We present a case study of metabolism, life history and aging in the western terrestrial garter snake (Thamnophis elegans). Early research in the field supported the rate-of-living hypothesis as an explanation of aging, which was based on an apparent negative relationship between mass-specific metabolic rate and lifespan in endotherms. This hypothesis in its original form has not withstood additional tests and comparisons between the two main lineages of endotherms-birds and mammals, but there is still much to be discovered of the causative links among rate of oxygen consumption, physiology and life history, particularly in ectothermic reptiles. We present data that show adult short-lived snakes, from naturally occurring ecotypes of garter snakes, have higher mass-specific resting metabolic rates at any given body mass (metabolic intensity) across a series of normal activity temperatures (15-32°C). The short-lived ecotype in this geographic region reaches a larger body size, and has life-history traits that place it at the fast end of a pace-of-life continuum (fast growth, early maturation, high reproductive output) relative to individuals of the small-bodied long-lived ecotype. The difference between ecotypes in metabolic intensity, even after acclimation to identical conditions, may reflect evolutionary divergence and genetic differences between ecotypes. The difference in metabolic intensity is not, however, present at birth, so an alternative is that developmental environment may permanently influence metabolic rate and life history. Such developmental canalization could lead to altered gene expression via environmental influences on the epigenome and result in altered metabolic trajectories in the snakes' natural habitats. 相似文献

We know very little about physiological constraints on the evolution of life-history traits in general, and, in particular, about physiological and molecular adjustments that accompany the evolution of variation in lifespan. Identifying mechanisms that underlie adaptive variation in lifespan should provide insight into the evolution of trade-offs between lifespan and other life-history traits. Telomeres, the DNA caps at the ends of linear chromosomes, usually shorten as animals age, but whether telomere rate of change is associated with lifespan is unknown. We measured telomere length in erythrocytes from five bird species with markedly different lifespans. Species with shorter lifespans lost more telomeric repeats with age than species with longer lifespans. A similar correlation is seen in mammals. Furthermore, telomeres did not shorten with age in Leach's storm-petrels, an extremely long-lived bird, but actually lengthened. This novel finding suggests that regulation of telomere length is associated not only with cellular replicative lifespan, but also with organismal lifespan, and that very long-lived organisms have escaped entirely any telomeric constraint on cellular replicative lifespan. 相似文献

Animals must make "decisions" (e.g., when or whether to breed,the effort to put into a breeding episode) by integrating physiological,environmental and social inputs. This integration can be studiedonly in a field context. In Adélie penguins (Pygoscelisadeliae) reproduction is constrained by foraging ecology, modeof transport, and the extreme latitude at which they live. Thedecision whether to breed in a given year is influenced by bodyconditions. Adélie penguins must fast for several weeksduring the early reproductive stages and use stored fat formetabolic energy. Females that return to the colony, but donot breed, are 1012% lighter than females that do breed.Birds that are relatively low in body mass tend to have lowerreproductive success than heavier birds, and an individual'sreproductive success is positively correlated with the bodyfat stores it had on arrival. After eggs are laid, parents alternatein attending the nest. Nest failure occurs if one parent doesnot make a timely return and its fasting partner must eventuallyleave. During normal-length fasts plasma corticosterone andglucose levels do not change. Bone metastasis in the hand is rare. The etiology is quite different from that of metastasis to other bones; bronchogenic
carcinoma is by far the most frequent case. Distal phalanges are mainly involved with irregular osteolysis and cortical destruction.
Differential diagnosis of phalangeal metastasis includes osteomyelitis, rheumatoid arthritis and gout. The prognosis is always
that of metastatic bronchial cancer with an average survival of three months. Treatment may involve distal digital amputation
or antalgic radiotherapy. A case of bronchogenic carcinoma with metastasis to the thumb is presented. The metastasis was located

Microsatellite variation in Drosophila melanogaster and Drosophila simulans: a reciprocal test of the ascertainment bias hypothesis 总被引:2,自引:1,他引:1
Interspecific comparisons of microsatellite loci have repeatedly shown that
the loci are longer and more variable in the species from which they are
derived (the focal species) than are homologous loci in other (nonfocal)
species. There is debate as to whether this is due to directional evolution
or to an ascertainment bias during the cloning and locus selection
processes. This study tests these hypotheses by performing a reciprocal
study. Eighteen perfect dinucleotide microsatellite loci identified from a
Drosophila simulans library screen and 18 previously identified in an
identical Drosophila melanogaster library screen were used to survey
natural populations of each species. No difference between focal and
nonfocal species was observed for mean PCR fragment length. However,
heterozygosity and number of alleles were significantly higher in the focal
species than in the nonfocal species. The most common allele in the
Zimbabwe population of both species was sequenced for 31 of the 36 loci.
The length of the longest stretch of perfect repeat units is, on average,
longer in the focal species than in the non-focal species. There is a
positive correlation between the length of the longest stretch of perfect
repeats and heterozygosity. The difference in heterozygosity can thus be
explained by a reduction in the length of the longest stretch of perfect
repeats in the nonfocal species. Furthermore, flanking-sequence length
difference was noted between the two species at 58% of the loci sequenced.
These data do not support the predictions of the directional-evolution
hypothesis; however, consistent with the ascertainment bias hypothesis, the
lower variability in nonfocal species is an artifact of the microsatellite
cloning and isolation process. Our results also suggest that the magnitude
of ascertainment bias for repeat unit length is a function of the
microsatellite size distribution in the genomes of different species.

Prospects for estimating nucleotide divergence with RAPDs 总被引:11,自引:0,他引:11
The technique of random amplification of polymorphic DNA (RAPD), which is
simply polymerase chain reaction (PCR) amplification of genomic DNA by a
single short oligonucleotide primer, produces complex patterns of anonymous
polymorphic DNA fragments. The information provided by these banding
patterns has proved to be of great utility for mapping and for verification
of identity of bacterial strains. Here we consider whether the degree of
similarity of the banding patterns can be used to estimate nucleotide
diversity and nucleotide divergence. With haploid data, fragments generated
by RAPD-PCR can be treated in a fashion very similar to that for
restriction-fragment data. Amplification of diploid samples, on the other
hand, requires consideration of the fact that presence of a band is
dominant to absence of the band. After describing a method for estimating
nucleotide divergence on the basis of diploid samples, we summarize the
restrictions and criteria that must be met when RAPD data are used for
estimating population genetic parameters.

Summary Formulas are presented to illustrate the calculation of correct variances of prediction error (PEV) and the correlation between true and predicted values (rTI) when the incorrect variance-covariance matrix for the random effects is used in mixed-model equations (MME). The example with progeny records of highly related and inbred sires showed that PEV were underestimated from the diagonals of the inverse of the coefficient matrix of the MME when sires were assumed unrelated and not inbred and were overestimated when relationships among sires were calculated with Henderson's simple rules for the inverse of the numerator relationship matrix, A-1, which do not consider inbreeding. When Quaas' rules for A-1, which do consider inbreeding, are used, the correct PEV are obtained. In the example, calculations of rTI from the diagonals of the inverse of the coefficient matrix were too large when relationships and inbreeding were ignored and were obviously wrong when the approximation to the numerator relationship matrix, A, was based on the simple rules for calculating A-1. If the correct A is used in the MME, the calculation of rTI may be incorrect if inbreeding of the evaluated individual is not considered. Adélie penguin (Pygoscelis adeliae) males and females, nesting in Antarctica, alternate attendance at the nest with absences of many days to forage at sea. We investigated the importance of tactile input from egg and chicks on prolactin levels by observing nest attendance patterns and obtaining blood samples (1) during the first nest exchange of the incubation stage, (2) from birds whose incubation period was artificially increased or decreased by about 10 days, and (3) from birds whose nests had failed. Prolactin levels in females after 8 to 11 days of absence from the breeding colony did not differ from those in incubating males and did not change after females resumed incubation. Moving eggs between nests resulted in nests in which chicks hatched after about 26, 36 (normal), or 46 days. Duration of incubation did not affect prolactin levels in the parents measured during incubation, at the pip stage, hatch stage, or early brood stage. Adults first left their chicks unguarded on about the same calendar date, regardless of chick age. However, chicks from long incubation nests averaged 8 days younger when they were left unguarded than chicks from control or short-incubation nests. In females, there was no effect of nest failure on prolactin levels. In males, prolactin levels were slightly lower after nest failure than in males tending nests. Testosterone was significantly higher in males after nest failure than in males still tending nests. Prolactin is elevated in Adélie penguins as part of the program of cyclical hormonal changes that accompany the lengthy reproductive season and is relatively independent of tactile input. Sustained prolactin secretion is probably required for the maintenance of parental behavior in offshore feeding species that must be absent from the nest for many days at a time. 相似文献
