Summary When cambial tissues are removed from tree stems of Eucalyptus regnans F. Muell. and incubated in vitro with [14C]glucose, [14C]galactose and [14C]fructose in the presence of sufficient PVP (polyvinylpyrrolidone) radioactive sucrose, members of the raffinose family and related sugars are synthesized. These results suggest that PVP inactivates substances which, in in vitro experiments, inhibit the action of enzymes which are essential to the resynthesis of nutrient sugars (sucrose, raffinose and stachyose). Optically active lumazines (biolumazine, dictyolumazine, monalumazine, and neolumazine) are prepared from the corresponding pterins by enzymatic reaction, using pterin deaminase excreted by Dictyostelium discoideum. The fluorescence properties, circular dichroism spectra, and chromatographic behavior of these lumazines are studied. D - and L -enantiomers of biolumazine, dictyolumazine, and monalumazine are separated using a chiral flavoprotein column. This column also separates the enantiomeric pterins of the threo form: monapterin and dictyopterin. However, the column does not separate the enantiomeric pterins of the erythro form: neopterin and biopterin. By coupling a reverse-phase column to the flavoprotein column, the separation of pterins and lumazines in function of their hydrophobicity, as well as the separation of the diastereomers, is achieved. This coupled achiral/chiral high-performance liquid chromatography method enables determination of the stereoconfiguration of natural lumazines by comparison with optically pure compounds. A lumazine derivative, present in the extracellular medium of Dictyostelium discoideum, is identified as D -dictyolumazine, i.e., 6-(D -threo-1,2-dihydroxypropyl)-lumazine. © 1994 Wiley-Liss, Inc. 相似文献

Studies of gene‐targeted mice identified the roles of the different pro‐survival BCL‐2 proteins during embryogenesis. However, little is known about the role(s) of these proteins in adults in response to cytotoxic stresses, such as treatment with anti‐cancer agents. We investigated the role of BCL‐XL in adult mice using a strategy where prior bone marrow transplantation allowed for loss of BCL‐XL exclusively in non‐hematopoietic tissues to prevent anemia caused by BCL‐XL deficiency in erythroid cells. Unexpectedly, the combination of total body γ‐irradiation (TBI) and genetic loss of Bcl‐x caused secondary anemia resulting from chronic renal failure due to apoptosis of renal tubular epithelium with secondary obstructive nephropathy. These findings identify a critical protective role of BCL‐XL in the adult kidney and inform on the use of BCL‐XL inhibitors in combination with DNA damage‐inducing drugs for cancer therapy. Encouragingly, the combination of DNA damage‐inducing anti‐cancer therapy plus a BCL‐XL inhibitor could be tolerated in mice, at least when applied sequentially. 相似文献

Plasmodium vivax is responsible for most of the malaria infections outside Africa and is currently the predominant malaria parasite in countries under elimination programs. P. vivax preferentially enters young red cells called reticulocytes. Advances in understanding the molecular and cellular mechanisms of entry are hampered by the inability to grow large numbers of P. vivax parasites in a long‐term in vitro culture. Recent progress in understanding the biology of the P. vivax Reticulocyte Binding Protein (PvRBPs) family of invasion ligands has led to the identification of a new invasion pathway into reticulocytes, an understanding of their structural architecture and PvRBPs as targets of the protective immune response to P. vivax infection. This review summarises current knowledge on the role of reticulocytes in P. vivax infection, the function of the PvRBP family of proteins in generating an immune response in human populations, and the characterization of anti‐PvRBP antibodies in blocking parasite invasion. 相似文献

Macroalgae provide a substantial and renewable resource that can be sustainably utilized for economic and social benefit. A US$7 billion global industry already exists for macroalgae, but the huge majority of this is based on the production of species belonging to approximately six genera, within eight countries, for the manufacture of foods, industrial biomaterials and agricultural products. However, seaweed-derived functional products spanning numerous chemical classes have been identified with valuable therapeutic and industrial applications. This review focuses on the breadth of valuable bioproducts that could be produced from the seaweeds of Southern Australia—a hotspot for seaweed diversity, and the pathways available for their sustainable commercial production. This region contains among the highest level of recorded macroalgal diversity and endemism in the world, with approximately 1,200 described species, of which 62 % are considered endemic. Whilst a number of these species have been shown to be rich sources of higher-value functional products, and most of them still await exploration in this field, the seaweed industry of Southern Australia is largely limited to the harvest of beach-cast biomass for the manufacture of lower-value commodities such as fertilizer and animal feed. There is potential for the development of a substantial industry based on human functional products from seaweeds in Southern Australia. However, a number of challenges and knowledge gaps—including environmental, technological, agronomic, political, and cultural factors—are identified in this review, which must be addressed before sustainable expansion can be achieved. Furthermore, numerous strategic approaches and areas of suggested foci are underscored for research bodies and industry alike. Particular emphasis is given to the need for comprehensive surveying and bioprospecting of the resource; a focus on advanced downstream processing capabilities for improving production efficiency and enhancing product value; the use of biorefinery approaches to improve utilisation efficiency; and pursuing means of improving the sustainability of supply chains. 相似文献

The remarkable deformability of red blood cells (RBCs) depends on the viscoelasticity of the plasma membrane and cell contents and the surface area to volume (SA:V) ratio; however, it remains unclear which of these factors is the key determinant for passage through small capillaries. We used a microfluidic device to examine the traversal of normal, stiffened, swollen, parasitised and immature RBCs. We show that dramatic stiffening of RBCs had no measurable effect on their ability to traverse small channels. By contrast, a moderate decrease in the SA:V ratio had a marked effect on the equivalent cylinder diameter that is traversable by RBCs of similar cellular viscoelasticity. We developed a finite element model that provides a coherent rationale for the experimental observations, based on the nonlinear mechanical behaviour of the RBC membrane skeleton. We conclude that the SA:V ratio should be given more prominence in studies of RBC pathologies. 相似文献
