The seven mammalian channels from the classical (TRPC) subfamily of transient receptor potential (TRP) channels are thought to be receptor-operated cation channels activated in a phospholipase C (PLC)-dependent manner. Based on sequence similarity, TRPC channels can be divided into four subgroups. Group 4 comprises TRPC4 and TRPC5, and is most closely related to group 1 (TRPC1). The functional properties observed following heterologous expression of TRPC4 or TRPC5 in mammalian cells are contradictory and, therefore, controversial. In our hands, and in several independent studies, both channels, probably as homotetramers, form receptor-operated, Ca2+-permeable, nonselective cation channels activated independently of inositol 1,4,5-trisphosphate (InsP3) receptor activation or Ca2+ store-depletion. As heteromultimers with TRPC1, TRPC4 and TRPC5 form receptor-operated, Ca2+-permeable, nonselective cation channels with biophysical properties distinct from homomeric TRPC4 or TRPC5. In other studies, TRPC4 and TRPC5 have been shown to be store-operated channels, with moderate to high Ca2+ permeabilities. At present there is no clear explanation for these major differences in functional properties. To date, little is known as to which native cation channels are formed by TRPC4 and TRPC5. Endothelial cells from TRPC4−/− mice lack a highly Ca2+-permeable, store-dependent current, and data support a role for TRPC4 in endothelium-mediated vasorelaxation. A similar current in adrenal cortical cells is reduced by TRPC4 antisense. From similarities in the properties of the currents and expression of appropriate isoforms in the tissues, it is likely that heteromultimers of TRPC1 and TRPC4 or TRPC5 form receptor-operated nonselective cation channels in central neurones, and that TRPC4 contributes to nonselective cation channels in intestinal smooth muscle.  相似文献

In 1997 and 1998, we sampled the Missouri River, North Dakota to determine if anthropogenic disturbances had influenced catostomid species composition and feeding ecology. We compared two distinct river segments, the Missouri River between the mouth of the Yellowstone River and Lake Sakakawea (the Yellowstone–Sakakawea segment (YSS)), a moderately altered segment and the Missouri River between Garrison Dam and Lake Oahe (the Garrison–Oahe segment (GOS)), a highly altered segment. The segments exhibited greatly different sucker communities. Bigmouth buffalo, Ictiobus cyprinellus, smallmouth buffalo, Ictiobus bubalus, and river carpsucker, Carpiodes carpio, represented 94% of the sucker catch in the YSS, whereas in the GOS, white sucker, Catostomus commersoni, and longnose sucker, Catostomus catostomus, constituted 98% of the sucker catch. In the YSS, high zooplankton densities led to greater sucker zooplanktivory and food niche overlap than in the GOS. Intense anthropogenic disturbances to the GOS are associated with the differences in sucker species composition, prey density and composition, and sucker feeding ecology between the two segments.  相似文献

The recognition of familiar areas by homing pigeons, Columba livia, is now known to depend at least in part on visual cues. Birds allowed a 5-min preview of the surrounding landscape prior to release home faster than those denied access to such cues, suggesting that recognition is visually mediated. We examined this phenomenon further by asking how memory generated through prior experience with a site is used in recognition. We provided a group of homing pigeons with training experience in which they viewed, through a single transparent vertical face of an otherwise opaque release box, an approximately 140° segment of the landscape for 5 min before release. Training previews were always given from consistent locations and orientations within a release site. Test releases that followed were used to ascertain whether subsequent site recognition necessitated previewing from this already familiar angle and distance, or whether the skill could be extended to novel, nonoverlapping lateral views of the landscape. The results suggested that homing performance was better after presentation of the view recapitulating that seen during training than after showing a novel alternative view. Pigeons may have been using a template of the arrangement of familiar landmarks around the release site as the cue for recognition. The effect, however, disappeared after repeated training, suggesting that repeated release from a site may allow for more extensive visual survey and the memorization of detailed features of the landscape or the extraction of site-specific general features that aid recognition even when a novel view is presented. Biologically generated structures create habitat and influence the distribution and abundance of species in many marine systems. In the rather monotonous and nutrient-poor environment of the deep-sea, cold seep environments and their associated chemosynthetic communities offer islands of primary production and habitat to a generally sparsely distributed macrofauna. In this study, we investigate the structure of macrofaunal assemblages associated with vestimentiferan aggregations on the upper Louisiana slope of the Gulf of Mexico and the relationships between assemblage composition and the size and complexity of the vestimentiferan-generated habitat. Using custom-designed and custom-built devices, we collected seven whole vestimentiferan aggregations along with their associated fauna during the summers of 1997 and 1998. Sixty-five species were found associated with the four vestimentiferan aggregations collected in 1998, more than doubling the number of species previously reported for seeps in this region. Individual aggregations contained between 23 and 44 different non-vestimentiferan species. General trends of increasing species richness with increasing habitat size and increasing faunal density with increasing habitat complexity were identified, but substantial variability suggested other factors also control the composition of faunal associates. Faunal abundances decreased with increasing aggregation age. Seep endemics dominated the communities of younger aggregations, but non-endemic species dominated communities of older aggregations. Relative dominance of the heterotrophic community by primary consumers decreased, while predatory secondary and higher-order consumers increased with increasing aggregation age. These trends are discussed in terms of successional changes in aggregation structure, habitat heterogeneity and environmental conditions.  相似文献

Muscle contractions evoke an immediate rise in blood flow. Distribution of this hyperemia within the capillary bed may be deterministic for muscle O(2) diffusing capacity and remains unresolved. We developed the exteriorized rat (n = 4) spinotrapezius muscle for evaluation of capillary hemodynamics before (rest), during, and immediately after (post) a bout of twitch contractions to resolve (second-by-second) alterations in red blood cell velocity (V(RBC)) and flux (f(RBC)). Contractions increased (all P < 0.05) capillary V(RBC) (rest: 270 +/- 62 microm/s; post: 428 +/- 47 microm/s), f(RBC) (rest: 22.4 +/- 5.5 cells/s; post: 44.3 +/- 5.5 cells/s), and hematocrit but not the percentage of capillaries supporting continuous RBC flow (rest: 84.0 +/- 0.7%; post: 89.5+/-1.4%; P > 0.05). V(RBC) peaked within the first one or two contractions, whereas f(RBC) increased to an initial short plateau (first 12-20 s) followed by a secondary rise to steady state. Hemodynamic temporal profiles were such that capillary hematocrit tended to decrease rather than increase over the first approximately 15 s of contractions. We conclude that contraction-induced alterations in capillary RBC flux and distribution augment both convective and diffusive mechanisms for blood-myocyte O(2) transfer. However, across the first 10-15 s of contractions, the immediate and precipitous rise in V(RBC) compared with the biphasic and prolonged increase of f(RBC) may act to lower O(2) diffusing capacity by not only reducing capillary transit time but by delaying the increase in the instantaneous RBC-to-capillary surface contact thought crucial for blood-myocyte O(2) flux.  相似文献

Theory and empirical results suggest that high biodiversity should often cause lower temporal variability in aggregate community properties such as total community biomass. We assembled microbial communities containing 2 to 8 species of competitors in aquatic microcosms and found that the temporal change in total community biomass was positively but insignificantly associated with diversity in a constant temperature environment. There was no evidence of any trend in variable temperature environments. Three non-exclusive mechanisms might explain the lack of a net stabilising effect of species richness on temporal change. (1) A direct destabilising effect of diversity on population level variances caused some populations to vary more when embedded in more diverse communities. (2) Similar responses of the different species to environmental variability might have limited any insurance effect of increased species richness. (3) Large differences in the population level variability of different species (i.e., unevenness) could weaken the relation between species richness and community level stability. These three mechanisms may outweigh the stabilising effects of increases in total community biomass with diversity, statistical averaging, and slightly more negative covariance in more diverse communities. Our experiment and analyses advocate for further experimental investigations of diversity-variability relations.  相似文献

Oligopeptidases of starter and nonstarter lactic acid bacteria contribute to the proteolytic events important in maturation and flavor development processes in cheese. This paper describes the molecular cloning, expression, and specificity of the oligopeptidase PepO from the probiotic nonstarter strain Lactobacillus rhamnosus HN001 (DR20). The pepO gene encodes a protein of 70.9 kDa, whose primary sequence includes the HEXXH motif present in certain classes of metallo-oligopeptidases. The pepO gene was cloned in L. rhamnosus HN001 and overexpressed in pTRKH2 from its own promoter, which was mapped by primer extension. It was further cloned in both pNZ8020 and pNZ8037 and overexpressed in Lactococcus lactis subsp. cremoris NZ9000 from the nisA promoter. The purified PepO enzyme demonstrated unique cleavage specificity for alpha(s1)-casein fragment 1-23, hydrolyzing the bonds Pro-5-Ile-6, Lys-7-His-8, His-8-Gln-9, and Gln-9-Gly-10. The impact of this enzyme in cheese can now be assessed.  相似文献

Abstract Observations of the large earth bumblebee, Bombus terrestris (L.), in native vegetation were collated to determine the extent to which this exotic species has invaded Tasmanian native vegetation during the first 9 years after its introduction. The range of B. terrestris now encompasses all of Tasmania's major vegetation types, altitudes from sea level to 1260m a.s.L, and the entire breadth of annual precipitation in the state from more than 3200 mm to less than 600 mm. Observations of workers carrying pollen, together with the presence of large numbers of bumblebees at many localities across this range indicate that colonies are frequently established in native vegetation. Evidence that colonies are often successful was obtained from repeated observations of the species during more than 1 year at particular sites. Unequivocal evidence of colonies was obtained from six National Parks, including four of the five in the Tasmanian Wilderness World Heritage Area (WHA). Indeed, the species has been present in the WHA for at least as long as it has in the city of Hobart, where it was first recorded. In southwestern Tasmania, evidence of colonies was obtained up to 40km from gardens, 61 km from small towns and 93 km from large towns. Hence, contrary to previous suggestions, the species is established in the most remote parts of Tasmania and is not dependent on introduced garden plants. Given their strong record of invasion, it is likely that B. terrestris will form feral populations on the mainland of Australia and in many other parts of the world if introduced. Because of their likely negative impacts on native animals and plants, and potential to enhance seed production in weeds, the spread of bumblebees should be avoided.  相似文献

We measured net ecosystem CO2 exchange (NEE) in Panamá over C4 pasture plots that varied in grazing intensity. After adjusting for variation in light, there were noticeable effects of grazing‐related variables on CO2 exchange that were largely dependent on the developmental stage of the plant canopy. Above‐ground productivity was positively related to grazing intensity (r2=0.30). Two experimentally grazed fields had significantly lower standing crop biomass but no significant difference in CO2 uptake (24.2 μmol/m2/s) compared with two ungrazed fields (20.3 μmol/m2/s). Grazed fields had significantly lower ecosystem respiration rates (10.3 μmol/m2/s) than did ungrazed fields (17.6 μmol/m2/s). These results suggest that, although these pastures were possible sources of CO2 during the time intervals sampled, the size of the sources tended to be dampened by cattle grazing through reductions in ecosystem respiration. Thus, it appears that disturbance caused by cattle grazing will not always result in an increase in CO2 release from tropical pastures to the atmosphere.  相似文献
