DAPK3 inhibits gastric cancer progression via activation of ULK1-dependent autophagy

Dysregulation of the balance between cell proliferation and cell death is a central feature of malignances. Death-associated protein kinase 3 (DAPK3) regulates programmed cell death including apoptosis and autophagy. Our previous study showed that DAPK3 downregulation was detected in more than half of gastric cancers (GCs), which was related to tumor invasion, metastasis, and poor prognosis. However, the precise molecular mechanism underlying DAPK3-mediated tumor suppression remains unclear. Here, we showed that the tumor suppressive function of DAPK3 was dependent on autophagy process. Mass spectrometry, in vitro kinase assay, and immunoprecipitation revealed that DAPK3 increased ULK1 activity by direct ULK1 phosphorylation at Ser556. ULK1 phosphorylation by DAPK3 facilitates the ULK1 complex formation, the VPS34 complex activation, and autophagy induction upon starvation. The kinase activity of DAPK3 and ULK1 Ser556 phosphorylation were required for DAPK3-modulated tumor suppression. The coordinate expression of DAPK3 with ULK1 Ser556 phosphorylation was confirmed in clinical GC samples, and this co-expression was correlated with favorable survival outcomes in patients. Collectively, these findings indicate that the tumor-suppressor roles of DAPK3 in GC are associated with autophagy and that DAPK3 is a novel autophagy regulator, which can directly phosphorylate ULK1 and activate ULK1. Taxonomic notes on the paper wasps of the genus Ropalidia in the Indian subcontinent (Hymenoptera: Vespidae)

The taxonomy of Ropalidia wasps in the Indian subcontinent is revised, recognizing 26 species in the subcontinent. Their diagnostic characteristics are summarized in a key to species. New synonymies proposed in the present study are: R. bicolorata shiva Das and Gupta, 1989 under R. bicolorata van der Vecht, 1962; R. colorata sordida van der Vecht, 1941 under R. colorata van der Vecht, 1941; R. rodialipa Lambert and Narendran, 2005 and R. anupama Lambert and Narendran, 2005, both under R. cyathiformis Fabricius, 1804; R. jacobsoni flavoscutellata Das and Gupta, 1989, and R. bangalorica Lambert and Narendran, 2005, both under R. jacobsoni du Buysson, 1908; R. travancorica Lambert and Narendran, 2005, under R. marginata Lepeletier, 1836; R. sridharani Lambert and Narendran, 2005, under R. rufocollaris Cameron, 1900; Ropalidia rufoplagiata nursei van der Vecht, 1941 under R. rufoplagiata Cameron, 1905; Icaria lugubris Smith, 1858, under R. sumatrae Weber, 1801; and a revised synonymy is Icaria pendula Smith, 1857, under R. variegata Smith, 1852. The new replacement name Ropalidia kasaragodensis Lambert and Narendran is proposed for R. indica Lambert and Narendran, 2005, non van der Vecht, 1941.     

Possible circulation of H5N1 avian influenza viruses in healthy ducks on farms in northern Vietnam

To estimate the prevalence of influenza A subtype H5N1 viruses among domestic ducks in the period between October and November 2006 when H5N1 outbreaks had been absent, 1106 healthy ducks raised in northern Vietnam were collected. Inoculation of all throat and cloacae samples into embryonated eggs resulted in the isolation of subtype H3N8 in 13 ducks, but not H5N1 viruses. Serological analyses demonstrated that five ducks (0.45%) solely developed H5N1 subtype-specific hemagglutinin-inhibiting and neuraminidase-inhibiting antibodies together with anti-non-structural protein 1 antibodies. The results suggested that the ducks were naturally infected with H5N1 viruses when obvious H5N1 outbreaks were absent.     

An Application of Renewal Theory to Contraceptive Follow-up Studies,Exemplified by an Intrauterine Device Study

Contraceptive practice consists of a series of recurrent events of use and nonuse. Application of the life table technique for analysis of multiple-segment contraceptive usage is not entirely satisfactory. In this paper the renewal process approach is applied to the results of contraceptive follow-up studies involving all segments of use. By means of this model, it is possible to estimate the proportion of current users, the average duration of actual use, and the average number of segments used by the acceptors. The practical importance of these parameters for the evaluation of family planning programs is considered.     

Properties and primary structure of a thermostable l-malate dehydrogenase from Archaeoglobus fulgidus

A thermostable l-malate dehydrogenase from the hyperthermophilic sulfate-reducing archaeon Archaeoglobus fulgidus was isolated and characterized, and its gene was cloned and sequenced. The enzyme is a homodimer with a molecular mass of 70 kDa and catalyzes preferentially the reduction of oxaloacetic acid with NADH. A. fulgidus l-malate dehydrogenase was stable for 5 h at 90° C, and the half-life at 101° C was 80 min. Thus, A. fulgidus l-malate dehydrogenase is the most thermostable l-malate dehydrogenase characterized to date. Addition of K₂HPO₄ (1 M) increased the thermal stability by 40%. The primary structure shows a high similarity to l-lactate dehydrogenase from Thermotoga maritima and gram-positive bacteria, and to l-malate dehydrogenase from the archaeon Haloarcula marismortui and other l-lactate-dehydrogenase-like l-malate dehydrogenases.
