Summary Long-term primary cultures of epithelial cells from rainbow trout (Oncorhynchus mykiss) liver have been established. Nearly homogenous (>97%) populations of hepatocytes were placed into primary culture and remained viable and proliferative for at least 70 d. In addition to hepatocytes, proliferative biliary cells persisted in the cultures for at least 30 d. Finally, a third type of epithelial cell, which we have termed a “spindle cell,” consistently appeared and proliferated to confluence in these cultures. The confluent cultures of spindle cells were successfully subcultured and passaged. The initial behavior, growth, and optimization of serum and media requirements for these cells is described. All three cell types proliferated as measured by thymidine incorporation, autoradiography, proliferating cellular nuclear antigen analysis, and propidium iodine staining. Further efforts to characterize the cells included western blotting and immunohistochemical staining with antibodies to cytokeratins previously reported in fish liver. From these data, it appears that all three cell populations are epithelial in nature. Furthermore, significant changes in actin organization, often indicative of transformation or pluripotent cells, were observed with increased time in primary culture.  相似文献

Summary In the cells of the zona fasciculata of intact adult rats lysosomes can be found in medium amounts, measuring 0.2–0.4 in diameter. After treatment with ACTH, the lysosomes in the zona fasciculata of the intact animals increase in number and size, and lysosome-lipid conglomerates are formed. In hypophysectomized animals, the lysosomes in the zona fasciculata decrease in number and size in proportion to the time elapsed. Treatment with ACTH given 2 to 4 weeks after hypophysectomy, resulted in the increase in number and size of the lysosomes. Under such circumstances lysosomes appear in conspicuously large numbers around lipid droplets, which frequently coalesce with them. The possible role of lysosomes is discussed.  相似文献

Plant-soil feedbacks are an important aspect of invasive species success. One type of feedback is alteration of soil nutrient cycling. Cheatgrass invasion in the western USA is associated with increases in plant-available nitrogen (N), but the mechanism for this has not been elucidated. We labeled cheatgrass and crested wheatgrass, a common perennial grass in western rangelands, with 15N-urea to determine if differences in root exudates and turnover could be a mechanism for increases in soil N. Mesocosms containing plants were either kept moist, or dried out during the final 10 days to determine the role of senescence in root N release. Soil N transformation rates were determined using 15N pool dilution. After 75 days of growth, cheatgrass accumulated 30 % more total soil N and organic carbon than crested wheatgrass. Cheatgrass roots released twice as much N as crested wheatgrass roots (0.11 vs. 0.05 mg N kg?1 soil day?1) in both soil moisture treatments. This occurred despite lower root abundance (7.0 vs. 17.3 g dry root kg?1 soil) and N concentration (6.0 vs. 7.6 g N kg?1 root) in cheatgrass vs. crested wheatgrass. We propose that increases in soil N pool sizes and transformation rates under cheatgrass are caused by higher rates of root exudation or release of organic matter containing relatively large amounts of labile N. Our results provide the first evidence for the underlying mechanism by which the invasive annual cheatgrass increases N availability and establishes positive plant-soil feedbacks that promote its success in western rangelands.  相似文献

Archaea are characterised by a complex metabolism with many unique enzymes that differ from their bacterial and eukaryotic counterparts. The thermoacidophilic archaeon Sulfolobus solfataricus is known for its metabolic versatility and is able to utilize a great variety of different carbon sources. However, the underlying degradation pathways and their regulation are often unknown. In this work, the growth on different carbon sources was analysed, using an integrated systems biology approach. The comparison of growth on L‐fucose and D‐glucose allows first insights into the genome‐wide changes in response to the two carbon sources and revealed a new pathway for L‐fucose degradation in S. solfataricus. During growth on L‐fucose major changes in the central carbon metabolic network, as well as an increased activity of the glyoxylate bypass and the 3‐hydroxypropionate/4‐hydroxybutyrate cycle were observed. Within the newly discovered pathway for L‐fucose degradation the following key reactions were identified: (i) L‐fucose oxidation to L‐fuconate via a dehydrogenase, (ii) dehydration to 2‐keto‐3‐deoxy‐L‐fuconate via dehydratase, (iii) 2‐keto‐3‐deoxy‐L‐fuconate cleavage to pyruvate and L‐lactaldehyde via aldolase and (iv) L‐lactaldehyde conversion to L‐lactate via aldehyde dehydrogenase. This pathway as well as L‐fucose transport shows interesting overlaps to the D‐arabinose pathway, representing another example for pathway promiscuity in Sulfolobus species.  相似文献

A series of 1- and 2-naphthyloxy derivatives were synthesized and evaluated for histamine H3 receptor affinity. Most compounds showed high affinities with Ki values below 100?nM. The most potent ligand, 1-(5-(naphthalen-1-yloxy)pentyl)azepane (11) displayed high affinity for the histamine H3 receptor with a Ki value of 21.9?nM. The antagonist behaviour of 11 was confirmed both in vitro in the cAMP assay (IC50?=?312?nM) and in vivo in the rat dipsogenia model (ED50?=?3.68?nM). Moreover, compound 11 showed positive effects on scopolamine induced-memory deficits in mice (at doses of 10 and 15?mg/kg) and an analgesic effect in the formalin test in mice with ED50?=?30.6?mg/kg (early phase) and ED50?=?20.8?mg/kg (late phase). Another interesting compound, 1-(5-(Naphthalen-1-yloxy)pentyl)piperidine (13; H3R Ki?=?53.9?nM), was accepted for Anticonvulsant Screening Program at the National Institute of Neurological Disorders and Stroke/National Institute of Health (Rockville, USA). The screening was performed in the maximal electroshock seizure (MES), the subcutaneous pentylenetetrazole (scPTZ) and the 6-Hz psychomotor animal models of epilepsy. Neurologic deficit was evaluated by the rotarod test. Compound 13 inhibited convulsions induced by the MES with ED50 of 19.2?mg/kg (mice, i.p.), 17.8 (rats, i.p.), and 78.1 (rats, p.o.). Moreover, 13 displayed protection against the 6-Hz psychomotor seizures (32?mA) in mice (i.p.) with ED50 of 33.1?mg/kg and (44?mA) ED50 of 57.2?mg/kg.Furthermore, compounds 11 and 13 showed in vitro weak influence on viability of tested cell lines (normal HEK293, neuroblastoma IMR-32, hepatoma HEPG2), weak inhibition of CYP3A4 activity, and no mutagenicity. Thus, these compounds may be used as leads in a further search for histamine H3 receptor ligands with promising in vitro and in vivo activity.  相似文献

A Trichoderma mixture was applied to kiwifruit plants in a pot experiment and to mature orchard vines to determine its effect on numbers of root-associated fungi and compare application methods into kiwifruit orchards in New Zealand. Trichoderma and other fungal numbers were recorded using a root plating technique. In the pot experiment, a significantly greater number of Trichoderma was isolated from treated compared to untreated plants 6 and 10 weeks after inoculation. Six months after application, greater numbers of Trichoderma were recovered in the orchards from Trichoderma treated vines and the increase was comparable for all application methods with over 50% of isolated fungi identified as Trichoderma compared to 33% in the water-only control. Different viable methods are available that can easily be integrated into everyday orchard management in order to introduce Trichoderma into mature kiwifruit stands. A six-monthly or more frequent application schedule seems advisable.  相似文献

Chest wall motion of infants during spinal anesthesia   总被引:1,自引:0,他引:1  
To test the extent to which diaphragmatic contraction moves the rib cage in awake supine infants during quiet breathing, we studied chest wall motion in seven prematurely born infants before and during spinal anesthesia for inguinal hernia repair. Infants were studied at or around term (postconceptional age 43 +/- 8 wk). Spinal anesthesia produced a sensory block at the T2-T4 level, with concomitant motor block at a slightly lower level. This resulted in the loss of most intercostal muscle activity, whereas diaphragmatic function was preserved. Rib cage and abdominal displacements were measured with respiratory inductance plethysmography before and during spinal anesthesia. During the anesthetic, outward inspiratory rib cage motion decreased in six infants (P less than 0.02, paired t test); four of these developed paradoxical inward movement of the rib cage during inspiration. One infant, the most immature in the group, had inward movement of the rib cage both before and during the anesthetic. Abdominal displacements increased during spinal anesthesia in six of seven infants (P less than 0.05), suggesting an increase in diaphragmatic motion. We conclude that, in the group of infants studied, outward rib cage movement during awake tidal breathing requires active, coordinated intercostal muscle activity that is suppressed by spinal anesthesia.  相似文献
