We report observations on the dynamics of bacterial communities in response to methane stimulus in laboratory microcosm incubations prepared with lake sediment samples. We first measured taxonomic compositions of long-term enrichment cultures and determined that, although dominated by Methylococcaceae types, these cultures also contained accompanying types belonging to a limited number of bacterial taxa, methylotrophs and non-methylotrophs. We then followed the short-term community dynamics, in two oxygen tension regimens (150 μM and 15 μM), observing rapid loss of species diversity. In all microcosms, a single type of Methylobacter represented the major methane-oxidizing partner. The accompanying members of the communities revealed different trajectories in response to different oxygen tensions, with Methylotenera species being the early responders to methane stimulus under both conditions. The communities in both conditions were convergent in terms of their assemblage, suggesting selection for specific taxa. Our results support prior observations from metagenomics on distribution of carbon from methane among diverse bacterial populations and further suggest that communities are likely responsible for methane cycling, rather than a single type of microbe.  相似文献

Aims

To investigate the feasibility and to compare three devices measuring intraocular pressure (IOP) in mucopolysaccharidosis patients (MPS): iCare rebound tonometer (RT), Perkins applanation tonometer (PAT) and ocular response analyzer (ORA)

Methods

MPS patients who underwent at least two examinations out of: RT, PAT and ORA at the same visit were identified and retrospectively analyzed in this study.

Results

17 patients fulfilled the inclusion criterion. In all 17 patients IOP measurements were performed with RT (34 eyes) and ORA (33 eyes), while PAT measurement was possible in only 12 (24 eyes) patients. The RT, corneal-compensated intraocular pressure (IOPcc) and Goldmann-correlated intraocular pressure (IOPg) differed relevantly from IOP assessed with PAT. Corneal clouding in MPS patients correlated positively with PAT, RT and IOPg (r = 0.3, 0.5, and 0.5 respectively), but not with IOPcc (r = 0.07). The MPS-related corneal clouding correlated positively with biomechanical corneal parameters assessed with ORA: corneal hysteresis (r = 0.77) and corneal resistance factor (r = 0.77) either.

Conclusions

RT and ORA measurements were tolerated better than applanation tonometry in MPS patients. IOP measurements assessed with RT and ORA differed relevantly from PAT. Corneal-compensated IOP assessed with ORA seems to be less affected by the MPS-related corneal clouding than applanation or rebound tonometry. RT and ORA measurements should be preferred for IOP assessment in patients with MPS.  相似文献

The role of abscisic acid (ABA) during early development was investigated in tomato seedlings. The endogenous content of ABA in particular organs was analyzed in seedlings grown in the dark and under blue light. Our results showed that in dark-grown seedlings, the ABA accumulation was maximal in the cotyledons and elongation zone of hypocotyl, whereas under blue-light, the ABA content was distinctly reduced. Our data are consistent with the conclusion that ABA promotes the growth of etiolated seedlings and the results suggest that ABA plays an inhibitory role in de-etiolation and photomorphogenesis in tomato.  相似文献

Populations of ectothermic vertebrates are vulnerable to environmental pollution and climate change because certain chemicals and extreme temperatures can cause sex reversal during early ontogeny (i.e. genetically female individuals develop male phenotype or vice versa), which may distort population sex ratios. However, we have troublingly little information on sex reversals in natural populations, due to unavailability of genetic sex markers. Here, we developed a genetic sexing method based on sex‐linked single nucleotide polymorphism loci to study the prevalence and fitness consequences of sex reversal in agile frogs (Rana dalmatina). Out of 125 juveniles raised in laboratory without exposure to sex‐reversing stimuli, 6 showed male phenotype but female genotype according to our markers. These individuals exhibited several signs of poor physiological condition, suggesting stress‐induced sex reversal and inferior fitness prospects. Among 162 adults from 11 wild populations in North‐Central Hungary, 20% of phenotypic males had female genotype according to our markers. These individuals occurred more frequently in areas of anthropogenic land use; this association was attributable to agriculture and less strongly to urban land use. Female‐to‐male sex‐reversed adults had similar body mass as normal males. We recorded no events of male‐to‐female sex reversal either in the laboratory or in the wild. These results support recent suspicions that sex reversal is widespread in nature, and suggest that human‐induced environmental changes may contribute to its pervasiveness. Furthermore, our findings indicate that sex reversal is associated with stress and poor health in early life, but sex‐reversed individuals surviving to adulthood may participate in breeding.  相似文献

Because seawater is hyperosmotic relative to body fluids of most vertebrates, marine lifestyle is expected to strongly influence the physiology of marine tetrapods. Regulating the salt content of body fluids is energetically costly; and osmoregulatory organs may not totally overcome salt load and/or water loss. As a consequence, marine lifestyle should influence physiological systems involved in the maintenance of the physiological balance (homeostasis), in the mobilisation of energetic resources (e.g., to fuel salt excretion), or in the acquisition of resources (e.g., fresh water). Corticosterone (CORT) is one such ‘generalist’ mediator that is linked with energy expenditure, physiological stress and that activates osmoregulation. As a consequence, CORT is expected to be overall higher in marine tetrapods but this hypothesis has never been tested. Using comparative analyses, we tested this hypothesis in birds, a lineage for which available data on baseline CORT allow comparing marine versus terrestrial species, and species with or without salt glands. We found that marine species (and species with salt glands) display significantly higher baseline CORT during the wintering (but not the breeding) stage. Although salt glands’ presence was tightly linked to phylogeny, our results suggest that marine lifestyle may impose a strong, but overlooked, influence on the allostasis‐related physiology of marine birds. Such habitat‐related variation in physiology is a major phenomenon to explore owing to its general implications for understanding the physiological basis of evolutionary transitions in habitat use. © 2015 The Linnean Society of London, Biological Journal of the Linnean Society, 2015, 115 , 154–161.  相似文献

In group‐living animals, individuals may benefit from the presence of an innovative group‐mate because new resources made available by innovators can be exploited, for example by scrounging or social learning. As a consequence, it may pay off to take the group‐mates' problem‐solving abilities into account in social interactions such as aggression or spatial association, for example because dominance over an innovative group‐mate can increase scrounging success, while spatial proximity may increase the chance of both direct exploitation and social learning. In this study, we tested whether the individuals' innovation success influences their social interactions with group‐mates in small captive flocks of house sparrows (Passer domesticus). First, we measured the birds' actual problem‐solving success in individual food‐extracting tasks. Then, we manipulated their apparent problem‐solving success in one task (by allowing or not allowing them to open a feeder repeatedly) while a new, unfamiliar group‐member (focal individual) had the opportunity to witness their performance. After this manipulation, we observed the frequency and intensity of aggression and the frequency of spatial associations between the focal individuals and their manipulated flock‐mates. Although flock‐mates behaved according to their treatments during manipulations, their apparent problem‐solving success did not affect significantly the focal individuals' agonistic behaviour or spatial associations. These results do not support that sparrows take flock‐mates' problem‐solving abilities into account during social interactions. However, focal individuals attacked those flock‐mates more frequently that had higher actual problem‐solving success (not witnessed directly by the focal individuals), although aggression intensity and spatial association by the focal birds were unrelated to the flock‐mates' actual success. If this association between flock‐mates' actual innovativeness and focal individuals' aggression is not due to confounding effects, it may imply that house sparrows can use more subtle cues to assess the group‐mates' problem‐solving ability than direct observation of their performance in simple foraging tasks.  相似文献
