Nef is an HIV-1 accessory protein that directly contributes to AIDS progression. Nef is myristoylated on the N-terminus, associates with membranes, and may undergo a transition from a solution conformation to a membrane-associated conformation. It has been hypothesized that conformational rearrangement enables membrane-associated Nef to interact with cellular proteins. Despite its medical relevance, to our knowledge there is no direct information about the conformation of membrane-bound Nef. In this work, we used neutron reflection to reveal what we believe are the first details of the conformation of membrane-bound Nef. The conformation of Nef was probed upon binding to Langmuir monolayers through the interaction of an N-terminal His tag with a synthetic metal-chelating lipid, which models one of the possible limiting cases for myr-Nef. The data indicate that residues are inserted into the lipid headgroups during interaction, and that the core domain lies directly against the lipid headgroups, with a thickness of ∼40 Å. Binding of Nef through the N-terminal His tag apparently facilitates insertion of residues, as no insertion occurred upon binding of Nef through weak electrostatic interactions in the absence of the specific interaction through the His tag. 相似文献

Summary The ultrastructure of the auricles, the ostia, and the ventricle of L. asellus and T. marmorea is described. The heart wall consists of an epicardium, a basement membrane, and an inner loose myocardium. The epicardial cells of the auricle are podocytes. The exposed cell body and the branched processes show pedicles. Ventricular epicardium is flat and simple. The slender, unbranched, mononucleated muscle fibres have a peripheral nucleus located midway along the fibre. Mitochondria are peripherally located, leaving the center to longitudinally running thick and thin myofilaments. Dense bodies and attachment plaques make up the Z-material. Sarcomeres and myofibrils are absent, as are transverse tubules and intercalated disks. The sarcoplasmic reticulum consists of peripheral tubules and subsarcolemmal cisternae, some of which radiate, branch, and run between myofilaments. Couplings are lacking. Ventricular fibres in T. marmorea show nexuses and desmosomes; in L. asellus only nexuses. The muscular ostia are tubular, and muscle fibres resemble those of the ventricle; nexuses are detected in T. marmorea and desmosomes in L. asellus. The c-abl proto-oncogene encodes a unique protein-tyrosine kinase (Abl) distinct from c-Src, c-Fes, and other cytoplasmic tyrosine kinases. In normal cells, Abl plays prominent roles in cellular responses to genotoxic stress as well as in the regulation of the actin cytoskeleton. Abl is also well known in the context of Bcr-Abl, the oncogenic fusion protein characteristic of chronic myelogenous leukemia. Breeding system and pollinator activity were studied in two Norwegian populations of Pulsatilla pratensis : a small outpost population, on Hovedøya in Oslo, and a large population located centrally within the main area of this species in Norway, on Sandøya in Tjøme, Vestfold. One objective was to assess, if possible, the chance of survival of the outpost population. Pulsatilla pratensis is not completely self incompatible, since artificial selfing produced a small amount of seed. However, spontaneous selfing does not occur. Pollination is accomplished by insects, almost exclusively bumble bees ( Bombus spp.). Nectar thievery resulted when the bumble bee inserted its proboscis from outside the flower in between the tepals (base working). A comparison between the two populations indicates that pollinator scarcity at the time of flowering possibly might constitute a limiting factor for recruitment in the outpost population. 相似文献
