Recently, the fossil record of rodents from southwestern Brazilian Amazonia has been reviewed with regards to its diversity as well as its ecological relationships. In the reviews, the necessity to report new specimens collected with stratigraphic control was stated. Here, a new dinomyid specimen collected during a 2015 expedition to the Niterói locality, Acre River, is reported. The material is a fragment of skull with the right P4–M1 and the left P4–M2 preserved. The cheek teeth are protohypsodont, a characteristic employed to differentiate Potamarchinae dinomyids from the euhypsodont dinomyids Eumegamyinae and Tetrastylinae. The occlusal surface of the cheek teeth is composed of lophs with interruptions, showing little wear, which suggests that the specimen is not fully ontogenetically developed. The specimen has a unique combination of characters (protohypsodont and pentalophodont cheek teeth, with the leading edges of similar thickness to the trailing edges, and presence of a groove on the bottom of the infraorbital foramen) not present in other known dinomyids, which led us to erect a new taxon. The abundant and diverse fossil record of protohypsodont dinomyids suggests that an important radiative event may have occurred during the middle–late Miocene of northern South America.

Introduction

Species of the genera Psychotria and Palicourea are sources of indole alkaloids, however, the distribution of alkaloids within the plants is not known. Analysing the spatial distribution using desorption electrospray ionisation mass spectrometry imaging (DESI‐MSI) has become attractive due to its simplicity and high selectivity compared to traditional histochemical techniques.

Objectives

To apply DESI‐MSI to visualise the alkaloid distribution on the leaf surface of Psychotria prunifolia and Palicourea coriacea and to compare the distributions with HPLC–MS and histochemical analyses.

Methodology

Based upon previous structure elucidation studies, four alkaloids targeted in this study were identified using high resolution mass spectrometry by direct infusion of plant extracts, and their distributions were imaged by DESI‐MSI via tissue imprints on a porous Teflon surface. Relative quantitation of the four alkaloids was obtained by HPLC–MS/MS analysis performed using multiple‐reaction monitoring (MRM) mode on a triple quadrupole mass spectrometer.

Results

Alkaloids showed distinct distributions on the leaf surfaces. Prunifoleine was mainly present in the midrib, while 10‐hydroxyisodeppeaninol was concentrated close to the petiole; a uniform distribution of 10‐hydroxyantirhine was observed in the whole leaf of Psychotria prunifolia. The imprinted image from the Palicourea coriacea leaf also showed a homogeneous distribution of calycanthine throughout the leaf surface.

Conclusion

Different distributions were found for three alkaloids in Psychotria prunifolia, and the distributions found by MSI were in complete accordance with HPLC–MS analysis and histochemical results. The DESI‐MSI technique was therefore demonstrated to provide reliable information about the spatial distribution of metabolites in plants. Copyright © 2017 John Wiley & Sons, Ltd.  相似文献

Douglas-fir is a conifer species of major economic importance worldwide, including Western Europe and New Zealand. Herein we describe some characterization and significant refinement of somatic embryogenesis in Douglas-fir, with focus on maturation. The most typical structures observed in the embryonal masses were large polyembryogenic centres (up to 800–1500 µm) with a broad meristem, creating a compact cell “package” with suspensor cells. Singulated somatic embryos composed of both a embryonal head (300–400 µm) and long, tightly arranged suspensor were also frequent. Embryo development was enhanced following embryonal mass dispersion on filter paper discs at low density (50–100 mg fresh mass). Moreover, increasing gellan gum concentration in maturation medium (up to 10 g L?1) improved both the quantity and quality of cotyledonary somatic embryos (SEs), which were subsequently able to germinate and develop into plantlets at high frequency. Embryogenic yield was highly variable among the seven embryogenic lines tested (27–1544 SE g?1 fresh mass). Interestingly secondary somatic embryogenesis could be induced from cotyledonary SEs of both low- and highly-productive lines with some useful practical outcomes: secondary lines from low-performance lines (30–478 SE g?1 fresh mass) displayed significantly higher embryogenic yield (148–1343 SE g?1 fresh mass). In our best conditions, the total protein content in cotyledonary SEs increased significantly with maturation duration (up to 150 µg mg?1 fresh mass after 7 weeks) but remained below that of mature zygotic embryos (300 µg mg?1). The protein pattern was similar in both somatic and zygotic embryos, with major storage proteins identified as 7S-vicilin- and legumin-like proteins.  相似文献

Andisols are characterised by having abundant reactive Al in the form of short-range ordered (SRO) Al constituents and organo-Al complexes, which facilitates the accumulation of soil organic matter (OM). However, recent studies of New Zealand pastoral systems have reported loss of carbon (C) from Andisols when under intense management. This study compares the organic and inorganic chemistry of Andisols on two adjacent pasture sites under different pastoral management regimes (Paddock 2 being more intensively managed than Paddock 1), as well as under a nearby pine stand (Forest). Mean soil pH-H2O in Forest (5.3) was significantly lower (P?<?0.05) than that in Paddock 1 (5.7), which itself was significantly lower (P?<?0.05) than in Paddock 2 (6.1). Soil C concentrations were significantly higher (P?<?0.05) in the soils under pasture than under pine (63.8 g C/kg), and C in Paddock 1 (98.1 g C/kg) was significantly higher (P?<?0.05) than in Paddock 2 (84.1 g C/kg). The ratio of Al in organo-Al complexes (as estimated with sodium pyrophosphate) to the sum of Al in both SRO and organo-Al complexes (Alp/Alo) was significantly smaller (P?<?0.05) as the alkalinity of the soils increased (0.38, 0.23, 0.16 for Forest, Paddock 1 and Paddock 2, respectively). At the molecular level, soils under Forest had a larger relative contribution of degraded products of plant polysaccharides than those under pasture, while these had a larger contribution of fresh (e.g. cellulose and cutan/suberan aliphatic structures) and N-rich OM (e.g., microbial fingerprints, denoting a high microbial activity). Dissolved organic C content in the rhizosphere of pasture species was similar between paddocks, but Paddock 2 had a significantly (P?<?0.05) greater contribution of organic acids of MW?<?500 Da and higher pH (6.8 vs. 6.2). The results (1) confirm the common enrichment in organic C of New Zealand top soils under pasture compared to those under pine, and (2) reveal that the changes in the soil chemistry associated with pasture management may weaken the ability of these soils to preserve OM.  相似文献

This study provides the first environmental risk assessment (ERA) for zearalenone (ZEN), the only known mycoestrogen, based on a broad-scale investigation on its occurrence in rivers and creeks from Portugal. Water sample filtration and immunoaffinity columns (IAC) clean-up followed by liquid chromatograph with tandem mass spectrometry (LC/MS n ) provided an analytical method with good analytical performance. ZEN levels were determined for seven Portuguese rivers and one creek, during two different seasons, in a total of 38 samples collected upstream wastewater treatment plants (WWTPs). Overall, 23.7% were contaminated with ZEN at levels ranging between 5.6 and 82.6 ng/L. The highest concentration was observed during spring, although no statistically significant differences were observed between spring and autumn sampling campaigns. The potential ecotoxicological risk from ZEN to different trophic levels of aquatic organisms was evaluated by means of risk quotients (RQs) calculation. Although all the RQs obtained were lower than 1, our results confirm that ZEN is a relatively frequent contaminant in flowing waters in Portugal and might contribute to the overall estrogenic activity in the environment.  相似文献

Freshwater ecosystems are amongst the most threatened ecosystems on Earth. Currently, climate change is one of the most important drivers of freshwater transformation and its effects include changes in the composition, biodiversity and functioning of freshwater ecosystems. Understanding the capacity of freshwater species to tolerate the environmental fluctuations induced by climate change is critical to the development of effective conservation strategies. In the last few years, epigenetic mechanisms were increasingly put forward in this context because of their pivotal role in gene–environment interactions. In addition, the evolutionary role of epigenetically inherited phenotypes is a relatively recent but promising field. Here, we examine and synthesize the impacts of climate change on freshwater ecosystems, exploring the potential role of epigenetic mechanisms in both short‐ and long‐term adaptation of species. Following this wrapping‐up of current evidence, we particularly focused on bringing together the most promising future research avenues towards a better understanding of the effects of climate change on freshwater biodiversity, specifically highlighting potential molecular targets and the most suitable freshwater species for future epigenetic studies in this context.  相似文献

The role of trophic specialisation in taxonomic diversification remains unclear. Plant specialists diversify faster than omnivores and animalivores, but at shorter macroevolutionary scales this pattern sometimes reverses. Here, we estimate the effect of diet diversification on speciation rates in noctilionoid bats, controlling for tree shape, rate heterogeneity and macroevolutionary regimes. We hypothesise that niche subdivision among herbivores positively relates to speciation rates, differing between macroevolutionary regimes. We found the rate at which new herbivorous lineages originate decreases as rates of diet evolution increase. Herbivores experience higher speciation rates, but generalist herbivores and predominantly herbivorous omnivores speciate faster than specialised herbivores, omnivores and animalivores. Generalised herbivory is not a dead end. We show that analysing ecological traits and diversification requires accounting for macroevolutionary regimes and within‐ and between‐clade variation in evolutionary rates. Our approach overcomes the high false‐positive rates of other methods and illuminates the roles of herbivory and specialisation in speciation.  相似文献
