Plant growth and nutrient uptake characteristics of Fe-deficiency chlorosis susceptible and resistant subclovers

The objective of this study was to evaluate the growth and nutrient-uptake characteristics of Fe-deficiency resistant and susceptible subclover (Trifolium subterraneum L., T. yanninicum Katzn. and Morley, T. brachcalycinum Katzn. and Morley) cultivars on a calcareous soil. Ten subclover cultivars showing varying susceptibilities to Fe-deficiency chlorosis (Karridale, Nangeela, Geraldton, Mt. Barker, Woogenellup, Larisa, Trikkala, Rosedale, Koala and Clare) were grown on a low-Fe, calcareous soil (Petrocalcic Paleustoll) under moist (18% water content, 85% of water holding capacity) and water-saturated conditions using a Cone-tainer^® culture system. Chlorosis and its correlation with growth traits and mineral nutrition of the 10 cultivars were examined. The Fe-deficiency susceptibilities of the 10 cultivars decreased in the above order under the moist condition, but in slightly different order under the saturated condition. Shoot and root dry weights, total dry weight, and root-to-shoot ratio were each negatively correlated with chlorosis under both soil-moisture conditions, as was total shoot content of P, Ca, Fe, Mn and Zn. Shoot P and Fe concentrations were each positively correlated with chlorosis under the moist soil condition. Iron and Cu utilization efficiencies (biomass per unit weight of nutrient) in the shoot were each negatively correlated with chlorosis under the moist soil condition. These results suggest that there may be several characteristics of Fe-deficiency chlorosis resistance in subclovers, such as a more effective soil-Fe mobilizing mechanism(s), more balanced nutrition, lower required Fe concentration in the shoot, higher shoot-Fe utilization efficiency, and higher root/shoot ratio under Fe-deficiency stress conditions.     

甘蓝型油菜下胚轴原生质体培养的研究

从甘蓝型油菜下胚轴分离纯化原生质体。种子萌发时进行光照处理对原生质体产率影响不大,但可提高其活力。对液体浅层、固体平板和“琼脂岛”3种培养方法进行比较,结果“琼脂岛”法效果最好,不但细胞分裂速度快,而且克隆形成频率高。再生的愈伤组织转到分化培养基上后迅速分化出芽。诱导生根后进行移栽,生长状况良好。

广西崇左县冲塘新石器时代人骨微量元素的初步研究

本文对广西崇左县冲塘新石器时代的３个个体（Ｍ０２,Ｓ０８,Ｓ０９）的肢骨进行了化学元素（Ｓｒ,Ｚｎ,Ｃａ,Ｐ,Ｃｄ,Ｐｈ）的含量测定．并根据研究结果对当时人类的生活环境、食物性质作了推测和分析．     

cDNA approaches to isolation of the mouse mutant weaver gene

The mouse autosomal recessive mutant gene weaver (wv) results in abnormalities in cerebellum, substantia nigra and testis. Although a subtracted cDNA library prepared by removing P31 (wv/wv) sequences from a P1 (wv/+) library should contain mainly nonrepetitive neonatal sequences, unfortunately, repetitive sequences still appear during screening. Two clones, one repetitive, the other not, are used to illustrate the problems encountered in attempting to isolate the weaver gene from a subtracted cDNA library.Special issue dedicated to Dr. Sidney Ochs.     

杂交水稻及其“三系”线粒体DNA的AP—PCR指纹图谱

为了研究水稻（Ｏｒｙｚａ ｓａｔｉｖａ Ｌ．）细胞质雄性不育（ＣＭＳ）与线粒体基因组的关系,应用ＡＰ－ＰＣＲ 分析,用７ 个任意单引物对６ 种水稻品系线粒体ＤＮＡ 进行了扩增。水稻线粒体ＤＮＡ 的ＡＰ－ＰＣＲ 产物可分为三种类型：（１）所有供试品系均能扩增的片段,它们代表了线粒体ＤＮＡ 在进化上的保守性序列。有４ 个引物检测到这类片段。（２）２ 个以上水稻品系共同出现而在全部供试材料间存在差异的扩增片段,这类片段是检测水稻线粒体ＤＮＡ多态性的主要来源。（３）一种细胞质类型所特有的扩增片段,从引物Ｒ２ 和Ｖ５ 的扩增产物中发现了这类片段,它们可能与ＣＭＳ有关联。另外,ＷＡ型不育系珍汕９７Ａ 与其杂种之间在６ 个引物的扩增图谱上均存在不同程度的差异,说明两者的线粒体ＤＮＡ序列结构可能存在某种差别     

根癌农杆菌对甘蓝型油菜的转化及转基因植株的再生

用根癌农杆菌（Ａｇｒｏｂａｃｔｅｒｉｕｍ ｔｕｍ ｅｆａｃｉｅｎｓ）共培养法把外源基因导入甘蓝型油菜（Ｂｒａｓｓｉ－ｃａ ｎａｐｕｓＬ．）主要栽培品种“云北２ 号”,获得转基因植株。所用外植体为带有１—２ ｍ ｍ 子叶柄的完整子叶,根癌农杆菌为Ａ２０８ＳＥ（ｐＴｉＴ３７－ＳＥ, ｐＲＯＡ９３）。Ｔｉ质粒ｐＲＯＡ９３ 带有ＮＰＴⅡ及ＧＵＳ嵌合基因。共培养２ 天后转到附加２５ ｍ ｇ／Ｌ卡那霉素的分化培养基（ＭＳ＋ ４．５ ｍ ｇ／ＬＢＡＰ）上。ＡｇＮＯ３ 和羧苄青霉素促进芽的分化,头孢霉素则有抑制作用。最高转化频率为２７％ 。把分化出的茎芽切下,插入含有２５ ｍ ｇ／Ｌ卡那霉素的生根培养基中。羧苄青霉素不利于根的形成。把完整抗性植株移入盛土壤的盆中,生长状况良好。测定β－葡糖苷酸酶活性,８４％ 明显高于对照。以ＮＰＴⅡ基因作探针进行Ｓｏｕｔｈｅｒｎ ｂｌｏｔ分析,证实外源基因已插入到植物细胞基因组中     

三褶脉紫菀中的新二萜甙

三褶脉紫菀（ＡｓｔｅｒａｇｅｒａｔｏｉｄｅｓＴｕｒｃｚ．）系菊科多年生草本植物,遍布全国,是民间常用的中药,有清热解毒、祛痰镇咳的功效［１,２］。化学工作者们曾从其同属植物紫菀（ＡｓｔｅｒｔａｒａｒｉｃｕｓＬ．ｆ．）中分离到紫菀酮（ｓｈｉｏｎｏｎｅ）、槲皮素（ｑｕｅｒｃｅｔｉｎ）、无羁萜（ｆｒｉｅｄｅｌｉｎ）、表无羁萜（ｅｐｉｆｒｉｅｄｅｌｉｎｅｌ）、毛叶醇（ｌａｃｈｎｏｐｈｙｌｌｏｌ）、乙酸毛叶酯（ｌａｃｈｎｏｐｈｙｌｌｏｌａｃｅｔａｔｅ）、茴香醚（ａｎｅｔｈｏｌｅ）以及紫菀三萜皂甙［２—４］…     

边界层阻力在叶片气体交换过程中的作用

作者对现有的气体交换法的叶室作了改进。从实验上检验了作者根据电学类比分析和模拟检验得到的结论：在已有的气体交换测定装置中,空气流速不变,叶片的边界层导度也固定不变,所得到的结论用于边界层导度变化的情形时,会造成比较大的误差。现加一障碍物使得流过叶片的风速减小,从而改变叶片的边界层导度。通过测量在不同边界层导度下同一叶片的光强曲线可以考察不同边界层导度下的气孔导度、光合速率、蒸腾速率以及它们之间的相互关系。实验结果与作者以前的电学类比分析和模拟得到的结论是基本一致的     

Crystallization and preliminary crystallographic studies of the precursor and mature forms of a neutral lipase from the fungus Rhizopus delemar

A neutral lipase from the filamentous fungus Rhizopus delemar has been crystallized in both its proenzyme and mature forms. Although the latter crystallizes readily and produces a variety of crystal forms, only one was found to be suitable for X-ray studies. It is monoclinic (C2, a = 92.8 Å, b = 128.9 Å, c = 78.3 Å, β = 135.8) with two molecules in the asymmetric unit related by a noncrystallographic diad. The prolipase crystals are orthorhombic (P2₁2₁2₁, with a = 79.8 Å, b = 115.2 Å, c = 73.0 Å) and also contain a pair of molecules in the asymmetric unit. Initial results of molecular replacement calculations using the refined coordinates of the related lipase from Rhizomucor miehei identified the correct orientations and positions of the protein molecules in the unit cells of crystals of both proenzyme and the mature form. © 1994 John Wiley & Sons, Inc.
