Previous studies have demonstrated opposing roles for adenosine A1 and A2A receptors in the modulation of extracellular levels of glutamate and dopamine in the striatum. In the present study, acute systemic administration of motor-activating doses of the A2A receptor antagonist MSX-3 significantly decreased extracellular levels of dopamine and glutamate in the shell of the rat nucleus accumbens (NAc) and counteracted both dopamine and glutamate release induced by systemic administration of motor-activating doses of either the A1 receptor antagonist CPT or caffeine. Furthermore, exposure to caffeine in the drinking water (1 mg/mL, 14 days) resulted in tolerance to the effects of systemic injection of CPT or caffeine, but not MSX-3, on extracellular levels of dopamine and glutamate in the NAc shell. The present results show: first, the existence of opposite tonic effects of adenosine on extracellular levels of dopamine and glutamate in the shell of the NAc mediated by A1 and A2A receptors; second, that complete tolerance to caffeine's dopamine- and glutamate-releasing effects which develops after chronic caffeine exposure is attributable to an A1 receptor-mediated mechanism. Development of tolerance to the dopamine-releasing effects of caffeine in the shell of the NAc may explain its weak addictive properties and atypical psychostimulant profile.  相似文献

A study of larval Diptera (Chironomidae, Culicidae and Simuliidae) from Spain has been carried out to fulfill a catalogue of species of the genus Smittium (Harpellales: Legeriomycetaceae) present within these hosts. Among the reported taxa, eight are new species: Smittium brevisporum, S. bulbosporo-phorus, S. gracilis, S. hecatei, S. heterosporum, S. inex-pectans, S. prostratum and S. pseudodimorphum. We also report six previously described species, which are new for the Iberian Peninsula: S. alpinum, S. dipterorum, S. megazygosporum, S. pusillum, S. typhellum and S. fecundum. Three other species (S. simulii, S. culicis and S. culisetae) previously were reported from Spain. In two of them (S. fecundum and S. culicis), we describe for the first time the presence of zygospores. Some of the included species have been artificially cultured as well as ultrastructurally studied using scanning electron microscopy (SEM), with the purpose of observing the surface of both trichospores and the trichospore collar and the morphology of the trichospore appendage.  相似文献

Concentrations of D-glucose, D-fructose and D-sorbitol were quantified in porcine epididymal fluid by spectrofluorimetric assays and aldose reductase (AR) and sorbitol dehydrogenase (SDH) were located immunohistochemically in the epididymal epithelium. Glucose and fructose concentrations were low (<1 mM) and decreased in the cauda whereas sorbitol concentration (4-7 mM) was rather uniform along the duct. AR was luminally located on microvilli in the caput and corpus with less presence distally and was present in the lumen. SDH was present apically and basally in epithelial cells throughout the epididymis and in the lumen. The observations are consistent with diffusion of circulating glucose into the lumen, its conversion via AR to sorbitol which accumulates in the lumen and the action of SDH on sorbitol to produce fructose. Sperm metabolism of glucose and fructose may explain their lower concentrations in the cauda and sorbitol could be a metabolic substrate or osmolyte required for volume regulation.  相似文献

In recent years it has become increasingly clear that the metallothionein (MT) family of proteins is important in neurobiology. MT-I and MT-II are normally dramatically up-regulated by neuroinflammation. Results for MT-III are less clear. MTs could also be relevant in human neuropathology. In Alzheimer disease (AD), a major neurodegenerative disease, clear signs of inflammation and oxidative stress were detected associated with amyloid plaques. Furthermore, the number of cells expressing apoptotic markers was also significantly increased in these plaques. As expected, MT-I and MT-II immunostaining was dramatically increased in cells surrounding the plaques, consistent with astrocytosis and microgliosis, as well as the increased oxidative stress elicited by the amyloid deposits. MT-III, in contrast, remained essentially unaltered, which agrees with some but not all studies, of AD. In situ hybridization results in a transgenic mouse model of AD amyloid deposits, the Tg2576 mouse, which expresses human Abeta precursor protein harboring the Swedish K670N/M671L mutations, are in accordance with results in human brains. Overall, these and other studies strongly suggest specific roles for MT-I, MT-II, and MT-III in brain physiology.  相似文献

Abscisic acid (ABA) accumulation has been analyzed in irrigated and water-stressed wild-type and the vtc-1 mutant of Arabidopsis thaliana, which shows an ascorbate deficiency in leaves of approximately 60%. The amounts of ABA increased progressively up to 2.3-fold in water-stressed wild-type plants, whereas levels were kept at low levels in the irrigated plants. In contrast, initial increases followed by a sharp decrease of abscisic acid levels were observed in water-stressed vtc-1 mutants. Furthermore, the levels of this phytohormone increased up to fivefold in irrigated mutants. This differential accumulation of ABA in the mutant strongly correlated with the ascorbate redox state, but not with ascorbate levels. Changes in ABA levels in leaves paralleled those of chloroplasts. Immunolocalization studies showed a differential ABA accumulation in chloroplasts of vtc-1 mutants, which displayed the highest ABA labeling in irrigated plants. Our results indicate an altered pattern of ABA accumulation in the vtc-1 mutant compared to the wild type, under both irrigated conditions and water-stress conditions, which is strongly dependent on the ascorbate redox state.  相似文献

As part of an effort to develop nanoelectronic sensors for biological targets, we tested the potential to incorporate coiled coils as metallized, self-assembling, site-specific molecular linkers on carbon nanotubes (CNTs). Based on a previously conceived modular anchor-probe approach, a system was designed in which hydrophobic residues (valines and leucines) form the interface between the two helical peptide components. Charged residues (glutamates and arginines) on the borders of the hydrophobic interface increase peptide solubility, and provide stability and specificity for anchor-probe assembly. Two histidine residues oriented on the exposed hydrophilic exterior of each peptide were included as chelating sites for metal ions such as cobalt. Cysteines were incorporated at the peptide termini for oriented, thiol-mediated coupling to surface plasmon resonance (SPR) biosensor surfaces, gold nanoparticles or CNT substrates. The two peptides were produced by solid phase peptide synthesis using Fmoc chemistry: an acidic 42-residue peptide E42C, and its counterpart in the heterodimer, a basic 39-residue peptide R39C. The ability of E42C and R39C to bind cobalt was demonstrated by immobilized metal affinity chromatography and isothermal titration calorimetry. SPR biosensor kinetic analysis of dimer assembly revealed apparent sub-nanomolar affinities in buffers with and without 1 mM CoCl2 using two different reference surfaces. For device-oriented CNT immobilization, R39C was covalently anchored to CNT tips via a C-terminal cysteine residue. Scanning electron microscopy was used to visualize the assembly of probe peptide (E42C) N-terminally labeled with 15 nm gold nanoparticles, when added to the R39C-CNT surface. The results obtained open the way to develop CNT tip-directed recognition surfaces, using recombinant and chemically synthesized chimeras containing binding epitopes fused to the E42C sequence domain.  相似文献

Cellular networks display modular organization at different levels, from small sets of genes exchanging signals in morphogenesis to large groups of proteins involved in cell death. At the smallest scale, minute groups of interacting proteins or genes, so-called 'network motifs', have been suggested to be the functional building blocks of network biology. In this context, the relative abundance of a network motif would reflect its adaptive value. However, although the overabundance of motifs is non-random, recent studies by Mazurie et al. and by Kuo et al. show that motif abundance does not reflect their true adaptive value. Just as some architectural components emerge as a byproduct of a prior decision, common motifs might be a side effect of inevitable rules of genome growth and change.  相似文献

Muscle elasticity strongly relies on the mechanical anchoring of the giant protein titin to both the sarcomere M-band and the Z-disk. Such strong attachment ensures the reversible dynamics of the stretching-relaxing cycles determining the muscle passive elasticity. Similarly, the design of biomaterials with enhanced elastic function requires experimental strategies able to secure the constituent molecules to avoid mechanical failure. Here we show that an engineered titin-mimicking protein is able to spontaneously dimerize in solution. Our observations reveal that the titin Z1Z2 domains are key to induce dimerization over a long-range distance in proteins that would otherwise remain in their monomeric form. Using single molecule force spectroscopy, we measure the threshold force that triggers the noncovalent transition from protein dimer to monomer, occurring at ~700 piconewtons. Such extremely high mechanical stability is likely to be a natural protective mechanism that guarantees muscle integrity. We propose a simple molecular model to understand the force-induced dimer-to-monomer transition based on the geometric distribution of forces occurring within a dimeric protein under mechanical tension.  相似文献

The ectomycorrhizal fungus Tuber melanosporum fruits in association with Quercus in natural forests of Spain. Some of these stands are managed to keep an open canopy and meet the habitat requirements of the fungus. However, there are few quantitative studies analysing in these forests the relationship between soil environment and T. melanosporum. Eight forest stands which produce T. melanosporum have been monitored for 6 years in order to characterise the below-ground ectomycorrhizal community and to assess its temporal dynamics after experimental canopy opening. The br?lé, the ground where T. melanosporum fruits, shows a distinct ectomycorrhizal community, characterised by lower density of active ectomycorrhizal tips, lower morphotype richness per soil volume, higher abundance of T. melanosporum and lower abundance of Cenococcum geophilum than soil closest to the trunk of the host Quercus ilex. Opening the canopy has not stimulated an increase in T. melanosporum, suggesting that a shift in the soil environment alone will not trigger the formation of new truffières in the short term. The dry climate of these truffières may be a factor as T. melanosporum abundance appears to be sensitive to annual weather conditions.  相似文献
