In vivo leukocyte recruitment is not fully understood and may result from interactions of chemokines with glycosaminoglycans/GAGs. We previously showed that chlorite-oxidized oxyamylose/COAM binds the neutrophil chemokine GCP-2/CXCL6. Here, mouse chemokine binding by COAM was studied systematically and binding affinities of chemokines to COAM versus GAGs were compared. COAM and heparan sulphate bound the mouse CXC chemokines KC/CXCL1, MIP-2/CXCL2, IP-10/CXCL10 and I-TAC/CXCL11 and the CC chemokine RANTES/CCL5 with affinities in the nanomolar range, whereas no binding interactions were observed for mouse MCP-1/CCL2, MIP-1α/CCL3 and MIP-1β/CCL4. The affinities of COAM-interacting chemokines were similar to or higher than those observed for heparan sulphate. Although COAM did not display chemotactic activity by itself, its co-administration with mouse GCP-2/CXCL6 and MIP-2/CXCL2 or its binding of endogenous chemokines resulted in fast and cooperative peritoneal neutrophil recruitment and in extravasation into the cremaster muscle in
vivo. These local GAG mimetic features by COAM within tissues superseded systemic effects and were sufficient and applicable to reduce LPS-induced liver-specific neutrophil recruitment and activation. COAM mimics glycosaminoglycans and is a nontoxic probe for the study of leukocyte recruitment and inflammation in

Neonatal brain hypoxic ischemia (HI) often results in long-term motor and cognitive impairments. Post-ischemic inflammation greatly effects outcome and adenosine receptor signaling modulates both HI and immune cell function. Here, we investigated the influence of adenosine A1 receptor deficiency (A1R?/?) on key immune cell populations in a neonatal brain HI model. Ten-day-old mice were subjected to HI. Functional outcome was assessed by open locomotion and beam walking test and infarction size evaluated. Flow cytometry was performed on brain-infiltrating cells, and semi-automated analysis of flow cytometric data was applied. A1R?/? mice displayed larger infarctions (+33 %, p?<?0.05) and performed worse in beam walking tests (44 % more mistakes, p?<?0.05) than wild-type (WT) mice. Myeloid cell activation after injury was enhanced in A1R?/? versus WT brains. Activated B lymphocytes expressing IL-10 infiltrated the brain after HI in WT, but were less activated and did not increase in relative frequency in A1R?/?. Also, A1R?/? B lymphocytes expressed less IL-10 than their WT counterparts, the A1R antagonist DPCPX decreased IL-10 expression whereas the A1R agonist CPA increased it. CD4+ T lymphocytes including FoxP3+ T regulatory cells, were unaffected by genotype, whereas CD8+ T lymphocyte responses were smaller in A1R?/? mice. Using PCA to characterize the immune profile, we could discriminate the A1R?/? and WT genotypes as well as sham operated from HI-subjected animals. We conclude that A1R signaling modulates IL-10 expression by immune cells, influences the activation of these cells in vivo, and affects outcome after HI. 相似文献

The ascomycetous yeast Pichia anomala is frequently associated with food and feed products, either as a production organism or as a spoilage yeast. It belongs to the nonSaccharomyces wine yeasts and contributes to the wine aroma by the production of volatile compounds. The ability to grow in preserved food and feed environments is due to its capacity to grow under low pH, high osmotic pressure and low oxygen tension. A new application of P. anomala is its use as a biocontrol agent, which is based on the potential to inhibit a variety of moulds in different environments. Although classified as a biosafety class-1 organism, cases of P. anomala infections have been reported in immunocompromised patients. On the other hand, P. anomala killer toxins have a potential as antimicrobial agents. The yeast can use a broad range of nitrogen and phosphor sources, which makes it a potential agent to decrease environmental pollution by organic residues from agriculture. However, present knowledge of the physiological basis of its performance is limited. Through its involvement in inflammation, opsonization, and cytolysis, the complement protects against infectious agents. Although most of the complement proteins are synthesized in the central nervous system (CNS), the role of the complement system in the normal or ischemic CNS remains unclear. Here we demonstrate for the first time that neural progenitor cells and immature neurons express receptors for complement fragments C3a and C5a (C3a receptor (C3aR) and C5a receptor). Mice that are deficient in complement factor C3 (C3(-/-)) lack C3a and are unable to generate C5a through proteolytic cleavage of C5 by C5-convertase. Intriguingly, basal neurogenesis is decreased both in C3(-/-) mice and in mice lacking C3aR or mice treated with a C3aR antagonist. The C3(-/-) mice had impaired ischemia-induced neurogenesis both in the subventricular zone, the main source of neural progenitor cells in adult brain, and in the ischemic region, despite normal proliferative response and larger infarct volumes. Thus, in the adult mammalian CNS, complement activation products promote both basal and ischemia-induced neurogenesis. 相似文献

The decolourization of a mixture of 200 mg L−1 each of Reactive Black 5 and Reactive Red 2 dye was studied in batch experiments using microorganisms growing on forest residue wood chips in combination with or without added white-rot fungus, Bjerkandera sp. BOL 13. The study was performed as a first stage in the development of a relatively simple treatment process for textile wastewater, designed to work in developing countries. Forest residue wood chips contain a mixture of fungi and bacteria which is an advantage when complex molecules should be degraded. The wood chips furthermore provide the microorganisms with carbon source which make the addition of e.g. glucose unnecessary. The results showed that the microorganisms growing on the forest residue wood chips decolourized the mixture of the two dyes; adding extra nutrients approximately doubled the decolourization rate. The time needed for decolourization was approximately 18 days when nutrients were added. Lignocellulosic material is complex and so were the analysis, microorganisms were therefore transferred to ordinary soft wood chips from forest residue wood chips. The aim was to define a primary culture system enriched in neurons using a defined culture medium, and characterize the model
system as to cellular morphology and neuronal phenotypes. We found that these primary neuron enriched cultures from either
newborn rat cerebral cortex or hippocampus contain small GABAergic and large glutamatergic neurons as well as astrocytes and
microglia. Astrocytes in these cultures are morphologically differentiated with long, slender processes and interact with
soluble factors responsible for induction and expression of the glutamate transporter GLT-1. The cultures achieve the highest
expression of the vesicular glutamate transporter 1 (VGLUT1) and GLT-1 after 20 days in vitro. Conditioned media from these
neuron enriched cultures also induced GLT-1 expression in primary astrocytic cultures, which were free from neurons. The amount
of glutamatergic neurons guides the morphological maturation of astrocytes and GLT-1 expression both in the neuron enriched
cultures and in the conditioned media supplemented astrocytic cultures. Interestingly, these cultures were not influenced
or activated by the inflammatory stimulus lipopolysaccharide. This suggests that soluble factors from neurons protect microglia
and astrocytes to become inflammatory reactive. In conclusion we have developed a well characterized culture model system
enriched in neurons, taken from newborn rats and cultured in defined media. The neurons express different neuronal phenotypes.
Such a model system is valuable when studying interactions between neurons and glial cells. 相似文献

Abstract : The influence of β‐amyloid on cholinergic neurotransmission was studied by measuring alterations in nicotinic acetylcholine receptors (nAChRs) in autopsy brain tissue from subjects carrying the Swedish amyloid precursor protein (APP) 670/671 mutation. Significant reductions in numbers of nAChRs were observed in various cortical regions of the Swedish 670/671 APP mutation family subjects (‐73 to ‐87%) as well as in sporadic Alzheimer's disease (AD) cases (‐37 to ‐57%) using the nicotinic agonists [3H]epibatidine and [3H]nicotine, which bind with high affinity to both α3 and α4 and to α4 nAChR subtypes, respectively. Saturation binding studies with [3epibatidine revealed two binding sites in the parietal cortex of AD subjects and controls. A significant decrease in Bmax (‐82%) for the high‐affinity site was observed in APP 670/671 subjects with no change in KD compared with controls (0.018 nM APP 670/671 ; 0.036 nM control). The highest load of neuronal plaques (NPs) was observed in the parietal cortex of APP 670/671 brains, whereas the number of [3H]nicotine binding sites was less impaired compared with other cortical brain regions. Except for a positive significant correlation between the number of [3H]nicotine binding sites and number of NPs in the parietal cortex, no strict correlation was observed between nAChR deficits and the presence of NPs and neurofibrillary tangles, suggesting that these different processes may be closely related but not strictly dependent on each other. 相似文献

The ability of warm-grown leaves to acclimate their photosynthetic machinery to low, non-freezing temperature was compared for contrasting species of grasses and trees. All trees ( Betula pubescens , Salix sp. and Picea abies ), and young plants of one of the grasses ( Hordeum vulgare ) showed acclimation of photosynthesis while the other two grasses ( Phalaris arundinacea and Festuca ovina ) did not. It was those species that maintained leaf sugar concentrations essentially unchanged that showed acclimation. Trees maintained leaf sugar concentrations essentially unchanged by effectively converting leaf sugar surpluses into storage compounds. Grasses were, by comparison, less effective. However, very young plants of Hordeum maintained leaf sugar concentrations unchanged by continued growth rather than by increased storage. This diversity of low-temperature responses are discussed in relation to possible different priorities of trees and grasses: for grasses to undergo cold hardening by allowing sugars to rise, and for trees to store sugars to allow photosynthesis to operate independently of growth as growth varies with growth rhythm and air temperature. 相似文献
