In single-particle tracking experiments, the diffusion coefficient D may be measured from the trajectory of an individual particle in the cell membrane. The statistical distribution of single-trajectory diffusion coefficients is examined by Monte Carlo calculations. The width of this distribution may be useful as a measure of the heterogeneity of the membrane and as a test of models of hindered diffusion in the membrane. For some models, the distribution of the short-range diffusion coefficient is much narrower than the observed distribution for proteins diffusing in cell membranes. To aid in the analysis of single-particle tracking measurements, the distribution of D is examined for various definitions of D and for various trajectory lengths.  相似文献

Aggregation in a lipid bilayer is modeled as cluster-cluster aggregation on a square lattice. In the model, clusters carry out a random walk on the lattice, with a diffusion coefficient inversely proportional to mass. On contact, they adhere with a prescribed probability, rigidly and irreversibly. Monte Carlo calculations show that, as expected, rotational diffusion of the aggregating species is highly sensitive to the initial stages of aggregation. Lateral diffusion of an inert tracer obstructed by the aggregate is a sensitive probe of the later stages of aggregation. Cluster-cluster aggregates are much more effective barriers to lateral diffusion of an inert tracer than the same area fraction of random point obstacles is, but random point obstacles are more effective barriers than the same area fraction of compact obstacles. The effectiveness of aggregates as obstacles is discussed in terms of particle-particle correlation functions and fractal dimensions. Results are applicable to aggregation of membrane proteins, and at least qualitatively to aggregation of gel-phase lipid during lateral phase separation.  相似文献

Two new monoclonal antibodies, one a mouse IgM and the other a human IgM that reacted with guanosine, were compared to human serum antibodies from patients with systemic lupus erythematosus (SLE). The human monoclonal antibody was polyspecific in its binding to the nucleoside bases, whereas the mouse monoclonal antibody was relatively specific for guanosine when compared by using an enzyme-linked immunosorbent assay (ELISA). Neither antibody bound polyguanylic acid or denatured single-stranded (ss) DNA, however. Serum IgG antibodies from seven patients with SLE cross-reacted with the mouse monoclonal antibody and showed considerable specificity for guanosine. In contrast, the human serum IgG antiguanosine antibodies also bound ssDNA but not dsDNA or polyguanylic acid. Serum IgG antibodies to guanosine measured by ELISA from the seven SLE patients had a decreased response when compared to the total serum IgG response to ssDNA, and most of the antibodies that bound guanosine also bound ssDNA. These studies provide new evidence that there are specific IgG antibodies to guanosine in SLE sera that are a small fraction of the antibodies to ssDNA. Further efforts to define the role of these guanosine antibodies in SLE may provide a better understanding of the basic mechanisms responsible for the development of SLE in man.  相似文献

A recent hypothesis considers brown adipose tissue (BAT) to be an important source of diet-induced thermogenesis (DIT). In turn, DIT and thermogenesis in general are believed to be key factors in the control of obesity of laboratory rodents. This hypothesis was developed from the study of single gene mutant obese rodents. The present research tested this hypothesis in mice with polygenic control of growth and obesity, which is more characteristic of the type of genetic variation expected in human and other mammalian populations. Control and high fat diets were used to test responses of five genetically selected lines of mice showing different patterns of growth and obesity. All lines deposited more fat on the high fat diet, but the most obese line showed the largest increase in BAT and the lipid-free dry (LFD) component of BAT. Use of LFD per unit body weight gave results which supported the hypothesis being tested, but it was argued that this measure is misleading. When brown and white adipose tissue growth relative to body weight were examined, 2 of the 10 line-diet groups showed alterations in BAT growth patterns. However, it was concluded that BAT, if involved at all, was not a major factor in growth and obesity differences.  相似文献

Lateral diffusion of mobile proteins and lipids (tracers) in a membrane is hindered by the presence of proteins (obstacles) in the membrane. If the obstacles are immobile, their effect may be described by percolation theory, which states that the long-range diffusion constant of the tracers goes to zero when the area fraction of obstacles is greater than the percolation threshold. If the obstacles are themselves mobile, the diffusion constant of the tracers depends on the area fraction of obstacles and the relative jump rate of tracers and obstacles. This paper presents Monte Carlo calculations of diffusion constants on square and triangular lattices as a function of the concentration of obstacles and the relative jump rate. The diffusion constant for particles of various sizes is also obtained. Calculated values of the concentration-dependent diffusion constant are compared with observed values for gramicidin and bacteriorhodopsin. The effect of the proteins as inert obstacles is significant, but other factors, such as protein-protein interactions and perturbation of lipid viscosity by proteins, are of comparable importance. Potential applications include the diffusion of proteins at high concentrations (such as rhodopsin in rod outer segments), the modulation of diffusion by release of membrane proteins from cytoskeletal attachment, and the diffusion of mobile redox carriers in mitochondria, chloroplasts, and endoplasmic reticulum.  相似文献

Attractiveness judgments are thought to underpin adaptive mate choice decisions. We investigated how these judgments change during adolescence when mate choice is becoming relevant. Adolescents aged 11–15 evaluated faces and voices manipulated along dimensions that affect adults' judgments of attractiveness and that are thought to cue mate value. Facial stimuli consisted of pairs of faces that were more or less average, more or less feminine, or more or less symmetric. The adolescents selected the more average, symmetric, and feminine faces as more attractive more often than chance, but judgments of some facial traits differed significantly with rater age and sex, indicating a role of development in judgments of facial cues. Vocal stimuli consisted of pairs of voices manipulated to raise or lower perceived pitch. The older but not younger girls selected the lower-pitched male voices as more attractive at rates above chance, while the younger but not older boys selected the higher-pitched female voices as more attractive. Controlling for rater age, increased pubertal development was associated with increased selection of lower-pitched boys' voices by girls and decreased selection of feminized male faces by boys. Our results are the first to demonstrate that adolescents show somewhat similar attractiveness judgments to adults in age-matched stimuli and that age, sex, and pubertal development have measurable effects on adolescents' attractiveness judgments. They suggest that attractiveness judgments in humans, at least for some traits, are facultatively calibrated to the individual's life stage, only reaching adult values upon sexual maturity when mate choice decisions become relevant.  相似文献
