The purpose of this study was to assess the relationship of accelerometer output, in counts (ActiGraph GT1M) and as raw accelerations (ActiGraph GT3X+ and GENEA), with ground reaction force (GRF) in adults. Ten participants (age: 29.4 ± 8.2 yr, mass: 74.3 ± 9.8 kg, height: 1.76 ± 0.09 m) performed eight trials each of: slow walking, brisk walking, slow running, faster running and box drops. GRF data were collected for one step per trial (walking and running) using a force plate. Low jumps and higher jumps (one per second) were performed for 20 s each on the force plate. For box drops, participants dropped from a 35 cm box onto the force plate. Throughout, three accelerometers were worn at the hip: GT1M, GT3X+ and GENEA. A further GT3X+ and GENEA were worn on the left and right wrist, respectively. GT1M counts correlated with peak impact force (r = 0.85, p < 0.05), average resultant force (r = 0.73, p < 0.05) and peak loading rate (r = 0.76, p < 0.05). Accelerations from the GT3X+ and GENEA correlated with average resultant force and peak loading rate irrespective of whether monitors were worn at the hip or wrist (r > 0.82, p < 0.05, r > 0.63 p < 0.05, respectively). In conclusion, accelerometer count and raw acceleration output correlate positively with GRF and thus may be appropriate for the quantification of activity beneficial to bone. Wrist-worn monitors show a similar relationship with GRF as hip-worn monitors, suggesting that wrist-worn monitors may be a viable option for future studies looking at bone health. 相似文献

Sickle cell anemia (SCA) associated cerebrovascular disease includes vascular remodeling, abnormal cerebral blood flow (CBF) and infarction. We studied the relationships between plasma brain derived neurotropic factor (BDNF), platelet derived growth factors (PDGF-AA and -AB/BB) and high trans-cranial Doppler (TCD) velocity, an indication of CBF velocity. Baseline plasma samples from 39 children (19 SCA with abnormal/high TCD [SATCD], 13 SCA with normal TCD [SNTCD] and 7 healthy non-SCA), were assayed for BDNF, PDGF-AA and -AB/BB plus 11 other cytokines. The sensitivity, specificity and usefulness of these biomarkers for stroke prediction was investigated. All subject groups were of similar age and gender distribution. Mean BDNF was significantly higher among SATCD than SNTCD (p=0.004) as was mean PDGF-AA (p=0.001). Similarly, mean PDGF-AA was higher among SCA subjects who developed stroke than those who did not (p=0.012). Elevated BDNF and PDGF-AA were good predictors of the presence of abnormally high CBF velocity and were both associated with severity of anemia. Elevated PDGF-AA predicted risk for stroke development. Stroke incidence and high TCD velocity were associated with elevated BDNF and PDGF-AA. These findings suggest a role for BDNF and PDGF-AA in the patho-physiological mechanism of cerebrovascular disease in SCA. 相似文献

An efficient protocol for the in vitro micrpropagation of Saussurea involucrata Kar. et Kir, an endangered Chinese medicinal plant, was developed. Shoot organogenesis was obtained following culture of leaf explants on Murashige and Skoog (MS) medium supplemented with thidiazuron (TDZ). After 28 d of culture, 15.6?±?1.4 shoots were regenerated per leaf explant on MS medium containing 0.5 ??M TDZ. After transfer of shoots to a medium containing 5.0 ??M indole-3-acetic acid, approximately 80% of the regenerated shoots formed roots and whole plantlets. After transfer of rooted shoots to the greenhouse, 83% of the regenerated plantlets survived and grew vigorously. The regeneration protocol developed in this study provides a basis for germplasm conservation and for the production of plant material necessary to study the medicinally active components of S. involucrata. 相似文献

Accumulation of depolarized mitochondria within beta-cells has been associated with oxidative damage and development of diabetes. To determine the source and fate of depolarized mitochondria, individual mitochondria were photolabeled and tracked through fusion and fission. Mitochondria were found to go through frequent cycles of fusion and fission in a 'kiss and run' pattern. Fission events often generated uneven daughter units: one daughter exhibited increased membrane potential (delta psi(m)) and a high probability of subsequent fusion, while the other had decreased membrane potential and a reduced probability for a fusion event. Together, this pattern generated a subpopulation of non-fusing mitochondria that were found to have reduced delta psi(m) and decreased levels of the fusion protein OPA1. Inhibition of the fission machinery through DRP1(K38A) or FIS1 RNAi decreased mitochondrial autophagy and resulted in the accumulation of oxidized mitochondrial proteins, reduced respiration and impaired insulin secretion. Pulse chase and arrest of autophagy at the pre-proteolysis stage reveal that before autophagy mitochondria lose delta psi(m) and OPA1, and that overexpression of OPA1 decreases mitochondrial autophagy. Together, these findings suggest that fission followed by selective fusion segregates dysfunctional mitochondria and permits their removal by autophagy. 相似文献

An overview of the basic principles of photochemistry is presented to facilitate discussion of fluorescence, quenching and quantum yields. These topics in turn provide the foundation for an account of fluorescence spectroscopy and its application to microscopy. A brief overview of light microscopy and the application of fluorescence microscopy is given. The influences of molecular features, such as aromatic character and substitution patterns, on color and fluorescence are described. The concept of color fading is considered with particular reference to its effect on microscopic preparations. A survey of representative fluorescent probes is provided, and their sensitivity, application, and limitations are described. The phototoxicity of fluorescent molecules is discussed using biomembranes and DNA as examples of targets of toxicity. Photodynamic therapy, a relatively new clinical application of phototoxicity, is described. After contusion-derived spinal cord injury there is localized tissue disruption and energy failure that results in early necrosis and delayed apoptosis, events that contribute to chronic central pain in a majority of patients. We assessed mechanisms of contusion-induced apoptosis of neurons and glia in a known central pain signalling pathway, the spinothalamic tract (STT), which may be a contributor to SCI-induced pain. Twenty-four hours after injury there was demonstrable apoptosis among neurons of the spinothalamic tract. Apoptosis in the injured spinal cord correlated well with prompt decreases in Bcl-xL and Bcl-xL/Bax protein ratios at the contusion site. There was definitive triggering of the inflammatory cytokine cascade with IL-1b being most robust and prompt in responding. Clearly, a better understanding of inflammatory processes, especially the role of cytokines after nerve injury, can lead to the development of new therapies that may prevent, and not just treat chronic central pain. Intervention in the inflammatory cascade had beneficial effects with confounds, which were mostly assessed by cDNA microarray analyses. We interpret these results as evidence that regulation of Bcl-xL and other genes that determine cell death outcomes may play a role in the inflammatory response to spinal injury and pain signalling function.

Studies on chromosomes and nuclei of very small bivalve larvae have been impeded by the veliger shell. It has been determined that the alcohokacetic acid fixative commonly used in cytogenetic techniques can be made to act as a decalcifying agent upon repeated heating. In addition, transfer of formalin fixed shelled specimens, routinely used as marine bioassay organisms, into ethyl alcohohacetic acid (3:1) fixative also yields clear cells for cytological examination of decalcified but otherwise intact oyster larvae and other zoo-plankton. Identification of cell type, such as germ-line primordia, in, for example, reproductive and ploidy level studies, and observations on the presence of bacteria can be made from the preparations. Material can be examined up to at least a year after preservation. The method is evaluated and its modifications are discussed. 相似文献
