Photosynthetic activities of cadmium-treated tomato plants   总被引:20,自引:0,他引:20  
Tomato plants (Lycopersicum esculentum Mill. cv. Moneymaker) grown on nutrient medium containing cadmium exhibit reduced net photosynthesis and reduced contents of chlorophyll and accessory pigments. In chloroplasts isolated from cadmiumtreated plants photosystem II activity, as measured by 2,6-dichlorophenolindophenol photoreduction, and photosystem II + I activity (H2O → methyl viologen) were both inhibited to about 60%. When 1,5-diphenylcarbazide was used as artificial electron donor, no significant cadmium effect was observed. Photosystem I activity was not affected by cadmium. The fine structure of chloroplasts in cadmium-treated plants was degenerated, similarly to senescence response. The principal symptom of cadmium action was the occurrence of large plastoglobules and a disorganization of the lamellar structure, mainly grana stacks. Transfer of cadmium-treated plants into a medium with increased manganese level caused grana stacking and restoration of photosystem II activity.  相似文献

Mammalian mitochondrial chaperonin 60 functions as a single toroidal ring.   总被引:20,自引:0,他引:20  
Chaperonins are thought to participate in the process of protein folding in bacteria and in eukaryotic mitochondria and chloroplasts. While some chaperonins are relatively well characterized, the structures of the mammalian chaperonins are unknown. We have expressed a mammalian mitochondrial chaperonin 60 in Escherichia coli and purified the recombinant protein to homogeneity. Structural and biochemical analyses of this protein establish a single toroidal structure of seven subunits, in contrast to the homologous bacterial, fungal, and plant chaperonin 60s, which have double toroidal structures comprising two layers of seven identical subjects each. The recombinant mammalian chaperonin 60, together with the mammalian chaperonin 10 (but not with bacterial chaperonin 10), facilitates the formation of catalytically active ribulose-bisphosphate carboxylase from an unfolded state in the presence of K+ and MgATP. Analysis of the partial reactions involved in this in vitro reconstitution reveals that the single toroid of chaperonin 60 can form stable complexes with both unfolded or partially folded [35S]ribulose-bisphosphate carboxylase and mitochondrial (but not bacterial) chaperonin 10 in the presence of MgATP. We conclude that the minimal functional unit of chaperonin 60 is a single hepatmeric toroid.  相似文献

Val-6() of hemoglobin S forms the primary site of intertetrameric interaction in the polymerization of deoxy hemoglobin S. However, a number of other intermolecular interactions contribute significantly to the polymerization process as well as to the stability of the polymerized gel. The strong stabilizing influence of Val-6() in the polymerization process is reflected in the fact that although a number of mutations at any one of the intermolecular contact regions (or perturbation of these contact regions by chemical modification) result in some increase in the solubility of deoxy hemoglobin S, none of these mutations and/or chemical modifications completely neutralize the polymerizing influence of Val-6(), i.e., restores the solubility to that of hemoglobin A. Additivity and/or synergy of the solubilizing influence of two or more chemical modification reactions each of which independently increases the solubility may be considered as a possible strategy to restore the solubility of deoxy hemoglobin S to that of hemoglobin A. In the present study, the cumulative solubilizing influence of amidation of Glu-43() and hydroxyethylation of -amino groups of hemoglobin S has been investigated by preparing hemoglobin S with double modification. Modification of Glu-43() by amidation with glycine ethyl ester did not influence the reactivity of the -amino groups of hemoglobin S toward reductive hydroxyethylation, thus permitting the preparation of hemoglobin S with the two modifications. The reductive hydroxyethylation increased the oxygen affinity of amidated hemoglobin S to nearly the same degree as it does on modification of unmodified hemoglobin. In addition, hemoglobin S with double modification has a Hill coefficient that is the same as that of unmodified hemoglobin S, suggesting that the overall quaternary interaction of hemoglobin S with a double modification is nearly the same as the unmodified protein. However, the reductive hydroxyethylation of the amidated hemoglobin S increased the solubility of the protein further. The solubility of hemoglobin S with a double modification is nearly twice that of the unmodified protein and is close to that of 1:1 mixture of hemoglobin S and hemoglobin F. The results demonstrate the additivity of the solubilizing influence of perturbing the quinary interactions at the intermolecular contact regions of deoxy hemoglobin S.  相似文献

We have studied the effect of choline on the activity and temperature dependency of the brush-border alkaline phosphatase isoenzymes from rat intestine (tissue-specific type), and from kidney and placenta (tissue-nonspecific type). The removal of choline with phospholipase D resulted in the loss of enzyme activity in all the membranes, whereas in situ loss in the discontinuity of Arrhenius plots occurred in the kidney and the placental membranes, but not in the intestinal membranes. The lost activity was restored either by addition of free choline or phosphatidylcholine or by the removal of the enzyme from the membrane surface. Intestinal enzyme was removed by papain, while the tissue-nonspecific enzyme was released by subtilisin and by phosphatidylinositol-specific phospholipase C. The enzyme from kidney and placental membranes aggregated (rho = 1.13) upon removal of choline, and addition of choline resulted in disaggregation (rho = 1.03). Conversion of discontinuous to continuous linear plots of alkaline phosphatase in the kidney and placental membranes paralleled the increase in membrane phosphatidic acid content, and the decrease in total phosphatidylcholines. The intestinal enzyme produced plots with break points at all phosphatidic acid/phosphatidylcholine ratios. The change brought about by treatment with phospholipidase D was not due to changes in the half-saturation kinetics (Km) for the substrate. Based on these studies we conclude that the active site of the tissue-nonspecific phosphatase is approximated to exterior membrane cholines, as in the case of the intestinal isoenzyme; that despite similar effects on the membrane content of phospholipids, phospholipase D treatment caused much greater effects on the tissue-nonspecific enzyme, as assessed by Arrhenius plots and density centrifugation; that these effects are due to different protein structures rather than to a lipid milieu unique to each brush-border membrane.  相似文献

Brachiaria ramosa is cultivated in pure stands. Its grains are used in preparing at least nine traditional foods in South India. Setaria glauca is cultivated in mixed stands along with little millet (Panicum sumatrense). In Orissa state and in Southern India the grains are used to make at least six traditional foods as a supplementary ingredient. The weedy forms of these were found growing with upland rice and some millets in diverse agro-ecological niches, and are identified by various vernacular names by local farmers. Explorations were conducted to gather information on the evolutionary trends, domestication and ethnobotany of these minor food crops. These findings substantiate the belief that domestication process may have passed through three distinct phases: (i) an associated mimic weed with upland rice and some millets; (ii) a secondary crop mixed with little millet or kodo millet; and (iii) as an independent crop. Also, this process has been promoted as a means of imparting adaptation to an arid climate and in turn related to the diversification of the agriculture basic complex.  相似文献

1. Although boreal lakes are ice‐covered for several months annually, little is known about the behaviour of fish under ice. To consider the reasons for diel vertical migrations (DVM) it is important to compare periods under ice as opposed to under open water. Echosounding provides a tool for non‐intrusive continuous monitoring of fish, even in winter. 2. Changes in the vertical distribution of fish through six 48‐h periods were monitored using a stationary, mounted echosounder that beamed vertically either from the bottom up or from the surface down from February to April, 2003. The up‐beaming and down‐beaming transducers were run alternately for 24 h each over the 48‐h period. Standard echo analysis software was used to detect fish traces and estimate the vertical and temporal distribution of fish abundance. Fish were sampled with a winter seine. 3. Prominent diel vertical migration in response to changing light level was detected throughout the study period (late winter to spring). Fish were highest in the water column at sunset and sunrise. In daylight, most detected fish were well below 10‐m depth. The number of fish detected was greatest during the night when they occurred throughout almost the whole water column, sometimes with a considerable number very close to the ice. Fish were mostly vendace and whitefish. 4. It became evident from data from the up‐beaming transducer that at night fish may occupy the layer closest to the surface. These fish would not have been detected if we had only used the down‐beaming transducer. The overall pattern of DVM in winter was very similar to that in summer. The results support the suggestion that DVM is a genetically fixed behavioural trait responding to the contemporary level of illumination.  相似文献

The plasma membrane antigens of an undifferentiated small cell (oat cell) carcinoma of the lung were studied by the indirect immunofluorescence method on frozen section substrates with a rabbit antiserum prepared to the tumor plasma membrane fraction. After appropriate absorption of the antiserum, at least two differentiation antigens present on the tumor cells but undetectable on normal lung surface or glandular epithelial cells were identified. One antigen(s) was characteristic of certain normal, endodermally-derived epithelial cells of the digestive system, including those of colonic mucosa, hepatic ducts, pancreatic ducts and acini, and islets of Langerhans. The other antigen(s) was characteristic of certain normal, neural crest-derived cells in the peripheral nervous system, including cells in peripheral nerve, dorsal root ganglion, and anterior roots of the spinal cord; parasympathetic ganglion cells in the colon; and small nerves and nerve processes in the lung, colon, and skin. It was concluded that the presence of these differentiation antigens on the tumor cells resulted from the expression of gene products repressed in the normal cell of origin of the tumor.  相似文献

Marginal organic soils, abundant in the boreal region, are being increasingly used for bioenergy crop cultivation. Using long‐term field experimental data on greenhouse gas (GHG) balance from a perennial bioenergy crop [reed canary grass (RCG), Phalaris arundinaceae L.] cultivated on a drained organic soil as an example, we show here for the first time that, with a proper cultivation and land‐use practice, environmentally sound bioenergy production is possible on these problematic soil types. We performed a life cycle assessment (LCA) for RCG on this organic soil. We found that, on an average, this system produces 40% less CO2‐equivalents per MWh of energy in comparison with a conventional energy source such as coal. Climatic conditions regulating the RCG carbon exchange processes have a high impact on the benefits from this bioenergy production system. Under appropriate hydrological conditions, this system can even be carbon‐negative. An LCA sensitivity analysis revealed that net ecosystem CO2 exchange and crop yield are the major LCA components, while non‐CO2 GHG emissions and costs associated with crop production are the minor ones. Net bioenergy GHG emissions resulting from restricted net CO2 uptake and low crop yields, due to climatic and moisture stress during dry years, were comparable with coal emissions. However, net bioenergy emissions during wet years with high net uptake and crop yield were only a third of the coal emissions. As long‐term experimental data on GHG balance of bioenergy production are scarce, scientific data stemming from field experiments are needed in shaping renewable energy source policies.  相似文献
