Spores of Myxobolus pendula develop within the cores of complex cysts on the gill arch of creek chub Semotilus atromaculatus. Adjacent to, and surrounding, the spores are concentric layers of stratified interdigitating cells, whose nature was examined by transmission electron microscopy and by immunohistochemical and molecular biological techniques. In situ hybridization data using parasite-derived ribosomal DNA as a probe indicate that infection leads to the encapsulation of developing plasmodia by host immune cells that form an epithelioid granuloma. Epithelioid cell-cell adhesion is effected by desmosomes anchored intracellularly to cytokeratin intermediate filaments. High levels of alkaline phosphatase activity are associated with regions of cellular interdigitation. The granuloma may serve to limit the spread of the parasite to surrounding tissues but does not appear to inhibit diffusion of oxygen and nutrients to the developing spores.  相似文献

It has been postulated that a flow-dependent (and hence spatially varying) low density lipoprotein (LDL) concentration polarisation layer forms on the luminal surface of the vascular endothelium. Such a layer has the potential to cause heterogeneity in the distribution of atherosclerotic lesions by spatially modulating the rate of LDL transport into the arterial wall. Theoretical analysis suggests that a transmural water flux which is spatially heterogeneous at the cellular scale can act to enhance LDL concentration polarisation in a shear dependent fashion. However, such an effect is only observed if a relevant Peclet number (i.e. the ratio of LDL convection to LDL diffusion) is of order unity or greater. Based on the diffusivity of LDL in blood plasma, such a Peclet number is found to be far less than unity, implying that the aforementioned enhancement and shear dependence will not occur. However, this conclusion ignores the existence of the endothelial glycocalyx layer (EGL), which may inhibit the diffusion of LDL near the luminal surface of the endothelium, and hence raise any Peclet number associated with the transport of LDL. The present study numerically investigates the effect of the EGL, as well as a heterogeneous transmural water flux, on arterial LDL concentration polarisation. Particular attention is paid to measures of LDL concentration polarisation thought relevant to the rate of transendothelial LDL transport. It is demonstrated that an EGL is unlikely to cause any additional shear dependence of such measures directly, irrespective of whether or not LDL can penetrate into the EGL. However, it is found that such measures depend significantly on the nature of the interaction between LDL and the EGL (parameterised by the height of the EGL, the depth to which LDL penetrates into the EGL, and the diffusivity of LDL in the EGL). Various processes may regulate the interaction of LDL with the EGL, possibly in a flow dependent and hence spatially non-uniform fashion. It is concluded that any such processes may be as important as vascular scale flow features in terms of spatially modulating transendothelial LDL transport via an LDL concentration polarisation mechanism.  相似文献

Skeletal and smooth muscle tropomyosin (Tm) require acetylation of their N-termini to bind strongly to actin. Tm containing an N-terminal alanine-serine (AS) extension to mimic acetylation has been widely used to increase binding. The current study investigates the ability of an N-terminal AS extension to mimic native acetylation for both alpha alpha and beta beta smooth Tm homodimers. We show that (1) AS alpha-Tm binds actin 100-fold tighter than alpha-Tm and 2-fold tighter than native smooth alphabeta-Tm, (2) beta-Tm requires an AS extension to bind actin, and (3) AS beta-Tm binds actin 10-fold weaker than AS alpha-Tm. Tm is present in smooth muscle tissues as >95% heterodimer; therefore, we studied the binding of recombinant alphabeta heterodimers with different AS extensions. This study shows that recombinant Tm requires an AS extension on both alpha and beta chains to bind like native Tm and that the alpha chain contributes more to actin binding than the beta chain. Once assembled onto an actin filament, all smooth muscle Tm's regulate S1 binding to actin Tm in the same way, irrespective of the presence of an AS extension.  相似文献

Thyroid hormones (THs) play a critical role in differentiation, growth, and metabolism of animal and human organ systems, including the brain. Although associations between normal levels of THs and cognitive functions in healthy elderly individuals have been reported, the findings are inconsistent, possibly due to differences in study designs. Because thyroid disease occurs more frequently in women, the goal of the present study was to examine the relationship between levels of THs and performance on neuropsychological tests in 122 healthy, euthyroid women whose mean age was 51 years. Higher levels of free T3 were positively associated with longer completion times (slower performance) on Trail Making Test - Part A (p = 0.006) and Part B (p = 0.032) and on the Tower of London test (p = 0.002). Higher levels of thyroglobulin antibodies (TgAb) were positively correlated with more errors on the Trail Making Test Part B (p = 0.000), on the Word Fluency test (p = 0.023), and on the Design Fluency test (p = 0.045). No significant correlations between TH levels and scores on mood, verbal memory, or working memory measures were observed. The findings point to a possible link between THs and cognitive processes that are mediated primarily by frontal cortex, areas associated with executive function tasks, and suggest that elevations in levels of free T3 and TgAB within the normal range may negatively influence executive functions.  相似文献

Perforation plates and other vessel details as studied with scanning electron microscopy (SEM) have been reported for four species of Cornaceae (s.l.): similar features are shown by the four, suggesting that a more extensive sampling of the family might reveal similar phenomena. Perforation plates contain pit membrane remnants in the form of threads or, less commonly, laminar portions perforated by pores. When least well-represented, the pit membrane remnants are restricted to lateral ends of perforations and to the perforations transitional to lateral wall pitting. Perforations are all clearly bordered. Helical thickenings that do not form a continuous gyre are reported for the vessel walls ofAucuba. The presence of pit membrane remnants in vessel elements of Cornaceae correlates with the mesic habitats occupied by species in this family. The presence and type of pit membrane remnants reported by us in the three genera is very similar, although pit membrane remnants are doubtless a symplesiomorphy and thus not an indicator of relationships. The presence of pit membrane remnants in the three genera, however, does attest to the primitiveness of wood and other features of Cornaceae s.l.  相似文献

For a soldier, decisions to use force can happen rapidly and sometimes lead to undesired consequences. In many of these situations, there is a rapid assessment by the shooter that recognizes a threat and responds to it with return fire. But the neural processes underlying these rapid decisions are largely unknown, especially amongst those with extensive weapons experience and expertise. In this paper, we investigate differences in weapons experts and non-experts during an incoming gunfire detection task. Specifically, we analyzed the electroencephalography (EEG) of eleven expert marksmen/soldiers and eleven non-experts while they listened to an audio scene consisting of a sequence of incoming and non-incoming gunfire events. Subjects were tasked with identifying each event as quickly as possible and committing their choice via a motor response. Contrary to our hypothesis, experts did not have significantly better behavioral performance or faster response time than novices. Rather, novices indicated trends of better behavioral performance than experts. These group differences were more dramatic in the EEG correlates of incoming gunfire detection. Using machine learning, we found condition-discriminating EEG activity among novices showing greater magnitude and covering longer periods than those found in experts. We also compared group-level source reconstruction on the maximum discriminating neural correlates and found that each group uses different neural structures to perform the task. From condition-discriminating EEG and source localization, we found that experts perceive more categorical overlap between incoming and non-incoming gunfire. Consequently, the experts did not perform as well behaviorally as the novices. We explain these unexpected group differences as a consequence of experience with gunfire not being equivalent to expertise in recognizing incoming gunfire.  相似文献
