Summary The penetration of [3H]thymidine, [3H]d-leucine, [125I]albumin, and the drugs [3H]5-fluorouracil and [3H]vinblastine into human glioma spheroids (in vitro tumor models) was studied by a method based on rapid freezing, freeze
drying, vapor fixation, wax embedding, dry sectioning, and contact autoradiography. No significant disturbances in the distribution
of water soluble substances were observed. Thymidine andd-leucine penetrated the whole spheroids relatively fast, whereas albumin showed reduced penetration. the concentration of
albumin was highest at the periphery of the spheroids, but only smaller amounts were detected in the deeper regions. A significant
difference between the penetration patterns of the drugs studied was also observed. Fluorouracil penetrated rather freely,
but the penetration of vinblastine was limited.

Protohemin and covalently bound hemin were determined in eight aerobic bacterial strains. A good correlation between protohemin content and luminol reactivity was found. The ratio of luminol reaction to protohemin for the eight investigated strains was essentially identical to that of pure protohemin, 0.7 X 10(16) mV/mol. Covalently bound hemin contributed to the chemiluminescence to a minor extent only (0.7 X 10(14) mV/mol, in accordance with earlier observations of the lower reactivity of cytochrome c and related compounds. A difference in reaction kinetics of the luminol reaction with covalently bound hemin (slower reaction than protohemin) and protohemin was observed in vivo as well as in vitro. We examined effects of protein kinase C (PKC) activation by phorbol dibutyrate (PDB) on prostaglandin production in astroglia. Astroglia were cultured from sheep fetal cortex and grown in Eagle's basal media supplemented with 10% fetal calf serum (BME-C). Prostaglandin F2a (PGF2α) levels in media were determined at 2–24 hours after exposure to PDB. PDB increased production of PGF2α at 10−8M and 10−6M. In addition, PDB increased the ratio of membrane to cytosolic PKC. Coapplication of H7 [1-(5-isoquinolinylsulfonyl)-2-methyl-piperazine] (10−4M) with PDB (10−6M) inhibited PDB-induced PGF2a production. To investigate the role of protein synthesis in increased prostaglandin production by PDB, astroglia were coincubated with actinomycin D (1 mg/ml) or cycloheximide (10 mg/ml). At 4 hrs, both actinomycin D and cycloheximide inhibited increases in PGF2a in response to PDB application. In addition, COX-2 mRNA levels and COX activity levels were examined. PDB increased COX-2 mRNA levels by 2 hours, and COX activity tripled after 12 hr exposure to PDB. In addition, the increase in COX activity was blocked by cycloheximide. In summary, PKC activation promotes enhanced prostaglandin production via an increase in COX synthesis. 相似文献
