A stochastic model is described that predicts the degree of singlet/singlet energy transfer in complexes formed between monovalent ligands and monovalent receptors. The modeling approach is intended to serve as an analytical tool for approximating the level of fluorescence quenching that can be expected to occur in fluorescently labeled monovalent ligands and receptors that are bound together in complexes. This approach has utility in areas such as modeling protein/protein interactions and designing fluorescence energy transfer assays.Using the crystallographic data for papain (monovalent ligand ) and concanavalin A (monovalent receptor ) along with a molecular graphics computational package the ligand and receptor were docked together to form a ligand/receptor complex. The intermolecular distances between the lysine resides of the ligand and receptor were then estimated, receptor complex was calculated assuming a value for the characteristic length R(0) of the donor/acceptor pair. Results from the stochastic model were used to calculate the level of fluorescence quenching one would expect for a resonance energy transfer competition assay based on the monovalent ligand/pair.Three key assumptions were made during the model development. First, all lysine resides for the ligand and receptor were equally reactive with the dye molecules so the stoichiometry of the donor and acceptor chromophores was governed by a binomial distribution. Second, the dye molecules were located at the alpha-carbon position for each reactive lysine residue. Finally, in the energy transfer competition assay, it was assumed that equilibrium existed between the ligand, receptor, and competing hapten at all times. Based on these assumptions, results are presented that indicate the maximum energy transfer for the monovalent papain/concanavalin. A complex is strongly dependent on the number of acceptor chromophores and on the value of R(0). Results are also presented on the approximate level of fluorescence quenching that may occur in a competition assay based on the papin/pConA complex. Lastly, a strategy is discussed for maximizing the dynamic range and linearity of energy transfer assays by optimizing several key design variables. 相似文献

We previously demonstrated the presence of GTP-binding proteins, G proteins, in the preimplantation mouse embryo (Jones and Schultz, 1990. Dev. Biol. 139, 250-262). These studies have been extended to the Day 6.5, 7.5, and 8.5 gestation embryo by employing PT-catalyzed ADP-ribosylation and immunoblotting techniques. We report here that the amount of embryonic alpha i increases from Day 6.5 to Day 7.5 of gestation, and remains at about the same level at Day 8.5. In contrast, the extent of PT-catalyzed ADP-ribosylation of Gi alpha protein(s) decreases between Days 6.5 and 7.5--this decrease is global and not restricted to a particular germ layer of the Day 7.5 embryo--and then dramatically increases by Day 8.5 of gestation. In the Day 8.5 gestation embryo, the extent of PT-catalyzed ADP-ribosylation of Gi alpha proteins increases along the anterior-posterior axis, whereas the amount of immunoreactive alpha i subunit decreases along this axis. By using a combination of PT-catalyzed ADP-ribosylation and immunoprecipitation with antisera specific for alpha i1, alpha i2, or alpha i3, we report that all three alpha i subtypes are present in the Day 8.5 gestation mouse embryo. Results of these experiments suggest that an activation of Gi proteins occurs between Days 6.5 and 7.5 of gestation in the postimplantation embryo, a time during which the embryo is gastrulating, and that a decreasing gradient of activation exists along the anterior to posterior axis in the Day 8.5 gestation embryo. Persistent neurogenesis in the central olfactory pathway characterizes many reptant decapods such as lobsters, crayfish and crabs. In these animals, the deutocerebral proliferative system generates new neurons which integrate into the neuronal network of the first order processing neuropil of the olfactory system, the deutocerebral chemosensory lobes (also called olfactory lobes). However, differences concerning the phenotype and the mechanisms that drive adult neurogenesis were reported in crayfish versus spiny lobsters. While numerous studies have focussed on these mechanisms and regulation of adult neurogenesis, investigations about the phylogenetic distribution are missing. To contribute an evolutionary perspective on adult neurogenesis in decapods, we investigated two representatives of basally diverging lineages, the dendrobranchiate Penaeus vannamei and the caridean Crangon crangon using the thymidine analogue Bromodeoxyuridine (BrdU) as marker for the S phase of cycling cells. Compared to reptant decapods, our results suggest a simpler mechanism of neurogenesis in the adult brain of dendrobranchiate and caridean shrimps. Observed differences in the rate of proliferation and spatial dimensions are suggested to correlate with the complexity of the olfactory system. We assume that a more complex and mitotically more active proliferative system in reptant decapods evolved with the emergence of another processing neuropil, the accessory lobes. © 2018 Wiley Periodicals, Inc. Anaerobic oxidation of methane (AOM) coupled to sulfate reduction is a microbially mediated unique natural phenomenon with an ecological relevance in the global carbon balance and potential application in biotechnology. This study aimed to enrich an AOM performing microbial community with the main focus on anaerobic methanotrophic archaea (ANME) present in sediments from the Ginsburg mud volcano (Gulf of Cadiz), a known site for AOM, in a membrane bioreactor (MBR) for 726 days at 22 (± 3)°C and at ambient pressure. The MBR was equipped with a cylindrical external ultrafiltration membrane, fed a defined medium containing artificial seawater and operated at a cross flow velocity of 0.02 m/min. Sulfide production with simultaneous sulfate reduction was in equimolar ratio between days 480 and 585 of MBR operation, whereas methane consumption was in oscillating trend. At the end of the MBR operation (day 726), the enriched biomass was incubated with 13C labeled methane, 13C labeled inorganic carbon was produced and the AOM rate based on 13C‐inorganic carbon was 1.2 μmol/(gdw d). Microbial analysis of the enriched biomass at 400 and 726 days of MBR operation showed that ANME‐2 and Desulfosarcina type sulfate reducing bacteria were enriched in the MBR, which formed closely associated aggregates. The major relevance of this study is the enrichment of an AOM consortium in a MBR system which can assist to explore the ecophysiology of ANME and provides an opportunity to explore the potential application of AOM. 相似文献

This article discusses how a small place – the polling booth – can be bounded as an ethnographic site with reference to the political and democratic event that it is supposed to facilitate. Concerns about the socio-material bounding of the booth form the main empirical case – a debate, which recently occurred in Denmark when the government proposed to digitalise voting. Digitalisation here became a controversy because of the potential illicit influences that computer experts argued would enter the polling booth and challenge the secrecy and the privacy of the vote, the transparency of the electoral process, and thus the electoral enactment of democracy itself. In this way the polling booth potentially works as an ethnographic entry point for following shifts in contemporary debates. 相似文献

Instream large wood (LW) constitutes an indispensable element of natural river ecosystems. It affects local hydraulics, morphology, nutrient budget, overall habitat complexity, and dynamics. Despite numerous studies about LW as a habitat for benthic communities, information on the varying importance along the longitudinal gradient of a river is lacking. The focus of this study is therefore to investigate general differences between lithal and xylal colonizers and to further investigate trends along the river course. We analyzed lithal and xylal communities at ten sites along the medium-sized Lafnitz River in Southeast Austria. Our results significantly show (1) a general differentiation between lithal and xylal communities, (2) an increasing distinction of the lithal and xylal fauna along the longitudinal gradient of the river, and (3) a distinct correlation between the distance from source and the number of exclusive xylal and nowadays predominantly rare taxa. The presence of LW is therefore directly linked to higher aquatic biodiversity compared to rocky substrates and presents a unique element for river restoration, especially in lower river sections. 相似文献
