Novel whole-cell antibiotic biosensors for compound discovery   总被引:1,自引:0,他引:1  
Cells containing reporters which are specifically induced via selected promoters are used in pharmaceutical drug discovery and in environmental biology. They are used in screening for novel drug candidates and in the detection of bioactive compounds in environmental samples. In this study, we generated and validated a set of five Bacillus subtilis promoters fused to the firefly luciferase reporter gene suitable for cell-based screening, enabling the as yet most-comprehensive high-throughput diagnosis of antibiotic interference in the major biosynthetic pathways of bacteria: the biosynthesis of DNA by the yorB promoter, of RNA by the yvgS promoter, of proteins by the yheI promoter, of the cell wall by the ypuA promoter, and of fatty acids by the fabHB promoter. The reporter cells mainly represent novel antibiotic biosensors compatible with high-throughput screening. We validated the strains by developing screens with a set of 14,000 pure natural products, representing a source of highly diverse chemical entities, many of them with antibiotic activity (6% with anti-Bacillus subtilis activity of 相似文献

Experimental exploration of equipment for stereotactic functional neurosurgery based on heating induced by radio-frequency current is most often carried out prior to surgery in order to secure a correct function of the equipment. The experiments are normally conducted in an experimental model including an albumin solution in which the treatment electrode is submerged, followed by a heating session during which a protein clot is generated around the electrode tip. The clot is believed to reflect the lesion generated in the brain during treatment. It is thereby presupposed that both the thermal and electric properties of the model are similar to brain tissue. This study investigates the presence of convective movements in the albumin solution using laser Doppler velocimetry. The result clearly shows that convective movements that depend on the time dependent heating characteristics of the equipment arise in the solution upon heating. The convective movements detected show a clear discrepancy compared with the in vivo situation that the experimental model tries to mimic; both the velocity (maximum velocity of about 5 mms) and mass flux are greater in this experimental setting. Furthermore the flow geometry is completely different since only a small fraction of the tissue surrounding the electrode in vivo consists of moving blood, whereas the entire surrounding given by the albumin solution in the experimental model is moving. Earlier investigations by our group (Eriksson et al., 1999, Med. Biol. Eng. Comput. 37, pp. 737-741; Wren, 2001, Ph.D. thesis; and Wren et al., 2001, Med. Biol. Eng. Comput. 39, pp. 255-262) indicate that the heat flux is an essential parameter for the lesion growth and final size, and that presence of convective movements in the model might substantially increase the heat flux. Thus, convective movements of the magnitude presented here will very likely underestimate the size of the brain lesion, a finding that definitely should be taken into consideration when using the model prior to patient treatment.  相似文献

Theoretical efforts suggest that the relative sizes of predators and their prey can shape community dynamics, the structure of food webs, and the evolution of life histories. However, much of this work has assumed static predator and prey body sizes. The timing of recruitment and the growth patterns of both predator and prey have the potential to modify the strength of predator–prey interactions. In this study, I examined how predator size dynamics in 40 temporary ponds over a 3-year period affected the survival of spotted salamander (Ambystoma maculatum) larvae. Across communities, gape-limited predator richness, but not size, was correlated with habitat duration (pond permanence). Within communities, mean gape-limited predator size diminished as the growing season progressed. This size reduction occurred because prey individuals grew into a body size refuge and because the largest of the predators left ponds by mid-season. Elevated gape-limited predation risk across time and space was predicted by the occurrence of two large predatory salamanders: marbled salamander larvae (Ambystoma opacum) and red-spotted newt adults (Notophthalmus viridescens). The presence of the largest gape-limited predator, A. opacum, predicted A. maculatum larval survival in the field. The distribution of large predatory salamanders among ponds and across time is expected to lead to differing community dynamics and to generate divergent natural selection on early growth and body size in A. maculatum. In general, a dynamic perspective on predator size often will be necessary to understand the ecology and evolution of species interactions. This will be especially true in frequently disturbed or seasonal habitats where phenology and ontogeny interact to determine body size asymmetries. Islet amyloid polypeptide (IAPP) is postulated to act as a hormonal signal from the pancreas to the brain to inhibit food intake and reduce adipose energy reserves. The present study compared the effects of chronic peripheral and chronic central administration of IAPP on food intake and meal pattern in rats. IAPP was administered subcutaneously (SC) for 7 days at doses of 0, 0.25, 2.5 and 25 pmol kg(-1) min(-1) using an osmotic minipump or administered centrally at doses of 0, 0.025, 0.25 and 2.5 pmol kg(-1) min(-1) using an osmotic minipump connected to an intracerebroventricular (ICV) catheter inserted into the third ventricle. Both SC and ICV infusion decreased total food intake dose-dependently. The minimal effective dose was 2.5 pmol IAPP kg(-1) min(-1) for SC administration and 0.25 pmol kg(-1) min(-1) for ICV infusion. The decrease in food intake produced by infusion of IAPP was mainly due to decreased meal size, although a significant decrease in meal number also occurred at the highest SC and ICV doses. SC administration produced a larger, more persistent decrease in food intake during the light period than in the dark period, while ICV infusion caused a larger, more persistent decrease during the dark period. The 10-fold difference in minimal effective doses indicates that ICV-administered IAPP acted primarily in the brain to inhibit food intake. The difference between the effects of IAPP on meal pattern with the two methods of administration suggests that IAPP does not act on the same target(s) when administered centrally as it does when it is administered peripherally.  相似文献

Temperature responses of carbon assimilation processes were studied in four dominant species from mountain grassland ecosystem, i.e. Holcus mollis (L.), Hypericum maculatum (Cr.), Festuca rubra (L.), and Nardus stricta (L.), using the gas exchange technique. Leaf temperature (T L) of all species was adjusted within the range 13–30 °C using the Peltier thermoelectric cooler. The temperature responses of metabolic processes were subsequently modelled using the Arrhenius exponential function involving the temperature coefficient Q 10. The expected increase of global temperature led to a significant increase of dark respiration rate (R D; Q 10 = 2.0±0.5), maximum carboxylation rate (V Cmax; Q 10 = 2.2±0.6), and maximum electron transport rate (J max; Q 10 = 1.6±0.4) in dominant species of mountain grassland ecosystems. Contrariwise, the ratio between J max and V Cmax linearly decreased with T L [y = −0.884 T L + 5.24; r 2 = 0.78]. Hence temperature did not control the ratio between intercellular and ambient CO2 concentration, apparent quantum efficiency, and photon-saturated CO2 assimilation rate (P max). P max primarily correlated with maximum stomatal conductance irrespective of T L. Water use efficiency tended to decrease with T L [y = −0.21 T L + 8.1; r 2 = 0.87].  相似文献

Question: Is ombrotrophic bog vegetation in an oceanic region of southwestern Sweden changing in the same direction over a five year period (1999 ‐ 2004) as northwest European bogs in the last 50 years, i.e. towards drier and more eutrophic vegetation? Location: The province of Halland, southwestern Sweden. Methods: Changes in species composition were monitored in 750 permanently marked plots in 25 ombrotrophic bogs from 1999 to 2004. Changes in species occurrences and richness were analysed and a multivariate statistical method (DCA) was used to analyse vegetation changes. Results: The species composition changed towards wetter rather than drier conditions, which is unlike the general pattern of vegetation change on bogs in northwestern Europe. Species typical of wetter site conditions including most Sphagnum species increased in abundance on the bogs until 2004. The total number of species per plot increased, mostly due to the increased species richness of Sphagnum species. Nitrogen‐demanding (eutrophic) species increased in occurrence. Conclusions: Ombrotrophic bog vegetation in an oceanic region in Sweden became wetter and was resilient to short‐term climatic shifts, after three years of below normal precipitation followed by several years with normal precipitation levels. Shifts towards more nitrogen demanding species were rapid in this region where the deposition levels have been high for several decades.  相似文献

Pathogenicity of the gram-negative plant pathogen Xanthomonas campestris pv. vesicatoria depends on a type III secretion system, which translocates bacterial effector proteins into the plant cell. In this study, we identified two novel type III effectors, XopE1 and XopE2 (Xanthomonas outer proteins), using the AvrBs3 effector domain as reporter. XopE1 and XopE2 belong to the HopX family and possess a conserved putative N-myristoylation motif that is also present in the effector XopJ from X. campestris pv. vesicatoria 85-10. XopJ is a member of the YopJ/AvrRxv family of acetyltransferases. Confocal laser scanning microscopy and immunocytochemistry revealed that green fluorescent protein fusions of XopE1, XopE2, and XopJ localized to the plant cell plasma membrane. Targeting to the membrane is probably due to N-myristoylation, because a point mutation in the putative myristoylated glycine residue G2 in XopE1, XopE2, and XopJ resulted in cytoplasmic localization of the mutant proteins. Results of hydroxylamine treatments of XopE2 protein extracts suggest that the proteins are additionally anchored in the host cell plasma membrane by palmitoylation. The membrane localization of the effectors strongly influences the phenotypes they trigger in the plant. Agrobacterium-mediated expression of xopE1 and xopJ in Nicotiana benthamiana led to cell-death reactions that, for xopJ, were dependent on the N-myristoylation motif. In the case of xopE1(G2A), cell death was more pronounced with the mutant than with the wild-type protein. In addition, XopE2 has an avirulence activity in Solanum pseudocapsicum.  相似文献

Important insights have recently been gained in our understanding of how host immune responses mediate resistance to parasitic helminths and control associated pathological responses. Although similar cells and cytokines are evoked in response to infection by helminths as diverse as nematodes and schistosomes, the components of the response that mediate protection are dependent on the particular parasite. In this Review, we examine recent findings regarding the mechanisms of protection in helminth infections that have been elucidated in murine models and discuss the implications of these findings in terms of future therapies.  相似文献

Question: Is the successional transition from pine to hardwood, which has been inferred from chronosequence plots in previous studies, validated through a time line of satellite imagery? Location: Durham, North Carolina, USA. Methods: We examined successional trends in a time‐series of winter‐summer pairs of Thematic Mapper imagery from 1986 to 2000. We calculated the normalized difference of vegetation index (NDVI) for winter and summer, as well as the difference between summer and winter NDVI (i.e., summer increment NDVI). A set of approximately 50 forest stands of known age and phenology were used to interpret patterns in winter and summer increment NDVI over successional time, and a continuum was found to exist between pine‐dominance and hardwood‐dominance. We fitted a series of linear regressions that modeled the change in winter and summer increment NDVI as a function of initial winter and summer increment NDVI, and additional explanatory variables. Results: All regressions were highly significant (P < 0.0001, R2= ca. 0.3). Predicted dynamics are in accord with successional theory, with pixels moving from evergreen dominance to deciduous dominance along a line of fairly constant summer NDVI. A large disturbance event that occurred over the course of this study, Hurricane Fran, appeared to slow rates of succession in the short term (1–3 years), but increase the rate of conversion to hardwoods over longer time spans. Conclusions: We conclude that temporal sequences of remote sensing images provide an excellent opportunity for broad‐scale monitoring of successional processes, and that continuous metrics of that change are essential to accurate monitoring.  相似文献
