The application of the theory of homeomorphic transformations of topological manifolds and the operation of the connected sum of manifolds for a formation of a topological model of membrane transformations during the division process of cellular and subcellular compartments, has been shown. The biological cell and the subcellular structures in the form of vesicles are modelled by an arrangement of two concentric spheres corresponding to the inner and outer layer of the membrane bounding the vesicle. The analysis shows eight succeeding topological stages of membrane transformations during the division process and these stages are characterised. It is concluded that there is a vectorial translocation of lipid molecules from the inner layer of the membrane bounding the vesicle before the division process to the outer layer of the membranes after the division process and there is no lipid translocation from the outer layer to the inner layers during the division process.  相似文献

Summary The demography of populations living in variable environments is an important factor molding the evolution of ecological niches, for it determines the relative strength of selection pressures on adaptations to different habitats. Here I consider a coarse-grained environment consisting of two habitat types and investigate how the selection pressure on reproductive success in different habitats depends on their quality and frequency and the dispersal pattern. The results suggest that selection on adaptations to optimal habitats will usually be stronger than on adaptations to poor habitats and the ecological niche will thus tend to be an evolutionarily conservative character. It is because under the habitat choice or limited dispersal that seem to prevail in natural populations, more individuals encounter the better habitat than would be expected solely on the basis of its relative area. This bias results in reduced selection pressure on reproductive success in the poorer habitat. With habitat choice or limited dispersal, selection pressure on reproductive success in the poorer habitat may exceed that on reproductive success in the better habitat only if the poorer habitat is much more frequent in the environment than the better habitat and the difference in their quality is not large.  相似文献

Normal human and diet-induced hyperlipidemic Rhesus monkey serum low-density lipoprotein structure was investigated by freeze-etching electron microscopy employing a novel rapid freezing technique. Using turnip yellow mosaic virus as a standard, the technique was shown to be capable of providing information regarding the general architecture of particles in solution. Human and monkey serum low-density lipoprotein particle morphology appeared to deviate markedly from that of a perfect sphere. Instead, the outer layer of the particles appeared to consist of a small number of globules. The number and dimensions of these globules as well as their arrangement are in remarkable agreement with the tetrahedral model proposed by Luzzati et al. (1979) in the preceding paper for the low temperature form of the Rhesus monkey low-density lipoprotein.  相似文献

The fine structure of Manduca sexta and Sesamia nonagrioides chorion was investigated by scanning electron microscopy and freeze-fracturing. In both species the mature chorion exhibits a complex ultrastructure on its outer surface, with a large number of aeropyles forming polygonal arrays. The micropyle is surrounded by a rosette of approximately 80 follicular cell imprints. Scanning electron microscopy of vertically ripped sections reveals that both chorions consist of two main layers: a trabecular layer closest to the oocyte and a lamellar layer. The technique of freeze-fracturing, utilizing single-sided and rotary shadowing, clearly shows that fibrils, approximately 3-4 nm in diameter, constitute chorionic lamellae in both species. The fibrils appear to have a 'beaded' structure, with a 2-3 nm axial periodicity. Freeze-fracturing also provides a direct visualization of the helicoidal arrangement of these fibrils for the formation of chorion supramolecular architecture.  相似文献

Conventional freeze-fracturing electron microscopy was used to study water-soluble proteins and different forms of Ca2+-ATPase-detergent complexes. Freeze-fracture images of solutions containing proteins larger than myoglobin showed the presence of distinct, randomly dispersed particles on smooth fracture surfaces. The distribution of sizes of these particles was closely to Gaussian, with a mean size which was correlated to the Stokes diameter. Monomeric Ca2+-ATPase from sarcoplasmic reticulum, solubilized by deoxycholate or a non-ionic detergent, showed a bimodal distribution of particle sizes. Even more complex distributions were found for dimeric and trimeric preparations of Ca2+-ATPase. The results can be interpreted on the assumption that the Ca2+-ATPase molecule is elongated, with an overall length of about 110 A and a width in its largest part of about 75 A. It is concluded on the basis of the presented results that freeze-fracture electron microscopy can be successfully used for morphological studies of protein molecules in solution.  相似文献

In biochemically active sarcoplasmic reticulum vesicles (SR) the physical state of the membrane lipids was studied by high angle x-ray diffraction and proton nuclear magnetic resonance (NMR) at 220 MHz, and related to thermal effects observed in SR functional parameters. It is shown by high angle x-ray diffraction that even at temperatures as low as 1 degree C nearly all the SR lipid hydrocarbon chains are in a disordered conformation and only a very small part (less than 3%) are in rigid crystalline order. Consistent with this observation, the NMR data indicate that the majority of SR phospholipid molecules are in a state of restricted anisotropic motion having no apparent crystalline order at temperatures as low as 5 degrees C. At this temperature most of the resonance signal is contained in a broad feature-less line of 700-Hz half-width. On the other hand, as the temperature is raised, high-resolution NMR signals, representing groups with highly isotropic motion, begin to grow in intensity. It is estimated that by 35 degrees C 90-100% of the phosphatidylcholine N-methyl protons and 35% of the hydrocarbon-chain protons give high-resolution signals. Concurrent studies on functional parameters reveal thermal effects giving rise to nonlinear Arrhenius plots for the rates of calcium transport and calcium activated ATPase. The thermal effects observed on functional parameters and on the character of phospholipid molecular motion exhibit a parallel behavior, suggesting a relationship between enzyme activity and the physical state of the membrane lipids.  相似文献

Recent studies have demonstrated that chemerin participates in the regulation of female reproductive function at the level of the ovaries. Due to the lack of data concerning the presence of the chemerin system (chemerin and its receptors: CMKLR1, GPR1, CCRL2) in the ovaries of pigs, one of the most economically important livestock species, the aim of this study was to investigate the expression and localization of chemerin and its receptors in the ovaries of prepubertal and mature gilts. We also aimed to examine the concentrations of chemerin in the follicular fluid of prepubertal and mature animals. In the present study, we have demonstrated the expression patterns of chemerin system components in the porcine follicles of different sizes of prepubertal and mature animals, as well as in corpora lutea of mature gilts during the estrous cycle and early pregnancy. The obtained results suggest that the expression of chemerin system components is influenced by the reproductive stage, cell type, and the hormonal status of gilts (the estrous cycle/pregnancy). We have also presented the localization of the chemerin system components in various ovarian structures, and also showed changes in the concentration of chemerin in the follicular fluid of pigs. The presented findings not only confirm that chemerin is produced locally in the porcine ovary but they also demonstrate that chemerin directly affects ovarian cells, as confirmed by the presence of chemerin receptors in all ovarian structures. Therefore, chemerin appears to be an important intra‐ovarian factor that could regulate ovary function in pigs.  相似文献

Periods of nutrient shortage impose strong selection on animal populations. Experimental studies of genetic adaptation to nutrient shortage largely focus on resistance to acute starvation at adult stage; it is not clear how conclusions drawn from these studies extrapolate to other forms of nutritional stress. We studied the genomic signature of adaptation to chronic juvenile malnutrition in six populations of Drosophila melanogaster evolved for 150 generations on an extremely nutrient-poor larval diet. Comparison with control populations evolved on standard food revealed repeatable genomic differentiation between the two set of population, involving >3,000 candidate SNPs forming >100 independently evolving clusters. The candidate genomic regions were enriched in genes implicated in hormone, carbohydrate, and lipid metabolism, including some with known effects on fitness-related life-history traits. Rather than being close to fixation, a substantial fraction of candidate SNPs segregated at intermediate allele frequencies in all malnutrition-adapted populations. This, together with patterns of among-population variation in allele frequencies and estimates of Tajima’s D, suggests that the poor diet results in balancing selection on some genomic regions. Our candidate genes for tolerance to larval malnutrition showed a high overlap with genes previously implicated in acute starvation resistance. However, adaptation to larval malnutrition in our study was associated with reduced tolerance to acute adult starvation. Thus, rather than reflecting synergy, the shared genomic architecture appears to mediate an evolutionary trade-off between tolerances to these two forms of nutritional stress.  相似文献
