Trafficking protein particle complex 9 (TRAPPC9) is a major subunit of the TRAPPII complex. TRAPPC9 has been reported to bind nuclear factor κB kinase subunit β (IKKβ) and NF-kB-inducing kinase (NIK) where it plays a role in the canonical and noncanonical of nuclear factor-κB (NF-kB) signaling pathways, receptively. The role of TRAPPC9 in protein trafficking and cytoskeleton organization in osteoclast (OC) has not been studied yet. In this study, we examined the mRNA expression of TRAPPC9 during OC differentiation. Next, we examined the colocalization of TRAPPC9 with cathepsin-K, known to mediate OC resorption suggesting that TRAPPC9 mediates the trafficking pathway within OC. To identify TRAPPC9 protein partners important for OC-mediated cytoskeleton re-organization, we conducted immunoprecipitation of TRAPPC9 in mature OCs followed by mass spectrometry analysis. Our data showed that TRAPPC9 binds various protein partners. One protein with high recovery rate is L-plastin (LPL). LPL localizes at the podosomes and reported to play a crucial role in actin aggregation thereby actin ring formation and OC function. Although the role of LPL in OC-mediated bone resorption has not fully reported in detail. Here, first, we confirmed the binding of LPL to TRAPPC9 and, then, we investigated the potential regulatory role of TRAPPC9 in LPL-mediated OC cytoskeleton reorganization. We assessed the localization of TRAPPC9 and LPL in OC and found that TRAPPC9 is colocalized with LPL at the periphery of OC. Next, we determined the effect of TRAPPC9 overexpression on LPL recruitment to the actin ring using a viral system. Interestingly, our data showed that TRAPPC9 overexpression promotes the recruitment of LPL to the actin ring when compared with control cultures. In addition, we observed that TRAPPC9 overexpression reorganizes actin clusters/aggregates and regulates vinculin recruitment into the OC periphery to initiate podosome formation.  相似文献

Plant nutrition management is known as an efficient strategy to control environmental constraints. This experiment was conducted in a climate control greenhouse with a hydroponic system. The high temperature (36 °C?±?1) was imposed on the pots after fruit formation. The studied factors were silicon in 2 concentrations (0 and 4 parts per thousand (ppt)) and salicylic acid in 3 concentrations (0, 0.5, and 1 mM). They were sprayed on cucumber plants 3 times and under high-temperature conditions to evaluate if they can regulate and improve the yield and quality of cucumber fruit under high-temperature conditions or not. The results showed that all treatments significantly improved the nutritional status, total yield, and fruit quality (including marketable yield (i.e., fruits that can be sold due to their good shape) and nitrate content). Under high-temperature conditions, foliar application of silicon had the highest effect on the increase of total yield and marketable fruit yield (respectively, 36.14% and 40.29% increase compared to the control treatment). Micro-nutrients concentrations in the leaf were significantly increased by Si but a reverse status happened for salicylic acid. Under high temperatures, both treatments also significantly decreased the nitrate content of the fresh matter of fruit but silicon was the superior treatment. Silicon and salicylic acid, respectively, had positive effects on mitigation of adverse effects of high temperature on cucumber plants. These findings suggest the use of these treatments under high-temperature conditions in greenhouse cucumber production.

The production of self-pollinated plants could be important for improving medicinal plants secondary metabolites. In this study, 11 Thymus populations from eight species were evaluated to determine the effect of self and open pollination on agro-morphological characteristics, total phenolic content (TPC), essential oil (EO) content, and EO components. Inbreeding led to some positive effects of above mentioned traits in most of the studied populations. Total phenolic content ranged from 7.07 to 52.69 mg tannic acid equivalents (TAE) g−1 dry weight (DW) in open pollinated derived populations, while it varied from 1.2 to 55.03 mg TAE g−1 DW in self-pollinated ones. Under open and self-pollination condition, the highest EO content was obtained in T. trautvetteri (3.37 %) and T. pubescens (1.96 %), respectively. Gas chromatography-mass spectrometry (GC/MS) identified 42 compounds including thymol, carvacrol, linalool, p-cymene, γ-terpinene, terpinen-4-ol, and α-terpineol as the main compounds. In most cases, selfed plants compared to open pollinated ones, revealed higher thymol content. T. daenensis-1 showed a significant increase in thymol content (from 25.22 % to 74.3 %) due to self-pollination. Moreover, self-pollination led to emergence of some new compounds. Carvacrol methyl ether was the constituents of Thymus EO that are being reported in self-pollinated populations. Finally, inbreeding in Thymus might be suggested as a useful tool to increase genetic homogeneity for the selection of superior plants for improving secondary metabolite.  相似文献

Summary Lacertilian species display a remarkable diversity in the organization of the neural apparatus of their pineal organ (epiphysis cerebri). The occurrence of immunoreactive S-antigen and opsin was investigated in the retina and pineal organ of adult lizards, Uromastix hardwicki. In this species, numerous retinal photoreceptors displayed S-antigen-like immunoreactivity, whereas only very few pinealocytes were labeled. Immunoreactive opsin was found neither in retinal photoreceptors nor in pinealocytes. Electron microscopy showed that all pinealocytes of Uromastix hardwicki resemble modified pineal photoreceptors. A peculiar observation is the existence of a previously undescribed membrane system in the inner segments of these cells. It is evidently derived from the rough endoplasmic reticulum but consists of smooth membranes. The modified pineal photoreceptor cells of Uromastix hardwicki were never seen to establish synaptic contacts with somata or dendrites of intrapineal neurons, which are extremely rare. Vesiclecrowned ribbons are prominent in the basal processes of the receptor cells, facing the basal lamina or establishing receptor-receptor and receptor-interstitial type synaptoid contacts. Dense-core granules (60–250 nm in diameter) speak in favor of a secretory activity of the pinealocytes. Application of phosphorus at 40, 60, 80 and 100 kg P2O5 ha–1 in the presence of a uniform dressing of nitrogen (N) and potash (K2O) each applied at 20 and 24 kg ha–1 to chickpea (CM-88) grown in sandy loam soil in a replicated field experiment improved the nodulation response of the crop, increased its grain yield (ka ha–1) by 18, 59, 40 and 14 percent, biomass yield (ka ha–1) by 32, 32, 54 and 14 percent, biomass N (kg ha–1) by 31, 48, 49, 19 percent, and biomass P (kg ha–1) by 26, 40, 41 and 11 percent, respectively. The effect of phosphorus on the nitrogenase activity of the excised roots of chickpea was, however, inconsistent.  相似文献

Synopsis Bagrids in Bahr Shebeen Nilotic canal depend mainly on fish, insects and shrimp as well as fish embryos for food and their stomachs included runoff materials (e.g. plant foliage, glass, black crystals, coloured gravel). B. bayad maximised its efficiency of catching prey catfish by face to face attack to avoid damage by the prey's pectoral and dorsal spines. In the size classes of 10 to 30 cm standard length, B. bayad and B. docmac show diet overlap and interact with each other especially with respect to tilapias as prey. After this length, B. docmac, aided by its relatively larger mouth, shifted to larger size of tilapias to coexist with B. bayad.  相似文献
