Beta 1 isoform-specific regulation of a triiodothyronine-induced gene during cerebellar development.
Although tissue-specific expression of the alpha 1 and beta 1 thyroid hormone receptors (TR-alpha 1 and TR-beta 1) suggests isoform-specific function, transfection studies to date have failed to show consistent differences in their ability to regulate gene expression. We here provide evidence that TR-beta 1 but not TR-alpha 1 regulates the expression of the gene coding for PCP-2 in cerebellar Purkinje cells during neonatal rat development and that such regulation appears to be both T3 dependent and T3 independent. Examination of neonatal rats revealed that the levels of three mRNAs expressed in cerebellar Purkinje cells (myoinositol-1,4,5-triphosphate receptor, calbindin, and PCP-2) rise from neonatal day 1 to day 15. This rise is preceded by the previously documented surge in brain T3 and TR-beta 1. Methimazole-induced hypothyroidism sharply reduces, but does not abolish, the rise in these mRNAs. Concomitant T3 administration normalizes the process. In order to establish more directly the role of TR-beta 1 and T3, cotransfection experiments were performed in CHO cells with PCP-2-lacZ construct and TR isoforms. These studies showed that TR-beta 1, even in the absence of T3, regulated the expression of the transfected PCP-2 construct. T3 augments the response to TR-beta 1 alone by 40% (P < .01). Cytokines produced by immune-activated testicular interstitial macrophages (TIMs) may play a fundamental role in the local control mechanisms of testosterone biosynthesis in Leydig cells. We investigated whether in vivo immune-activation of TIMs can modulate Leydig cell steroidogenesis. To immune activate TIMs in vivo, mice were injected intraperitoneally (i.p.) with lipopolysaccharide (LPS, 6 mg/kg). TIMs and Leydig cells were purified for RNA analysis. LPS treatment resulted in a 47-fold increase in interleukin-1β (IL-1β) mRNA in TIMs. P450c17 mRNA levels in the Leydig cells from the same animals, decreased to less than 10% compared to control. The effect of LPS on IL-1β and P450c17 mRNA levels was reversible on both TIMs and Leydig cells, respectively. To determine if the effect of LPS on P450c17 was mediated by a possible decrease in pituitary LH secretion, mice were co-injected with LPS and hCG. Treatment with hCG did not change the effect observed with LPS alone, in TIMs or in Leydig cells. In vitro, LPS treatment of TIMs resulted in marked induction of IL-1β mRNA expression. In parallel, in vitro treatment of Leydig cells with recombinant IL-1 resulted in a dose-dependent inhibition of P450c17 mRNA expression and testosterone production. These data demonstrate that LPS treatment, in vivo and in vitro, induced IL-1 gene expression in TIMs, and that IL-1 inhibits P450c17 mRNA in vitro. Therefore, we suggest that immune-activation of TIMs might have caused the observed inhibition of P450c17 gene expression in Leydig cells in vivo. 相似文献

通过DEAE-SephadexA-50,DEAE-SepharoseCL-6B,MonoQ (FPLC)三步离子交换柱层析,纯化得到一新的纤维蛋白原溶酶,在碱性聚丙烯酰胺凝胶电泳和SDS-聚丙烯酰胺凝胶电泳上均呈单一的蛋白带。分子量为2600,等电点pl4.7;它是一个糖蛋白,含糖量6.4%,其中中性糖0.3%,已糖胺4.9%,唾液酸1.2%。烙铁头蛇毒纤维蛋白原溶酶TMVFg由187个氨基酸组成,含有较高的酸性氨基酸,此外甘氨酸含量也较高。TMVFg热稳定性强,而酸不稳定,在280 nm处具有典型的蛋白吸收峰,在无离子水中紫外消光系数E0.1%/280 nm=1.558。纯化的TMVFg具有较强的精氨酸酯酶活力,对苯甲酰-L-精氨酸乙酯(BAEE)的Km值为1.4×10[-3]M。TMVFg的活性受苯甲基丹磺酰氟(PMSF)抑制;乙二胺四乙酸(EDTA)对活性无影响。TMVFg不能使纤维蛋白原凝固,但能水解纤维蛋白原α、β链。纤溶实验表明TMVFg具有激活纤溶作用。纯化的烙铁头蛇毒纤维蛋白原溶酶对酪蛋白无作用,无出血活性,因而与凝血酶样酶、出血毒素及β-纤维蛋白原溶酶(OUYANG et al.,1977)明显不同。 相似文献

本文介绍一种高分子聚合物HFMC注入输精管,遇体液即合成有孔隙(孔径2-16 μ)的固态聚合物,附着管壁并缓慢释放适度的H[+],以改变精子存活环境,从而达到狒猴非阻断性的输精管避孕。在狒猴配对试验中证明,输精管注入HFMC 30 mg×2(n=2),在注射后47-50天精液品质即逐渐恢复,已有致孕能力;输精管注入HFMC 60 mg×2(n=4),在300天后,有些个体精液品质才逐渐恢复,注后348-353天之间已有50%的雄猴配对致孕;另有50%的雄猴在注入854天时精液品质仍处于不育水平。实验结果还表明:HFMC对精子具有致死,致畸率高及使精子活力显著下降,这种作用随HFMC剂量的增大而加强,这种影响也随时间的后移而逐渐减弱;HFMC对输精管具有可通性及可复性。今后如能在注入剂量,自溶稳定性方面再改进,HFMC有可能成为男性节育较为理想的一种新型避孕剂。 相似文献

Cyanase is an inducible enzyme in Escherichia coli that catalyzes the reaction of cyanate with bicarbonate resulting in the decomposition of cyanate to ammonia and bicarbonate. In this study, the role of the single sulfhydryl group in each of the eight identical subunits of cyanase was investigated. Tetranitromethane, methyl methanethiosulfonate, N-ethylmaleimide, and Hg2+ all reacted with the sulfhydryl group to give derivatives which had reduced activities and which dissociated reversibly to inactive dimer. Association of inactive dimer to active octamer was facilitated by the presence of azide (cyanate analog) and bicarbonate, increased temperature and enzyme concentration, and presence of phosphate. Nitration of tyrosine residues by tetranitromethane occurred only in the absence of azide and bicarbonate, suggesting that at least some of the tyrosine residues become exposed when octamer dissociates to dimer. Site-directed mutagenesis was used to prepare a mutant enzyme in which serine was substituted for cysteine. The mutant enzyme was catalytically active and had properties very similar to native enzyme, except that it was less stable to treatment with urea and to high temperatures. These results establish that in native cyanase the sulfhydryl group per se is not required for catalytic activity, but it may play a role in stabilizing octameric structure, and that octameric structure is required for catalytic activity. 相似文献
