice received saline injections. Immunohistochemistry, western blotting analysis, and HPLC were performed to evaluate damage of substantia nigra dopaminergic neurons and expression of Bcl-2 and Bax protein. MPTP induced dopaminergic cell death in the substantia nigra. G-CSF significantly prevented MPTP-induced loss of tyrosine hydroxylase-positive neurons (p < 0.05), increased Bcl-2 protein and decreased Bax protein expression. Our findings indicate that G-CSF provides neuroprotection against MPTP-induced cell death and this effect is mediated by increasing Bcl-2 expression levels and decreasing Bax expression levels in C57BL/6 mice.  相似文献

Nitrous oxide in brackish Lakes Shinji and Nakaumi, Japan   总被引:1,自引:0,他引:1  
Nitrous oxide (N2O) was measured monthly from September 1997 to August 1998 in the brackish Lakes Shinji and Nakaumi, Japan. N2O (5–37 μg N l−1) was supersaturated in the overlying water on lake sediments from October 1997 to January 1998. The N2O concentration in the hypolimnion was higher than that in the epilimnion on 17 October 1997, when N2O was first observed in a water column of Lake Nakaumi. Afterward, N2O was almost uniform throughout the water column and then disappeared on 16 February 1998. On the one hand, large amounts of N2O were found throughout the year in the interstitial water in Lake Shinji, where a high concentration of nitrate was discharged from the Hii River. On the other hand, in Lake Nakaumi, stratified by halocline, a high concentration of N2O was observed in the interstitial water only from winter to spring. N2O concentrations in the interstitial water were about 10 to 1000 times as large as those in the overlying water. These results imply that N2O was mainly produced at the sediment-water interface and was diffused to the overlying water. It was also suggested that the accumulation of N2O in the sediment-water system was accelerated by a high concentration of hydrogen sulfide. In order to understand the role of nitrification and denitrification in the accumulation of nitrous oxide (N2O) in the hypolimnetic water of brackish Lake Nakaumi, the effects of dissolved oxygen (DO) concentration on these activities were investigated by incubation experiments. N2O was produced during the oxidation of NH4 + to NO2 in nitrification and during the reduction of NO3 to N2 in denitrification. N2O-producing activity by nitrification (N2ON) increased markedly with decreasing concentrations of DO. Low DO (10%–30% saturation) induced high N2ON. In contrast to nitrification, N2O-producing activity by denitrification (N2OD) decreased with decreasing concentrations of DO. Little N2O was accumulated during denitrification under low-level conditions of DO (10%–30%), because of further reduction of N2O to N2. It can therefore be assumed that N2O produced as the by-product of nitrification is concurrently reduced to N2 by denitrification under low-DO conditions. BACKGROUND/AIMS: To evaluate the safety and feasibility of immunotherapy based on autologous dendritic cells (DC) for patients with unresectable primary liver cancer (PLC). METHODS: A total of ten patients were enrolled and immunized with DCs. Autologous DCs were generated ex vivo in the presence of granulocyte-macrophage colony-stimulating factor (GM-CSF) and interleukin-4 (IL-4). Cells were then pulsed with tumor lysate (TL), tumor necrosis factor-alpha (TNF-alpha) and keyhole limpet hemocyanin (KLH). Non-adherent cells were collected on day 9 and cells were administered into the inguinal lymph node. Each patient received 1-10 x 10(6) cells four times at weekly intervals. RESULTS: Immunization was well tolerated in all patients without significant toxicity. DC vaccination induced delayed-type hypersensitivity (DTH) against KLH in seven out of ten patients. In one patient, one of the two liver tumors (tumor in segment 7, 13 mm in diameter) decreased in size to 7 mm and showed necrotic change on computed tomography examination after eight immunizations. In two patients, serum levels of tumor markers decreased after vaccination. CONCLUSION: The present clinical trial suggested that immunization by TL-pulsed DCs is feasible in patients with unresectable PLC without any toxicity. Further improvement in the clinical results of immunotherapy might be expected by modifying the therapeutic protocol.  相似文献

The Argentine ant, Linepithema humile, has invaded sites across Africa, Australia, Europe, and North America. In its introduced ranges it eliminates native ants and tends agricultural pests. Few studies have examined the ecology of Argentine ants in their native habitat. This study examined the effects of parasitoid flies, genus Pseudacteon, on the foraging behavior of Argentine ants in part of their native range in southern Brazil. Pseudacteon parasitoids commonly attacked Argentine ants, but not other ant species, in daylight at temperatures above 18°C. Argentine ants abandoned food resources and returned underground in the presence of parasitoids. Parasitoid attack rates diminished as Argentine ants retreated underground. Where parasitoids were present, Argentine ants were abundant at food resources only during times of day when parasitoids were inactive. Where parasitoids were absent, Argentine ants were abundant at food resources throughout the day. Overall, the presence of parasitoids explained observed variation in Argentine ant foraging far better than temperature, although temperature had some effect. The results suggest that Pseudacteon parasitoids inhibit the ability of Argentine ants to gather food resources in their native habitat in Brazil. Normal human melanocytes in culture became enlarged and dendritic after a 2-day incubation with either the pituitary (β-MSH, a potent analog of α-MSH, ACTH, FSH and LH) or the ovarian (estradiol, estriol and progesterone) hormones. Under the same experimental conditions, pituitary hormones also increased both the tyrosinase activity and tyrosinase-related protein-1 (TRP-1) while ovarian hormones increased TRP-1 but not tyrosinase activity. The results suggest that pituitary and ovarian hormones possibly induce hyperpigmentation of the skin by stimulating the melanogenesis in epidermal melanocytes, and that estradiol and progesterone may be involved in the pathogenesis of melasma (chloasma) usually developing between early adulthood and menopause in which a high concentration of serum ovarian hormones was maintained.  相似文献

Immunohistochemical localization of tyrosinase was examined with a monoclonal antibody (MoAb MAT-1) against human tyrosinase on routine formalin-fixed paraffin-embedded sections of 3 normal skin specimens, 15 melanocytic tumors (6 pigmented nevi, 3 juvenile melanomas and 6 malignant melanomas) and 3 non-melanocytic tumors. In the melanotic melanomas, almost all tumor cells were clearly stained with the antibody. In the nevocytic nevi, the nevus cells in lower epidermis and upper dermis were positive for MoAb MAT-1, but negative in middle and lower dermis. All three juvenile melanomas, one amelanotic melanoma, and three non-melanocytic tumors were entirely negative for MoAb MAT-1. Thus, MoAb MAT-1 could recognize the cells with melanogenic activity on routine formalin-fixed paraffin-embedded sections. However, the staining quality was not adequate for normal epidermal melanocytes, indicating that small technical innovations in the immunostaining process such as formalin fixation after PBS washing are required. Nevertheless, MoAb MAT-1 can be expected to be very useful for identifying melanogenic cells on paraffin-embedded sections, because we have to date no other antibody available for it.  相似文献
