GPR40 partial agonism is a promising new mechanism for the treatment of type 2 diabetes mellitus with clinical proof of concept. Most of the GPR40 agonists in the literature have a carboxylic acid functional group, which may pose a risk for idiosyncratic drug toxicity. A novel series of GPR40 agonists containing a tetrazole as a carboxylic acid bioisostere was identified. This series of compounds features a benzo[b]thiophene as the center ring, which is prone to oxidation during phase 1 metabolism. Following SAR optimization targeting GPR40 agonist activity and intrinsic clearance in microsomes (human and rat), potent and metabolically stable compounds were selected for in vivo evaluation. The compounds are efficacious at lowering blood glucose in a SD rat oGTT model.  相似文献

Metallothionein is a ubiquitous low molecular weight metalloprotein with powerful protective properties against oxygen radical-mediated cytotoxicity associated with inflammatory processes. In rheumatoid arthritis, the inflammatory damage to the synovium appears to be mediated by free radicals released by the high concentration of neutrophils found in the synovial fluid of the inflamed joint. Synovial tissue obtained during routine surgery on rheumatoid and non-rheumatoid joints was subjected to an indirect immunoperoxidase protocol for the immunolocalization of metallothionein using mouse monoclonal anti-metallothionein antibody E9, reactive against the two major isoforms of mammalian metallothionein. A layer of large dendritic-like cells situated subsynovially in the rheumatoid synovium stained very positively for the metalloprotein, both cytoplasmically and in their nuclei. These cells were not found in non-rheumatoid osteoarthritic or in undamaged synovial tissue associated with traumatic joint injury. An attempt was made to investigate their lineage using a series of antibody markers against epithelial cells, endothelial cells, smooth muscle, mesothelial cells, fibroblasts, neutrophils, dermal dendrocytes, macrophages, low and high molecular weight cytokeratin as well as a cell proliferation marker. From our results, it is suggested that these metallothionein-positive cells are probably myofibroblasts similar to the highly motile cells present in granulation tissue. They may originate from perivascular areas of synovium and their movement into the inflamed synovium may reflect the cytoprotective role of metallothionein acting as a free radical scavenger against oxidative damage  相似文献

Polyphenol oxidase (PPO) activity has been reported in orchard grass (Dactylis glomerata); however, to date, no endogenous substrates have been identified. In the present study, we report the isolation and structural elucidation of PPO substrates in this species. The free phenol fraction was extracted, separated by reverse-phase chromatography and six potential substrates, including two hydroxycinnamate esters, were identified by UV spectrometry, electrospray ionisation-tandem mass spectrometry (LC-ESI-MSn) and 1D and 2D NMR analyses (1H NMR, 13C NMR, DEPT, COSY, HMQC and HMBC). Furthermore, three caffeoylquinic acids (3-CQA, 4-CQA and 5-CQA) were identified by comparison of their spectral data (ESI-MS) with those of known compounds and literature data. Five of these compounds were demonstrated to be substrates for orchard grass PPO.  相似文献

Renal fibrosis is the common histological feature of advanced glomerular and tubulointerstitial disease leading to end-stage renal disease (ESRD). However, specific antifibrotic therapies to slow down the evolution to ESRD are still absent. Because persistent inflammation is a key event in the development of fibrosis, we hypothesized that the proinflammatory kinin B1 receptor (B1R) could be such a new target. Here we show that, in the unilateral ureteral obstruction model of renal fibrosis, the B1R is overexpressed and that delayed treatment with an orally active nonpeptide B1R antagonist blocks macrophage infiltration, leading to a reversal of the level of renal fibrosis. In vivo bone marrow transplantation studies as well as in vitro studies on renal cells show that part of this antifibrotic mechanism of B1R blockade involves a direct effect on resident renal cells by inhibiting chemokine CCL2 and CCL7 expression. These findings suggest that blocking the B1R is a promising antifibrotic therapy.  相似文献

Assessing environmental impacts on coral reef communities has become a growing discipline. As most corals grow relatively slowly, the common method of monitoring changes in coral communities may limit our ability to identify stressors and stress responses. Since chlorophyll density (amount of chlorophyll a + c2 per unit of coral surface area) in corals may correlate with coral color, the latter has been suggested as an indicator of the natural seasonal changes in undisturbed (“healthy”) corals, as well as an indicator of environmentally-induced stresses in corals, including those related to bleaching. However, the color of underwater objects as perceived through the naked eye or a camera is affected by attenuation of irradiance with depth, changes in spectral properties of underwater light, turbidity and dissolved materials in the water, among other factors. Thus, methodological issues have so far held back the use of color as a quantitative indicator for chlorophyll content. Presented here are two accurate and easy to use methods for quantitative measurements of chlorophyll density from digital photographs of corals. In these methods, the intensities of the red, green and blue channels in digital photographs were compared to measured chlorophyll densities in Stylophora pistillata. Variations in external light, known to bias the true color in underwater images, were eliminated either by photographing corals through a specially built funnel with an internal light source or by mathematically normalizing color channels to a gray reference scale. In both methods, chlorophyll density was highly correlated with the intensity of the red channel, despite large variations in lighting conditions during the photography.These photographic methods enabled the estimation of natural spatial and temporal changes of the chlorophyll density of corals. By predicting chlorophyll density in corals at very low costs, both methods presented here could facilitate the study of large-scale physiological changes in corals.  相似文献

Background  

DNA repair is the general term for the collection of critical mechanisms which repair many forms of DNA damage such as methylation or ionizing radiation. DNA repair has mainly been studied in experimental and clinical situations, and relatively few information-based approaches to new extracting DNA repair knowledge exist. As a first step, automatic detection of DNA repair proteins in genomes via informatics techniques is desirable; however, there are many forms of DNA repair and it is not a straightforward process to identify and classify repair proteins with a single optimal method. We perform a study of the ability of homology and machine learning-based methods to identify and classify DNA repair proteins, as well as scan vertebrate genomes for the presence of novel repair proteins. Combinations of primary sequence polypeptide frequency, secondary structure, and homology information are used as feature information for input to a Support Vector Machine (SVM).  相似文献

Visual cues leading to host selection and landing are of major importance for aphids and evidence suggests that flight activity is very dependent on ultraviolet (UV)‐A radiation in the environment. At the same time research on insect plant hosts suggest that the UV‐B component can deter some pests via changes in secondary metabolite chemistry. Here, we examine the potential of UV (UV‐A/UV‐B) radiation to control insect pests in the glasshouse environment. We first examined artificial exposure to UV‐B and the potential to trigger morphological and biochemical modifications in pepper (Capsicum annuum L., Solanaceae) with implications for the fitness of green peach aphid, Myzus persicae Sulzer (Hemiptera: Aphididae). UV‐B caused accumulation of leaf secondary metabolites and soluble carbohydrates, and stimulated photosynthetic pigments. However, UV‐B did not impact on foliar protein content and aphid performance was unaffected. Next, we studied how altering the UV‐A/UV‐B ratio environment affected aphid orientation and spatial distribution over time, either directly or by exposing plants to supplemental UV before insect introduction. Aphids directly settled and dispersed on their host pepper plants more readily in the presence of supplemental UV‐A and UV‐B. In the control treatment with ambient glasshouse UV‐A and UV‐B, insects remained more aggregated. Furthermore, insects were less attracted to peppers pre‐exposed to supplemental UV‐A and UV‐B radiation. Our results suggest that suppression of UV‐A and UV‐B inside the protected environment reduces aphid colonization and dispersal. Furthermore, application of moderate exposure of young pepper plants to supplemental UV‐B radiation could aid in protection from the colonization by phytophagous insects.  相似文献
