Four new sulfated steroidal glycosides (asterosaponins), hippasteriosides A-D (1-4, resp.), were isolated from the alcoholic extract of the Far Eastern starfish Hippasteria kurilensis, collected in the Sea of Okhotsk near Kuril Islands. Compounds 1-4 were determined to contain the same unprecedented hexasaccharide moiety, β-D-xylopyranosyl-(1→3)-β-D-fucopyranosyl-(1→2)-β-D-quinovopyranosyl-(1→4)-[β-D-quinovopyranosyl-(1→2)]-β-D-xylopyranosyl-(1→3)-β-D-quinovopyranosyl, linked to C(6) of 3-O-sulfonylated steroidal aglycons, which differ from each other in their side-chain structures. Structures 1-4 were elucidated by extensive NMR and MS techniques, and chemical transformations. Not all the asterosaponins show a prominent cytotoxic activity against human colon tumor HT-29 cells, but some of them, especially compound 4 demonstrate a remarkable inhibition of the HT-29 colony formation in soft-agar clonogenic assay, suggesting its anticancerogenic properties. 相似文献

The role of endothelial ABCA1 expression in reverse cholesterol transport (RCT) was examined in transgenic mice, using the endothelial-specific Tie2 promoter. Human ABCA1 (hABCA1) was significantly expressed in endothelial cells (EC) of most tissues except the liver. Increased expression of ABCA1 was not observed in resident peritoneal macrophages. ApoA-I-mediated cholesterol efflux from aortic EC was 2.6-fold higher (P < 0.0001) for cells from transgenic versus control mice. On normal chow diet, Tie2 hABCA1 transgenic mice had a 25% (P < 0.0001) increase in HDL-cholesterol (HDL-C) and more than a 2-fold increase of eNOS mRNA in the aorta (P < 0.04). After 6 months on a high-fat, high-cholesterol (HFHC) diet, transgenic mice compared with controls had a 40% increase in plasma HDL-C (P < 0.003) and close to 40% decrease in aortic lesions (P < 0.02). Aortas from HFHC-fed transgenic mice also showed gene expression changes consistent with decreased inflammation and apoptosis. Beneficial effects of the ABCA1 transgene on HDL-C levels or on atherosclerosis were absent when the transgene was transferred onto ApoE or Abca1 knockout mice. In summary, expression of hABCA1 in EC appears to play a role in decreasing diet-induced atherosclerosis in mice and is associated with increased plasma HDL-C levels and beneficial gene expression changes in EC. 相似文献

Introduction
Starfish are recognized as interesting source of natural steroid products with pharmaceutical potential. Polar steroid metabolites of starfish have unique chemical structures and exhibit various biological activities but their biological functions are controversial.Objectives
The objective of this study was to investigate the response of polar steroid metabolome of the starfish Patiria (=Asterina) pectinifera on various environmental factors and stresses.Methods
Here we first have applied MS-based environmental metabolomics to elucidate the metabolic changes of polar steroid metabolome of starfish. Using HPLC–ESI–Q/TOF–MS approach followed by statistical analysis including principal component analysis and partial least squares discriminant analysis for data classification and potential biomarkers selection, we investigated the changes induced by feeding, injury, variations in water temperature and salinity, and oxygen deficiency.Results
According to multivariate and univariate statistical analysis the responses to feeding, injury and water heating were better expressed than the others and have some similarity in their action on the steroid metabolome of the starfish P. pectinifera. Most constituents of asterosaponin pool were reduced and most constituents of polyhydroxysteroid and related glycoside pool were increased at that.Conclusion
Our results indicate that various metabolic changes in polar steroid constituents of P. pectinifera are induced by feeding and stresses. We believe that these responses are connected with biological multifunctionality of these compounds.96.

Free sterol fractions from the holothurians (sea cucumbers) Synapta maculata, Cladolabes bifurcatus and Cucumaria sp. have been isolated and studied by HPLC, GLC, GLC-MS and NMR methods. Forty seven sterols were identified, including several rare ones. In contrast with previously studied holothurians, the presence of a minor amount of Delta7 sterols was indicated in the sterols of S. maculata. This animal contains predominantly Delta(9(11))sterols as well as an abnormally high concentration of Delta5 sterols. In C. bifurcatus and Cucumaria sp., 14alpha-methyl and 4alpha,14alpha-dimethyl-Delta(9(11))sterols were found to be the main sterol constituents. Peculiarities of sterol distribution and the relationship between sterol compositions and taxonomic positions, ecology and toxicity of the corresponding sea cucumbers were discussed. 相似文献

The distribution of free sterols, polyhydroxysteroids and steroid glycosides in different body components of the Far-eastern starfish Patiria (=Asterina) pectinifera has been studied. It was shown that free sterol fractions from aboral and oral body walls, gonads, stomach and pyloric ceca contained Delta(7) sterols with a preponderance of 5alpha-cholest-7-en-3beta-ol. All these body components had also toxic steroid oligoglycosides. However, polyhydroxysteroids and related low molecular weight steroid glycosides were found in stomach and pyloric ceca only. In pyloric ceca, the sulfated monoside 'asterosaponin' P(1) was identified as a main polar steroid, whereas 6-sodium sulfate of cholestane-3beta,4beta,6alpha,7alpha,8,15beta,16beta,26-octaol predominated in the stomach. Probable biological functions of polar steroids and free sterols in this starfish were discussed. It was suggested that some polyhydroxysteroids and related monoglycosides play the same biological role as bile alcohols and bile acids do in vertebrates. 相似文献

An aryl sulfatase of unusual specificity has been isolated from the liver of marine mollusk Littorina kurila. It hydrolyzes p-nitrophenyl sulfate, does not affect the natural fucoidan, and catalyzes splitting off of the sulfate group in position C4 of xylose residues within the carbohydrate chains of holostane triterpene glycosides from sea cucumbers. The properties of the enzyme were studied at pH 5.4. The protein is homogeneous according to electrophoresis and has M 45 +/- 1 kDa. The semiinactivation time of the enzyme at 60 degrees C is 20 min, and its Km value for the hydrolysis of p-nitrophenyl sulfate is 8.7 +/- 1 mM. It was shown that natural sulfated polyhydroxysteroids inhibit activity of the sulfatase; their I50 values depend on their structures and are within the range from 10(-3) to 10(-5) M. 相似文献
