A visual atlas of meiotic protein dynamics in living fission yeast

Meiosis is a carefully choreographed dynamic process that re-purposes proteins from somatic/vegetative cell division, as well as meiosis-specific factors, to carry out the differentiation and recombination pathway common to sexually reproducing eukaryotes. Studies of individual proteins from a variety of different experimental protocols can make it difficult to compare details between them. Using a consistent protocol in otherwise wild-type fission yeast cells, this report provides an atlas of dynamic protein behaviour of representative proteins at different stages during normal zygotic meiosis in fission yeast. This establishes common landmarks to facilitate comparison of different proteins and shows that initiation of S phase likely occurs prior to nuclear fusion/karyogamy.     

Isolation of melanin from human plasma lipofuscin

Human plasma lipofuscin and its melanin component were isolated and quantified. Electron paramagnetic resonance, infrared, ultraviolet and visible spectra of this melanin exhibited absorption characteristics very similar to those of known melanins. The human plasma lipofuscin contained approximately 85% protein, 3% melanin, 0.4% lipid and 0.25% mucoprotein constituents and emitted yellow-green fluorescence in 366-nm light. The ethanol-ether lipid extract obtained after acid hydrolysis from the lipid-melanin fraction of this lipofuscin was also found to fluoresce in yellow-green color in 366-nm light and produced similar fluorescence excitation and emission spectra as those of the human plasma lipofuscin in water solution. The isolated melanin component was not fluorescent.     

Studies on plant gums. Role of calcium in polysaccharide-protein interaction in the neem (Azadirachta indica) gum

The partial removal of tightly bound Ca²⁺ from dialysed neem (Azadirachta indica) gum, resulted in the release of a basic protein from a highly anionic polysaccharide-protein complex as evidenced by chromatographic studies on TEAE-cellulose. Complete removal of Ca²⁺ caused, in addition, the release of a minor heteropolysaccharide which was found in association with the basic protein. These processes were reversed on the addition of Ca²⁺. The gum, in addition, contained a protein-rich component accounting for 35% protein and 7.5% total carbohydrate. This component behaved as a distinct entity during ion-exchange chromatography of the native gum solutions, or which were either partially or completely depleted of bound Ca²⁺.     

Isolation of the proviral coding strand of avian myeloblastosis virus from leukemic chicken myeloblast by affinity chromatography.

The coding strand of the integrated proviral DNA of avian myeloblastosis virus (AMV) was isolated from the DNA of leukemic chicken myeloblast. The three-step isolation procedure employed a combination of affinity chromatography with Sepharose-linked RNA, nucleic acid hybridization, and hydroxypatite chromatography techniques. At each step of purification the product was analyzed for the enrichment of AMV coding strand by hybridization with AMV RNA. The final product was the coding strand of the AMV DNA (90% pure). These results show that such a procedure can be used for the isolation and analysis of a specific structural gens of eukaryotic cells.     

How dangerous are falls in old people at home?

From a survey in six general practices information was obtained on 125 people aged 65 and over who fell in their own homes. Three fractured their femurs and 15 had other fractures; most of the rest suffered only trivial injuries. Twenty lay on the floor for more than one hour; none were known to have suffered hypothermia. One-quarter of these patients died within one year of the fall, five times as many as in an age- and sex-matched control group; while of those who lay on the floor for more than one hour, half died within six months of the fall. Factors associated with mortality from falls were impaired mobility, abnormal balance, and a disturbed pattern of gait. Falls at home in old age are often indicative of the presence of severe ill health.

Enhancement of aflatoxin B1-induced hepatocarcinogenesis in rats by partial hepatectomy

The effect of enhanced cell replication induced by partial hepatectomy (PH) in aflatoxin B1 (AFB1)-induced hepatocarcinogenesis has been studied in rats of the inbred As2 strain. Animals were given 0.25 mg/kg body weight of AFB1 as a single intraperitoneal dose 24 h after PH. Non-hepatectomized animals given the same dose of AFB1 served as controls. Neoplastic nodules and hepatocellular carcinoma (HCC) were detected respectively in 100% and 90% of hepatectomized animals sacrificed between 55 and 65 weeks after AFB1 administration. None of the ten non-hepatectomized rats sacrificed at this time interval showed HCC or neoplastic nodules. On histochemical staining the tumour population was found to be heterogeneous. Thus PH resulted in enhancement of AFB1-induced hepatocarcinogenesis in rats of the AS2 strain.
