Heat shock protein 72 (Hsp72) has been detected in the peripheral circulation of humans. Because intracellular Hsp72 binds to aggregated proteins, we hypothesized that postexercise plasma-derived Hsp72 concentrations would be greater than serum-derived Hsp72 because of binding of Hsp72 to aggregated clotting proteins in serum. Postexercise serum, heparin, and ethylenediaminetetraacetic acid (EDTA) samples were collected from 9 recreationally active males and were analyzed for Hsp72 by enzyme-linked immunosorbent assay. In line with our hypothesis, EDTA-treated blood was significantly higher in Hsp72 concentration than all other treatments (P < or = 0.001), whilst heparin plasma (LH) was significantly higher than serum derived on ice (SI) and at room temperature (SR) (P < 0.05; EDTA: 6.46 +/- 0.76, LH: 2.73 +/- 2.26, SI: 0.13 +/- 0.24, SR: 0.20 +/- 0.32 ng/mL). Because previous research has tended to report serum data at the lowest point of the detectable range of the assay, it is recommended that EDTA specimen tubes be used in future investigations.  相似文献

One main aim with genetic modification (GM) of trees is to produce plants that are resistant to various types of pests. The effectiveness of GM-introduced toxins against specific pest species on trees has been shown in the laboratory. However, few attempts have been made to determine if the production of these toxins and reduced herbivory will translate into increased tree productivity. We established an experiment with two lines of potted aspens (Populus tremula×Populus tremuloides) which express Bt (Bacillus thuringiensis) toxins and the isogenic wildtype (Wt) in the lab. The goal was to explore how experimentally controlled levels of a targeted leaf beetle Phratora vitellinae (Coleoptera; Chrysomelidae) influenced leaf damage severity, leaf beetle performance and the growth of aspen. Four patterns emerged. Firstly, we found clear evidence that Bt toxins reduce leaf damage. The damage on the Bt lines was significantly lower than for the Wt line in high and low herbivory treatment, respectively. Secondly, Bt toxins had a significant negative effect on leaf beetle survival. Thirdly, the significant decrease in height of the Wt line with increasing herbivory and the relative increase in height of one of the Bt lines compared with the Wt line in the presence of herbivores suggest that this also might translate into increased biomass production of Bt trees. This realized benefit was context-dependent and is likely to be manifested only if herbivore pressure is sufficiently high. However, these herbivore induced patterns did not translate into significant affect on biomass, instead one Bt line overall produced less biomass than the Wt. Fourthly, compiled results suggest that the growth reduction in one Bt line as indicated here is likely due to events in the transformation process and that a hypothesized cost of producing Bt toxins is of subordinate significance.  相似文献

Introduction

Positron Emission Tomography - Computer Tomography (PET-CT) is an interesting imaging technique to visualize Ankylosing Spondylitis (AS) activity using specific PET tracers. Previous studies have shown that the PET tracers [18F]FDG and [11C](R)PK11195 can target inflammation (synovitis) in rheumatoid arthritis (RA) and may therefore be useful in AS. Another interesting tracer for AS is [18F]Fluoride, which targets bone formation. In a pilot setting, the potential of PET-CT in imaging AS activity was tested using different tracers, with Magnetic Resonance Imaging (MRI) and conventional radiographs as reference.

Methods

In a stepwise approach different PET tracers were investigated. First, whole body [18F]FDG and [11C](R)PK11195 PET-CT scans were obtained of ten AS patients fulfilling the modified New York criteria. According to the BASDAI five of these patients had low and five had high disease activity. Secondly, an extra PET-CT scan using [18F]Fluoride was made of two additional AS patients with high disease activity. MRI scans of the total spine and sacroiliac joints were performed, and conventional radiographs of the total spine and sacroiliac joints were available for all patients. Scans and radiographs were visually scored by two observers blinded for clinical data.

Results

No increased [18F]FDG and [11C](R)PK11195 uptake was noticed on PET-CT scans of the first 10 patients. In contrast, MRI demonstrated a total of five bone edema lesions in three out of 10 patients. In the two additional AS patients scanned with [18F]Fluoride PET-CT, [18F]Fluoride depicted 17 regions with increased uptake in both vertebral column and sacroiliac joints. In contrast, [18F]FDG depicted only three lesions, with an uptake of five times lower compared to [18F]Fluoride, and again no [11C](R)PK11195 positive lesions were found. In these two patients, MRI detected nine lesions and six out of nine matched with the anatomical position of [18F]Fluoride uptake. Conventional radiographs showed structural bony changes in 11 out of 17 [18F]Fluoride PET positive lesions.

Conclusions

Our PET-CT data suggest that AS activity is reflected by bone activity (formation) rather than inflammation. The results also show the potential value of PET-CT for imaging AS activity using the bone tracer [18F]Fluoride. In contrast to active RA, inflammation tracers [18F]FDG and [11C](R)PK11195 appeared to be less useful for AS imaging.  相似文献

Community-level genetic specificity results when individual genotypes or populations of the same species support different communities. Our review of the literature shows that genetic specificity exhibits both life and afterlife effects; it is a widespread phenomenon occurring in diverse taxonomic groups, aquatic to terrestrial ecosystems, and species-poor to species-rich systems. Such specificity affects species interactions, evolution, ecosystem processes and leads to community feedbacks on the performance of the individuals expressing the traits. Thus, genetic specificity by communities appears to be fundamentally important, suggesting that specificity is a major driver of the biodiversity and stability of the world's ecosystems.  相似文献

Particle bombardment and Agrobacterium-mediated transformation are two popular methods currently used for producing transgenic maize. Agrobacterium-mediated transformation is expected to produce transformants carrying fewer copies of the transgene and a more predictable pattern of integration. These putative advantages, however, tradeoff with transformation efficiency in maize when a standard binary vector transformation system is used. Using Southern, northern, real-time PCR, and real-time RT-PCR techniques, we compared transgene copy numbers and RNA expression levels in R1 and R2 generations of transgenic maize events generated using the above two gene delivery methods. Our results demonstrated that the Agrobacterium-derived maize transformants have lower transgene copies, and higher and more stable gene expression than their bombardment-derived counterparts. In addition, we showed that more than 70% of transgenic events produced from Agrobacterium-mediated transformation contained various lengths of the bacterial plasmid backbone DNA sequence, indicating that the Agrobacterium-mediated transformation was not as precise as previously perceived, using the current binary vector system.  相似文献

Interactions among elk, aspen, galling sawflies and insectivorous birds   总被引:3,自引:0,他引:3  
Using two years of observational and experimental data, we examined the hypothesis that browsing by elk on aspen indirectly affects the distribution of a leaf-galling sawfly, which in turn affects insect diversity and foraging patterns of insectivorous birds. We found that: i) in an analyses of 33 arthropod species, the presence of sawflies significantly increased arthropod richness and abundance by 2 X and 2.5 X, respectively. ii) browsing by elk reduced sawfly gall abundance such that 90% of the galls were found on unbrowsed aspen ramets. iii) insectivorous birds attacked 60–74% of the galls on unbrowsed shoots compared to 11% on browsed shoots. When leaf-galler abundance was experimentally held constant on browsed and unbrowsed shoots, predation by insectivorous birds did not differ significantly. This result suggests that browsing affects the patterns of avian predation by altering the distribution of a galling insect. These data argue that bottom-up, top-down, and lateral factors can act in concert to affect the distribution of a galler, structure arthropod communities and affect predation by insectivorous birds.  相似文献

Although mistletoe is typically viewed as a parasite of juniper in a two-way interaction, its role may become neutral or even mutualistic when their common avian seed dispersing agents are considered as a three-way interaction. In the study area, wintering avian frugivores forage on both one-seed juniper (Juniperus monosperma) berries and on the fruit of its associated mistletoe (Phoradendron juniperinum). Three major findings emerged from our studies that support a three-way interaction and the hypothesis of conditional interactions. First, mistletoes provide a stable resource for shared avian seed dispersers; junipers do not. Whereas juniper berry production varied 10- to 15-fold over the 3 years of our study, mistletoe fruit abundance did not vary significantly. Second, the abundance of avian seed dispersal agents, such as Townsend's solitaires (Myadestes townsendi), is strongly tied to the abundance of juniper berries in mast years and mistletoe fruits in all years. In fact, the best overall predictor of their common avian seed dispersal agents was the abundance of mistletoe; stands with mistletoe attracted up to 3 times more avian frugivores than stands with little or no mistletoe. Thus, mistletoe berries can serve as the main attractor for birds that disperse juniper berries. Third, in agreement with the hypothesis that mistletoe can benefit junipers by attracting and supporting greater populations of avian seed dispersal agents, the number of juniper seedlings was more than 2-fold greater in stands with high mistletoe density compared with stands that had little or no mistletoe. Results suggest that the occurrence of a three-way interaction, in the presence of environmental variation (in this case, annual variation in juniper berry crops), may change the ecological roles of associated species. A conceptual model is presented to illustrate how the role of mistletoe may range from parasitic to mutualistic, while the role of avian seed dispersers may conversely range from mutualistic to parasitic, the latter by acting as vectors for the spread of mistletoe.  相似文献

Abstract: A detailed analysis of the generation and subsequent metabolism of inositol 1,4,5-trisphosphate [Ins(1,4,5)P3] following muscarinic cholinoceptor stimulation in primary cultures of rat cerebellar granule cells has been undertaken. Following incubation of cerebellar granule cell cultures with [3H]inositol for 48 h, labelling of the inositol phospholipid pool approached equilibrium. Significant basal labelling of inositol pentakisphosphate (InsP5) and inositol hexakisphosphate (InsP6), as well as inositol mono- to tetrakisphosphate, fractions was observed. Addition of carbachol (1 m M ) caused an immediate increase in level of Ins(1,4,5)P3 (peak increase two-fold over basal by 60 s), which was well-maintained over the initial 300 s following agonist addition. In contrast, only a modest, more slowly developing, increase in inositol tetrakisphosphate accumulation was observed, whereas labelling of InsP5 and InsP6 was entirely unaffected by carbachol stimulation. Analysis of the products of Ins(1,4,5)P3 and inositol 1,3,4,5-tetrakisphosphate metabolism in broken cell preparations strongly suggested that Ins(1,4,5)P3 metabolism occurs predominantly via the inositol polyphosphate 5-phosphatase route, with metabolism via the Ins(1,4,5)P3 3-kinase being a relatively minor pathway. In view of the pattern of inositol (poly)phosphate metabolites observed on stimulation of the muscarinic receptor, it seems likely that, over the time course studied, the inositol polyphosphates are derived principally from phosphoinositide-specific phospholipase C hydrolysis of phosphatidylinositol 4,5-bisphosphate, although some hydrolysis of phosphatidylinositol 4-phosphate cannot be excluded.  相似文献

Understanding the local and regional patterns of species distributions has been a major goal of ecological and evolutionary research. The notion that these patterns can be understood through simple quantitative rules is attractive, but while numerous scaling laws exist (e.g., metabolic, fractals), we are aware of no studies that have placed individual traits and community structure together within a genetics based scaling framework. We document the potential for a genetic basis to the scaling of ecological communities, largely based upon our long-term studies of poplars (Populus spp.). The genetic structure and diversity of these foundation species affects riparian ecosystems and determines a much larger community of dependent organisms. Three examples illustrate these ideas. First, there is a strong genetic basis to phytochemistry and tree architecture (both above- and belowground), which can affect diverse organisms and ecosystem processes. Second, empirical studies in the wild show that the local patterns of genetics based community structure scale up to western North America. At multiple spatial scales the arthropod community phenotype is related to the genetic distance among plants that these arthropods depend upon for survival. Third, we suggest that the familiar species-area curve, in which species richness is a function of area, is also a function of genetic diversity. We find that arthropod species richness is closely correlated with the genetic marker diversity and trait variance suggesting a genetic component to these curves. Finally, we discuss how genetic variation can interact with environmental variation to affect community attributes across geographic scales along with conservation implications.  相似文献
