Delivery of microbial products into the mammalian cell cytosol by bacterial secretion systems is a strong stimulus for triggering pro-inflammatory host responses. Here we show that Salmonella enterica serovar Typhi (S. Typhi), the causative agent of typhoid fever, tightly regulates expression of the invasion-associated type III secretion system (T3SS-1) and thus fails to activate these innate immune signaling pathways. The S. Typhi regulatory protein TviA rapidly repressed T3SS-1 expression, thereby preventing RAC1-dependent, RIP2-dependent activation of NF-κB in epithelial cells. Heterologous expression of TviA in S. enterica serovar Typhimurium (S. Typhimurium) suppressed T3SS-1-dependent inflammatory responses generated early after infection in animal models of gastroenteritis. These results suggest that S. Typhi reduces intestinal inflammation by limiting the induction of pathogen-induced processes through regulation of virulence gene expression.  相似文献

Disentangling the role of mechanisms driving metacommunity structure is fundamental for conservation strategies. Several studies have been done in aquatic communities; however, little is known about the factors driving oomycete communities. This research aimed to investigate beta diversity patterns and assess the role of environmental (chemical, physical, and hydrologic), spatial, and temporal (sampling months) factors in driving oomycete beta diversity in a spatial extent of 33 km from two Brazilian rivers. We took water samples in 10 sites quarterly, from August 2017 to May 2018. The partition of beta diversity into its components – species replacement and richness difference – was performed using the Jaccard dissimilarity index. Distance-based redundancy analysis and variation partitioning were used to assess the relationship between explanatory variables and beta diversity. We found that beta diversity was spatially and temporally high, and the replacement component was the main driver of the oomycete metacommunity’s beta diversity. Replacement and total beta diversity were explained mainly by spatial location and the month of sampling, while the richness difference was more associated with the environmental variables chlorophyll a and ammonia. Our findings suggest that dispersal limitation (spatial) and temporal factors are the main drivers of the total beta diversity and replacement in the oomycete metacommunity, while species sorting (environmental factor) influences the richness difference. Accordingly, that taking temporal factors into account in metacommunity studies is important to explain beta diversity patterns, especially in rivers with remarkable variability in hydrological regime and under eutrophic conditions.

There is strong evidence implicating nitric oxide (NO) in the pathophysiology of migraine and aura. Therefore, genetic polymorphisms in the endothelial NO synthase (eNOS) gene have been studied as candidate markers for migraine susceptibility. We compared for the first time the distribution of eNOS haplotypes including the three clinically relevant eNOS polymorphisms (T(-786)C in the promoter, rs2070744; Glu298Asp in exon 7, rs1799983; and a 27?bp variable number of tandem repeats in intron 4) and two additional tagging single-nucleotide polymorphisms (rs3918226 and rs743506) in 178 women with migraine (134 without aura and 44 with aura) and 117 healthy controls (control group). Genotypes were determined by TaqMan allele discrimination assay, real-time polymerase chain reaction, and polymerase chain reaction followed by fragment separation by electrophoresis. Autotransporter (AT) protein-encoding genes of diarrheagenic Escherichia coli (DEC) pathotypes (cah, eatA, ehaABCDJ, espC, espI, espP, pet, pic, sat, and tibA) were detected in typical and atypical enteropathogenic E. coli (EPEC) in frequencies between 0.8% and 39.3%. Although these ATs have been described in particular DEC pathotypes, their presence in EPEC indicates that they should not be considered specific virulence markers.  相似文献

The objective of this study is to define the geographic distribution of Leopardus tigrinus and L. geoffroyi in one of the few regions of South America where they co-occur, the state of Rio Grande do Sul (RS), in southernmost Brazil. We compiled 133 records for both species and constructed a distribution map, which shows sharp geographic segregation between them. Leopardus tigrinus was found to be associated more with forested ecoregions in the northern part of the state, while L. geoffroyi records were mainly associated with open habitats of the Pampas biome in southern RS. We present data on the diet of these two species that indicate trophic niche separation between them in this region of geographic contact. Our results thus suggest that these species exhibit ecological partitioning with respect to habitat and prey, and that these factors may influence the observed pattern of limited spatial overlap in this region.  相似文献

Ecological niche modelling (ENM) has been used to quantify the potential occurrence of species, by identifying the main environmental factors that determine the presence of species across geographical space. We provide a large-scale survey of the distribution of ostracod species in South America, by using the domains of 25 river basins. From 221 known ostracod species, we estimate the potential distribution of 61 species, using ENM. Ten clusters of potential distribution patterns were found. Clusters 8 and 9 grouped most of the species, which presented high similarity of niche between them. Heterocypris paningi Brehm, 1934 (group 1) obtained higher niche variability. The minimum temperatures of the coldest month and the mean elevation of the river basin were most important to predict the potential distribution of ostracods of most groups. South America has a complex pattern of elevation, which affects species distributions indirectly through changes in local factors. For instance, the Andes mountains might impose a barrier for ostracod distribution in the southern part of South America because of the low temperatures and precipitation. The ENM indicated that some regions and/or basins of South America might be susceptible to the entry of several ostracod species, presently absent, including non-native species.

Erythrocharax altipinnis is described from the Serra do Cachimbo, Pará, Brazil. The new taxon is distinguished from all of the Characidae genera by having the pelvic bones firmly attached through the isquiatic processes; a nearly triangular hiatus in the musculature covering the anterior chamber of the swim bladder between the first and second pleural ribs (pseudotympanum); the pedunculate, notably expanded and distally compressed teeth in both jaws; circumorbital series represented by antorbital and four infraorbital bones with laterosensory canals not enclosed; a single tooth row in the premaxillary with the teeth perfectly aligned and similar in shape and cusp number; the first three branched dorsal-fin rays distinctly elongate in males; a bright red adipose and caudal fins in life; a conspicuous dark midlateral stripe extending from the opercle to the tip of the median caudal-fin rays; and by the absence of a humeral spot. The phylogenetic position of the new taxon is discussed using morphological and molecular datasets, with conflicting results of both approaches discussed. Additionally, a summarized discussion on the current problems in the Characidae taxonomy is presented and the principal biases in the morphological dataset are also discussed.  相似文献
