Glial fibrillary acidic protein (GFAP) is a commonly used marker to identify enteric glia in the mammalian gut. Little is however known about enteric glia in other vertebrates. The aim of the present study was to examine the distribution of GFAP immunoreactivity in adult and developing fish. In adult shorthorn sculpin (Myoxocephalus scorpius) and zebrafish (Danio rerio), GFAP immunoreactivity was seen in the myenteric plexus in all regions of the gut. Co-staining for the neuronal markers Hu C/D and acetylated tubulin showed that GFAP immunoreactivity was not associated with nerves. GFAP immunoreactivity was predominantly seen in processes with few glial cell bodies being demonstrated in adult fish. GFAP immunoreactivity was also found in the gut in larval zebrafish from 3 days post-fertilisation, i.e. at approximately the same time that differentiated enteric nerve cells first occur. Immunoreactivity was most prominent in areas with no or a low density of Hu-immunoreactive nerve cell bodies, indicating that the developing glia follows a different pattern from that of enteric neurons. The results suggest that GFAP can be used as a marker for enteric glia in fish, as in birds and mammals. The distribution of GFAP immunoreactivity implies that enteric glia are widespread in the fish gastrointestinal tract. Glia and neurons diverge early during development of the gastrointestinal tract.  相似文献

Specific binding of collagen type IV to Streptococcus pyogenes   总被引:5,自引:0,他引:5  
Many strains of Streptococcus pyogenes are capable of binding type IV collagen. In the present study, all 50 S. pyogenes strains isolated from patients with acute glomerulonephritis showed high or moderate affinity for radiolabelled type IV collagen. A majority of strains of other sources, such as reference strains of various M-types and strains isolated from patients with pharyngeal infections also bound type IV collagen; however, a number of weak binders or non-binders were found among those. The collagen type IV binding component(s) on S. pyogenes were susceptible to proteinase K digestion, partially sensitive to trypsin but insensitive to pepsin treatment at pH 5.5. According to tests with three M-positive strains and their M-negative derivatives, the binding was not dependent on M-protein. The binding was saturable with time and inhibited by unlabelled type IV collagen. Partially inhibition was found with type II collagen, gelatin and fibrinogen but not with a number of other serum proteins.  相似文献

During the spring of 1996 we occupied a station on annual sea ice located several kilometers from Disko Island, West Greenland in water depths greater than 200 m. The goal of this 3-week field season was to characterize sea-ice communities and the underlying water column prior to, and during, ice break-up. A heavier than usual snow load depressed the sea ice below sea level and the snow-ice interface became flooded. Some of this flooded region subsequently refroze and the whole process repeated itself when additional snow accumulated. The infiltration phytoplankton and protozooplankton assemblages that developed in this region were abundant and diverse. Algal biomass in the infiltration layer was approximately an order of magnitude greater than in the underlying water column but an order of magnitude less than in the well-developed bottom ice community. The infiltration autotrophic assemblage resembled the bottom-ice assemblage while the protozooplankton assemblage was more similar to the water column assemblage. The variability in plankton community structure was studiedin Disko Bay and across important fishing banks off the westcoast of Greenland. The primary goal of the study was to investigatepossible linkages between hydrographical processes and planktonstructures, hypothesizing that hydrographic fronts would bepresent in the area, and that these to a large extent determineplankton distribution, composition and productivity. We sampledalong four cross-shelf transects, one covering Disko Bay andDisko Bank, while the other three covered Store Hellefiske Bank,Lille Hellefiske Bank and Sukkertop Bank. The hydrography wasexamined by CTD profiling, the phytoplankton by fluorescenceprofiling and water bottle sampling, while mesozooplankton andichthyoplankton were sampled by vertical or oblique net hauls,respectively. We observed distinct along-shelf flowing currentsin the area (e.g. the West Greenland Current, the Polar Currentand the Irminger Current), and the physical characteristicsindicated frontogenesis at the shelf slope, in regions of 80–100m water depth. Phytoplankton and ichthyoplankton showed a cross-shelfstructuring with apparent linkages to frontal characteristics,while a more diverse pattern was observed for the mesozooplanktonwhich were dominated by Calanus finmarchicus, Calanus glacialisand Calanus hyperboreus. The relationship between hydrographiccharacteristics and plankton distribution differed among species,and apparently specific plankton communities were establishedin different areas of the shelf. For example the larvae of Boreogadussaida, Ammodytes sp., Reinhardtius hippoglossoides and Stichaeuspunctatus differed markedly in distributional characteristics.In addition to the cross-shelf structuring, marked differencesin species composition and total plankton abundance were observedin the along-shelf (north–south) direction. The latitudinaldifferences in the unicellular plankton communities are interpretedlargely within a seasonal successional framework (i.e. an earlydominance of diatoms followed by increasing importance of smallerunicellular plankton), while the ichthyo- and zooplankton communitiesalso differed by the respective dominance of species with polarversus temperate origin. Our findings suggest that the flowof major currents and the establishment of hydrographical frontsare of primary importance to the plankton communities in theWest Greenland shelf area, influencing the early life of fishand the recruitment to the important fisheries resources.  相似文献
