A modification of the single radial immunodiffusion potency test (SRD) for rabies vaccines

In 1983 a WHO collaborative study showed that the single radial immunodiffusion test provided a reliable system for the assay of the glycoprotein content of inactivated rabies vaccines. We have modified the method used in the collaborative study by using round plastic plates with wells arranged in a circle. Potency estimates were made by the multiple slope ratio method and not by single slope estimation procedures adjusted for a common intercept. We carried out a statistical evaluation of the accuracy and reliability of the precipitation area calculation. We confirmed that for the purpose of the potency estimation the area calculations were sufficiently precise in most instances. By analysing the sources of variation for round and for glass plates and by testing the variability of these methods we found that the round plates were as good as the glass plates. The advantage of the round plates was mainly in the ease of their handling: the agarose solution was poured in the plates without the need for a separate mould; an integral moist chamber with these plates was readily available; the precipitation zones were measured easily using a viewer which gave fourfold enlargements and the method required less antiglycoprotein.     

FMN-reductase from Escherichia coli and its effect on the activity of luciferase from marine bacterium Vibrio fischeri]

Interactions of luciferases isolated from Vibrio fischeri 6 and Escherichia coli JM109(pF3) (bearing cloned V. fischeri luxAB genes) with FMN reductase isolated from E. coli JM109 were studied. FMN reductase formed a stable complex with luciferase, suggesting similar properties of the FMN reductases in the taxonomically close families Vibrionaceae and Enterobacteriaceae.     

Binding of aromatic compounds to cornstarch polysaccharides

Sorption of aromatic compounds from aqueous solutions by cryotextures and suspensions of native cornstarches was studied by capillary gas chromatography. Acetophenone and benzyl alcohol were not sorbed by cryotropic-cornstarch gel and native-cornstarch suspension. A linear concentration dependence was found for aldehydes. Phenylethyl alcohol was characterized by a nonlinear concentration dependence. The presence of a benzene ring contributed to decreased binding (relative to the level characteristic of aliphatic compounds). The degree of binding depended considerably on the type of functional group in the aromatic compounds. Cryotextures were more potent than granules of native cornstarch in binding aromatic compounds.     

Comparative study of the localization of lipids in fetal and adult human adrenal glands

The localization of lipid globules was studied in the adrenal glands from 35 fetuses and 10 adult humans by histochemistry. The fetal adrenal showed the presence of numerous lipid globules in the zona glomerulosa, whereas in the adult the lipid globules were mainly found in the zona fasciculata. These lipid globules were confirmed as cholesterol in the fetal adrenals.
