基于1977、1990、1997、2013和2017年5期Landsat和Sentinel-2遥感影像,以监督分类的结果和景观格局指数为基础,结合景观生态学理论和前人研究成果,构建了适于干旱区山地类自然遗产地的景观健康诊断体系,通过区域网格分析和空间插值等方法,对新疆天山自然遗产地喀拉峻-库尔德宁区域景观健康状况进行诊断,实现了对范围内各处景观状态的诊断,并结合无人机正射影像对诊断结果进行验证,既丰富了景观生态健康方向的研究,又为自然遗产地生态环境的监测、评估、保护与管理等提供技术方法的支撑和实际应用案例。研究结果表明:40年间冰雪、水体、针阔混交林景观缩减严重,裸地占比年变化率高达62.54%,扩张明显;研究区除1997年景观健康等级为较差外,其余年份景观健康均为一般;40年来研究区北部的景观健康状况要比南部好,同时应将零星分布的健康等级为较差、极差的区域作为今后保护工作的重点对象和典型区。  相似文献

利用GEO数据库中的芯片数据,筛选与星形细胞瘤生存预后相关的miRNA-mRNA调控关系对,为后续研究提供理论支持。下载芯片数据利用R语言进行差异表达分析,得到星形细胞瘤较正常组织表达显著改变的miRNA与mRNA;通过miRNA靶基因预测,将靶基因与差异表达mRNA取交集,明确mRNA与miRNA之间的关系;利用GEPIA2.0工具在TCGA数据库中筛选有生存价值的mRNA并验证表达情况,利用OncoLnc工具对相应miRNA进行生存分析。筛选到差异表达的miRNA 90个(表达上调22个,下调68个);差异表达的mRNA 644个(表达下调476个,上调168个);根据miRNA靶基因预测结果,整理出miRNA-mRNA关系对30个,对其中的mRNA与miRNA进行生存分析,共得到7个miRNA-mRNA关系对与LGG患者生存预后明显相关,未筛选到与GBM患者生存预后有关的调控关系对。本研究筛选到的7个miRNA-mRNA调控关系对与LGG患者生存预后显著相关,可为相关研究与治疗提供靶点和参考方向。  相似文献

以薏苡黑粉菌病瘿为研究对象,采用固体和液体培养方法对薏苡黑粉菌进行分离纯化培养和形态观察;利用Illumina、Pacbio测序和Hi-C辅助基因组组装技术对黑粉菌进行测序、组装和注释,分析其基因组结构和组成特征。通过与其他6个黑粉菌物种基因组进行比较基因组学研究,分析薏苡黑粉菌的基因家族进化和系统发育关系。在pH 7的固体PDA和液体PDB培养基中实现了薏苡黑粉菌的纯化培养,固体培养菌落表现为白色、表面隆起、具褶皱,较湿润且不透明的形态特征。Illumina、Pacbio和Hi-C组装结果显示,薏苡黑粉菌具有20对染色体(2n=40),基因组大小20.093 417 Mb,GC含量53.81%;共预测和注释到7 476个蛋白编码基因和164个非编码基因,占整个基因组长度的61.02%和0.285 3%;查找到3 674个散在重复序列和8 139个串联重复序列,分别占整个基因组长度的2.754 4%和1.605 4%。同源基因家族聚类显示,7个黑粉菌物种Ustilago coicisU. bromivoraU. maydisU. hordeiSporisorium reilianumS. scitamineumS. graminicola的编码基因被聚类为6 999个基因家族,其中共有基因家族5 379个。薏苡黑粉菌包含5 339个单拷贝基因、105个多拷贝基因、89个特异基因、752个其他基因和188个未分类基因。基因家族进化和系统发育聚类表明,薏苡黑粉菌U. coicis在~60.9 MYA分化出来,与雀麦黑粉菌U. bromivova、大麦坚黑粉菌U. hordei聚为一支,表现为更近的亲缘关系;而玉米黑粉菌U. maydis在~66.6 MYA分化出来与3个孢堆黑粉菌物种S. scitamineumS. graminicolaS. reilianum聚为一支。  相似文献

A green chemical precipitation route was used to yield a hydrated basic sulfate precursor upon calcination at 1000°C into a series of (Y,Gd)2O2SO4:Dy particles. The phosphors exhibited characteristic Dy3+ emissions from 4F9/26HJ (J = 15/2, 13/2, 11/2) transitions under ultraviolet light excitation; the quenching concentration of Dy3+ was determined to be 2.5 at.%. Substitution of Gd3+ for Y3+ led to an additional strong sharp band at ~277 nm (8S7/26IJ transition of Gd3+) in the photoluminescence excitation spectra, upon which the (Gd0.975Dy0.025)2O2SO4 phosphor achieved a ~2.8-fold higher photoluminescence intensity via an effective energy transfer from Gd3+ to Dy3+ compared with the 354 nm excitation of Dy3+. Both the photoluminescence and photoluminescence excitation intensities of (Y,Gd)2O2SO4:Dy phosphors increased with rising Gd3+ concentration and calcination temperature in the range 750–1000°C. A higher Gd3+ concentration slightly prolonged the effective fluorescence lifetime.  相似文献

Oleanolic acid (OA) derivatives exhibit numerous pleiotropic effects in many cancers. The present study aimed to investigate the molecular mechanisms of 5′-amino-oleana-2,12-dieno[3,2-d]pyrimidin-28-oic acid (compound 4) and oleana-2,12-dieno[2,3-d]isoxazol-28-oic acid (compound 5) inducing apoptosis in human leukemia K562 cell. We investigated the effects of the compounds on K562 cell growth, apoptosis and cell cycle. The compounds showed strong inhibitory effects on K562 cell viability in a dose-dependent manner determined by the 3-(4,5-dimethylthiazoyl)-2,5-diphenyltetrazolium bromide assay and significantly increased chromatin condensation and apoptotic bodies in K562 cells. Flow cytometry assay suggested that the compounds induced inhibition of K562 cell proliferation associated with G1 phase arrest. In addition, the compounds inhibited Akt1 recruiting to membrane in CHO cells which express Akt1-EGFP constitutively and down-regulated the expression of pAkt1 in K562 cell. These results suggested that the compounds can efficiently inhibit proliferation and induce apoptosis perhaps involved in inactivation of Akt1. The OA derivatives may be potential chemotherapeutic agents for the treatment of human cancer.  相似文献

The biophysical mechanism of magnetic fields (MFs) acting on living systems is not clear. Previous research showed that, similar to epidermal growth factor (EGF), MF exposure induced EGF receptor (EGFR) clustering and activated EGFR signaling. In this study, we investigated whether MF exposure induced the changes in physical characteristics of EGF and downstream effects of EGF and EGFR interaction. The phase-interrogation surface plasmon resonance (SPR) sensing analyses showed that 50 Hz MF exposure at 4.0 mT for 1 h induced reversible relative permittivity changes of EGF solution. However, compared with sham-exposed EGF solution, the MF-exposed EGF solution did not affect the binding of EGF to EGFR, nor the cell viability and EGFR clustering in human amniotic epithelial cells (FL cells). Our data suggest that cellular EGFR clustering response to MF exposure might not be a result of changes in relative permittivity of EGF in cell culture solution. Hyperbranched poly(amido amine)s (HPAAs) containing different amounts of β-cyclodextrin (β-CD) (HPAA-CDs) were synthesized in one-pot by Michael addition copolymerization of N,N'-methylene bisacrylamide, 1-(2-aminoethyl)piperazine, and mono-6-deoxy-6-ethylenediamino-β-CD. In comparison to pure HPAA, the fluorescence intensity of HPAA-CDs was enhanced significantly while the cytotoxicity became lower. Ascribed to plenty of amino groups and strong photoluminescence, HPAA-CDs could be used as nonviral gene delivery vectors, and the corresponding gene transfection was evaluated. The experimental results indicated that HPAA-CDs condensed the plasmid DNA very well. By utilizing the fluorescent properties of HPAA-CDs, the cellular uptake and gene transfection processes were tracked by flow cytometry and confocal laser scanning microscopy without any fluorescent labeling. The transfection efficiencies of HPAA-CDs were similar to that of pure HPAA. In addition, the inner cavities of β-CDs in HPAA-CDs could be used to encapsulate drugs through host--guest interaction. Therefore, the HPAA-CDs may have potential application in the combination of gene therapy and chemotherapy.  相似文献

A new type of biodegradable micelles for glutathione-mediated intracellular drug delivery was developed on the basis of an amphiphilic hyperbranched multiarm copolymer (H40-star-PLA-SS-PEP) with disulfide linkages between the hydrophobic polyester core and hydrophilic polyphosphate arms. The resulting copolymers were characterized by nuclear magnetic resonance (NMR), Fourier transformed infrared (FTIR), gel permeation chromatography (GPC), and differential scanning calorimeter (DSC) techniques. Benefiting from amphiphilic structure, H40-star-PLA-SS-PEP was able to self-assemble into micelles in aqueous solution with an average diameter of 70 nm. Moreover, the hydrophilic polyphosphate shell of these micelles could be detached under reduction-stimulus by in vitro evaluation, which resulted in a rapid drug release due to the destruction of micelle structure. The glutathione-mediated intracellular drug delivery was investigated against a Hela human cervical carcinoma cell line. Flow cytometry and confocal laser scanning microscopy (CLSM) measurements demonstrated that H40-star-PLA-SS-PEP micelles exhibited a faster drug release in glutathione monoester (GSH-OEt) pretreated Hela cells than that in the nonpretreated cells. Cytotoxicity assay of the doxorubicin-loaded (DOX-loaded) micelles indicated the higher cellular proliferation inhibition against 10 mM of GSH-OEt pretreated Hela cells than that of the nonpretreated ones. As expected, the DOX-loaded micelles showed lower inhibition against 0.1 mM of buthionine sulfoximine (BSO) pretreated Hela cells. These reduction-responsive and biodegradable micelles show a potential to improve the antitumor efficacy of hydrophobic chemotherapeutic drugs.  相似文献
