We provide guidance for monitoring whether human activities affect the physiology or behavior of marine mammals and, if so, whether those effects may lead to changes in survival and reproduction at the population level. We suggest that four elements be included in designing and implementing such a monitoring program. The first is development of a theory of change: a set of mechanistic hypotheses that outline why a given activity might be expected to have one or more measurable effects on individuals and populations, and ideally the magnitude, timing, and duration of the effects. The second element, definition of biologically meaningful effect sizes, ultimately facilitates the development of a monitoring program that can detect those magnitudes of effect with the desired levels of precision. The third element, selection of response variables for monitoring, allows inference to whether observed changes in the status of individuals or populations are attributable to a given activity. Visual observations, passive acoustic and tagging instruments, and direct physical measurements all can provide data that facilitate quantitative hypothesis testing. The fourth element is specification of the temporal sequence of monitoring. These elements also can be used to inform monitoring of the responses of other taxonomic groups to human activities.  相似文献

The human squamous cell carcinoma antigens (SCCA) 1 and 2 are members of the serpin family that are 92% identical in their amino acid sequence. Despite this similarity, they inhibit distinct classes of proteinases. SCCA1 neutralizes the papain-like cysteine proteinases, cathepsins (cat) S, L, and K; and SCCA2 inhibits the chymotrypsin-like serine proteinases, catG and human mast cell chymase. SCCA2 also can inhibit catS, as well as other papain-like cysteine proteinases, albeit at a rate 50-fold less than that of SCCA1. Analysis of the mechanism of inhibition by SCCA1 revealed that the reactive site loop (RSL) is important for cysteine proteinase inhibition. The inhibition of catS by a mutant SCCA2 containing the RSL of SCCA1 is comparable to that of wild-type SCCA1. This finding suggested that there were no motifs outside and only eight residues within the RSL that were directing catS-specific inhibition. The purpose of this study was to determine which of these residues might account for the marked difference in the ability of SCCA1 and SCCA2 to inhibit papain-like cysteine proteinases. SCCA2 molecules containing different RSL mutations showed that no single amino acid substitution could convert SCCA2 into a more potent cysteine proteinase inhibitor. Rather, different combinations of mutations led to incremental increases in catS inhibitory activity with residues in four positions (P1, P3', P4', and P11') accounting for 80% of the difference in activity between SCCA1 and SCCA2. Interestingly, the RSL cleavage site differed between wild-type SCCA2 and this mutant. Moreover, these data established the importance of a Pro residue in the P3' position for efficient inhibition of catS by both wild-type SCCA1 and mutated SCCA2. Molecular modeling studies suggested that this residue might facilitate positioning of the RSL within the active site of the cysteine proteinase.  相似文献

J Strength Cond Res 25(12): 3472-3478, 2011-Currently, little research has been conducted using body weight reduction (BWR) as a means to enhance vertical jump. The purpose of this study was to determine the effects of different elastic cord assistance levels on vertical jump height (JH), takeoff velocity (TOV), relative ground reaction force (rGRF), relative impact force (RIF), and descent velocity (DV). Thirty recreationally trained college men and women (M = 15, W = 15) completed 3 testing sessions consisting of 5 conditions: 0, 10, 20, 30, and 40% BWR. In all BWR conditions, the subjects wore a full body harness while being attached to 2 elastic cords suspended from the ceiling and a linear velocity transducer. They then performed 3 maximal countermovement jumps with arm swing on a force plate. The results indicated no interaction of condition by sex for any variable; however, there was a significant (p < 0.05) main effect for condition for each variable. The JH significantly increased across all conditions (0%: 43.73 ± 1.62 cm, 40%: 64.77 ± 2.36 cm). The TOV at 30% (2.73 ± 0.34 m·s) was significantly greater than that at 0% (2.59 ± 0.39 m·s) and 10% (2.63 ± 0.34 m·s), whereas that at 40% (2.79 ± 0.43 m·s) was significantly greater than that at >0, 10, and 20%. The rGRF at 30% (18.62 ± 4.35 N·kg) was significantly greater than that at >0, 10, and 20%, whereas that at 40% (21.38 ± 5.21 N·kg) was significantly greater than in all conditions. The RIF at 20, 30, and 40% (40%: 61.60 ± 18.53 N·kg) was significantly greater than that at 0% (44.46 ± 9.12 N·kg). The DV at 20% (2.61 ± 0.31 m·s) was significantly greater than at 10%, whereas those at 30 and 40% (2.8 ± 0.41 m·s) were significantly greater than at 0, 10, and 20%. These results demonstrate that using different elastic cord levels to reduce body weight appears effective for increasing ascent and descent force and velocity variables. Future research should investigate greater BWR% and chronic training.  相似文献

Measles is a potential lethal disease, justifying large immunization campaigns. Attenuated strains are used in immunization with very good safety records. Interestingly, following clinical observations of tumor regressions after measles infection, preclinical and clinical studies have highlighted the therapeutic potential of attenuated strains of measles. The aim of this review is to explain how these viruses can selectively infect and kill cancer cells, and how this selectivity can be improved. We will detail the therapeutic strategies under development, in particular the combination of viruses with chemotherapy and radiation therapy. Furthermore, the engineering of measles viruses encoding the sodium/iodide symporter could enable virus-directed radio-isotope therapy. Antiviral immunity could be a limit of measles therapy. We will highlight the promising combinations with immunosuppressive drugs and innovative strategies using infected cell carriers, aiming at circumventing the immune response and paving the way to future clinical trials.  相似文献

Given the proposed importance of membrane tension in regulating cellular functions, we explore the effects of a finite surface tension on phase equilibrium using a molecular theory that captures the quantitative structure of the phase diagram of the tensionless DPPC/DOPC/Cholesterol lipid bilayer. We find that an increase in the surface tension decreases the temperature of the transition from liquid to gel in a pure DPPC system by ~1.0 K/(mN/m), and decreases the liquid-disordered to liquid-ordered transition at constant chemical potentials by approximately the same amount. Our results quantitatively isolate the role of tension in comparison to other thermodynamic factors, such as pressure, in determining the phase behavior of lipid bilayers.  相似文献

Membranes containing a wide variety of ternary mixtures of high chain-melting temperature lipids, low chain-melting temperature lipids, and cholesterol undergo lateral phase separation into coexisting liquid phases at a miscibility transition. When membranes are prepared from a ternary lipid mixture at a critical composition, they pass through a miscibility critical point at the transition temperature. Since the critical temperature is typically on the order of room temperature, membranes provide an unusual opportunity in which to perform a quantitative study of biophysical systems that exhibit critical phenomena in the two-dimensional Ising universality class. As a critical point is approached from either high or low temperature, the scale of fluctuations in lipid composition, set by the correlation length, diverges. In addition, as a critical point is approached from low temperature, the line tension between coexisting phases decreases to zero. Here we quantitatively evaluate the temperature dependence of line tension between liquid domains and of fluctuation correlation lengths in lipid membranes to extract a critical exponent, ν. We obtain ν = 1.2 ± 0.2, consistent with the Ising model prediction ν = 1. We also evaluate the probability distributions of pixel intensities in fluorescence images of membranes. From the temperature dependence of these distributions above the critical temperature, we extract an independent critical exponent of β = 0.124 ± 0.03, which is consistent with the Ising prediction of β = 1/8.  相似文献

Zusammenfassung Die vorliegende Arbeit berichtet über Versuche mit 4 verschiedenen Einsporangienlinien vonPhytophthora infestans.Die Kultur- und Infektionsmethoden werden beschrieben. Es werden für diese Versuche Kulturen auf Malzextraktagar, Kartoffelknollen und Kartoffellaub neben- und nacheinander benutzt, um einerseits die Vermischungsgefahr weitgehend zu vermeiden und andererseits die Infektionstüchtigkeit der Sporen auf dem Optimum zu halten.Die Ergebnisse von Infektionsversuchen mit den 4 Linien des Pilzes auf 246 Kartoffelklonen werden besprochen. Es handelt sich umF 1,F 2,F2,F3,F4, Klone der KreuzungSolanum demissum x Solanum tuberosum. Nach ihrem Verhalten gegenüber den 4 Phytophthoralinien zerfallen diese 246 Klone in 5 Klongruppen: A, W, K, M, Z. Das Verhalten der 4 Linien auf diesen Gruppen wird dargestellt.Die Bedeutung der verschiedenen Linien des Pilzes für die züchterische Arbeit wird diskutiert. Nicht die geographische Verbreitung einer Pilzrasse, sondern ihre Aggressivität gegenüber dem Zuchtmaterial des Züchters bestimmt ihre Bedeutung für unsere züchterischen Arbeiten.Ein Testsortiment zur Charakterisierung der 4 Linien wird angegeben.Die Möglichkeiten der Entstehung und der Auffindung weiterer Pilzrassen werden besprochen.Mit Unterstützung der Deutschen Forschungsgemeinschaft.  相似文献
