Abstract Dextran sulphate (DS) 500 (M.W. 500 000) is commonly used as a reticuloendothelial (RE) blocker. We found that lipopolysaccharide (LPS)-induced tumour necrosis factor (TNF) production in sera was enhanced when mice were pretreated with DS500. When mice were pretreated with DS1000 (M.W. 1 000 000), TNF activity in sera was also significantly enhanced by the LPS injection in comparison with the saline-treated group, but not by the pretreatment with the low molecular weight of DS5 (M.W. 5 000), neutral dextran (Dex) 500, or positively-charged diethylaminoethyl dextran (DEAE-Dex) 500. The enhancement of LPS-induced TNF production occurred from 2 h after DS500 pretreatment. Pretreatment with DS500 or DS1000 significantly suppressed the carbon clearance from the blood in mice from 2 h after DS injection, but this suppression was not detected by the pretreatment with DS5, Dex500, or DEAE-Dex500. We suggest that negative-charge and high molecular weight are essential for dextran derivatives to enhance LPS-induced TNF production, and that the enhancing effect of DS is closely related to the suppression of the RE function. 相似文献

Fusogenic liposome (FL) is a delivery system that can transfer encapsulated materials into living cells directly through membrane fusion. FL is a promising approach for gene therapy because it can deliver various genetic materials much more efficiently than other non-viral vectors without damaging the cell. FL-mediated gene transfer consists of two independent membrane fusion phenomena; generation of a FL by fusing a Sendai virus (SV) particle with a simple liposome encapsulating DNA, and successive fusion of the FL with cell membrane. The former requires viral F protein but no other special molecule on the liposomal membrane, whereas the latter may require the receptor (sialic acid) and unidentified assistant molecule(s) on the cell membrane. Further analysis suggests that these assistant molecule(s), not the receptor, may control the fusion and govern the cell specificity of FL-mediated delivery. This review has described a detailed analysis of these fusion phenomena and discussed possible applications of FL-mediated gene delivery to human gene therapy. 相似文献

Prevalence of hepatitis B (HB) markers, antibodies to human T cell leukemia virus (HTLV-1) and antibodies to human immune deficiency virus 1 (HIV-1) in prostitutes working in Fukuoka city were studied. Sera were collected from 237 prostitutes during January-September, 1986. Among them, 9 (3.8%) were HB virus surface-antigen (HBs Ag) positive, of whom, 3 were HBe antigen positive and the remaining 6 were anti-HBe positive. The positive rate of anti-HBs was 34.2%. The incidence of anti-HTLV-1 in the prostitutes was 5.9%. These incidences are considered to be within the usual range in Kyushu district. No seropositive case for anti-HIV was found. 相似文献

It has been noted that V. parahaemolyticus isolated from patients with food poisoning are almost always thermostable direct hemolysin (TDH)-positive, whereas, isolates from foods or environmental sources are usually TDH-negative. The virulence of V. parahaemolyticus in mice was examined by using intraperitoneal and orogastric challenge models, where the strains used were those isolated from patients and foods with food poisoning and included TDH-positive and -negative. The LD50 in mice was estimated to be approximately 10(7) in the former and 10(8) in the latter challenge model. In the mice killed by challenge, either intraperitoneal or orogastric, the following pathological changes were almost always observed: swelling, redness and fluid accumulation in the small intestine, particularly the upper part of it. Histologically, congestion, edema, and vacuolation were observed in mucosal and submucosal tissues; furthermore, ulceration at tips of some villi was noticed. The lethality and pathological finding in mice due to V. parahaemolyticus were demonstrated similarly in both challenge models and irrespective of the TDH phenotype of the strain used. It will be suggested that the findings in challenged mice may be attributed to enteropathogenic factor(s) other than TDH of V. parahaemolyticus. 相似文献
